        떡쑥 추출물의 항염증 및 항알러지 효과

        떡쑥(Gnaphalium affine D. Don, GA)은 동아시아 지역에서 식용으로 사용되고 있으며, 예로부터 전통적인 민간요법 약재로 사용되어 왔다. 현재 떡쑥 추출물(GA extract, GAE)의 항산화 활성과 항보체 활성 등은 알려져 있으나, 항염과 항알러지 효능 및 그 작용 기작은 자세히 알려져 있지 않다. 본 연구에서는 염증 매개인자인 산화질소, 프로스타글란딘 E<sub>2</sub>, Toll-유사수용체 4, 에오탁신-1, 히스타민의 활성화에 대한 GAE의 저해효과를 평가하였다. 본 연구를 통해, GAE는 유도성 산화질소 합성효소와 COX-2의 발현을 저해함을 확인하였으며, 이를 통해 산화질소와 프로스타글란딘 E<sub>2</sub>의 생성을 저해함을 확인하였다. GAE는 LPS로부터 유도된 Toll-유사수용체 4의 발현에도 영향을 미치는 것을 확인하였으며, A23187로부터 유도되는 비만세포의 히스타민 방출의 억제에도 효과적으로 작용하는 것을 확인하였다. 또한 IL-4로부터 유도된 에오탁신-1의 생성도 효과적으로 억제하는 결과를 확인하였다. 이상의 결과로부터 GAE는 항염증과 항알러지 효능을 가진다고 사료되며, 향후 항염증 및 항알러지 화장품 원료로서의 이용가능성을 보였다. Gnaphalium affine D. DON (GA) has been used as a vegetable as well as a folk medicine in East Asia. The antioxidant and anti-complementary activity of GA extract (GAE) has also been reported. However, little is known about its anti-inflammatory and anti-allergic effect and mechanism of action. In this study, we evaluated the inhibitory effects of GAE on the production of inflammatory mediators such as NO, PGE<sub>2</sub>, TLR4, eotaxin-1 and histamine. Our results suggest that GAE inhibits the production of NO and PGE<sub>2</sub> by inhibiting transcriptional activation via the involvement of iNOS and COX-2. The LPS-induced expression of Toll-like receptor 4 (TLR4) was also attenuated. In addition, GAE inhibited A23187-induced histamine release from MC/9 mast cells. It also inhibited the production of eotaxin-1 induced by IL-4. Collectively, these results suggest that GAE may have considerable potential as a cosmetic ingredient with anti-inflammatory and anti-allergic properties.

        천연복합 제제 BSASM의 항염효과 비교평가

        In the treatment of chronic inflammatory skin diseases, topical steroids are known to show strong efficacy in relieving inflammation. Nevertheless, because its use in treatment is limited due to the side effects of long-term use, development of a therapeutic agent using natural products is necessary. Studies on various natural products with antiinflammatory effects are currently being conducted, however few studies verifying the anti-inflammatory effect of natural products through direct comparison with steroids have been reported. In this study, the anti-inflammatory effect of BSASM, a complex of seven natural products, and hydrocortisone, a topical steroid, were directly compared, and the effectiveness of BSASM as an anti-inflammatory agent was evaluated. Compared with hydrocortisone (topical steroids), BSASM had a similar inhibitory effect on the production of nitric oxide and prostaglandin E2, and showed a better inhibitory effect than hydrocortisone in inhibiting the production of pro-inflammatory cytokines, TNF-α and IL-6. In addition, mRNA expression of collagen type I alpha 1 was decreased by hydrocortisone in human dermal fibroblasts, which is associated with skin atrophy, however expression of COL1A1 was not affected by BSASM. Based on these results, it is expected that BSASM can be applied as a natural alternative to hydrocortisone in treatment of chronic inflammatory skin diseases.

        고삼 추출물의 피부장벽 강화와 염증완화 효과

        아토피성 피부염은 피부장벽 기능장애, 염증 및 만성 소양증을 특징으로 하는 다인성의 염증성 피부질환이다. 아토피성 피부염은 유전적, 면역학적, 환경적 요인 등의 복합적인 요인으로 피부장벽 기능과 면역기능의 장애를 유발한다고 알려져 있다. 고삼 추출물은 중국전통의학에서 사용되고 있으나, 이의 항아토피 효능에 대한 연구는 거의 진행되지 않았다. 본 연구에서는 아토피성 피부염의 주요 증상인 피부장벽 기능과 면역이상 개선에 대한 고삼추출물의 효과를 평가하였다. 고삼추출물은 피부장벽 기능에서 중요한 역할을 하는 각질세포막의 형성을 강화하는 결과를 나타내었다. 또한 피부의 보습작용에 있어서 중요한 히알루론산의 발현을 증가시키는 결과를 나타내었다. 아토피성 피부염 병변에서 특이적으로 증가하는 황색포도상구균에 대한 고삼추출물의 효능도 확인하였으며, 고삼추출물이 황색포도상구균으로부터 유도된 전염증성사이토카인의 생성을 억제함을 확인하였다. 또한 피부 스트레스 등으로 부터 생성되는 신경전달 물질인 substance P에 의해 유도된 전염증성사이토카인의 발현도 억제하는 것을 확인하였다. 이러한 결과들은 고삼추출물이 피부장벽기능과 면역반응 개선을 통해 아토피 피부염 치료에 사용될 수 있는 잠재적 후보물질임을 제시한다. Atopic dermatitis (AD) is a common and multifactorial inflammatory skin disease that is characterized by skin barrier dysfunction, inflammation, and chronic pruritus. AD has a complex etiology that includes genetic, immunological, and environmental factors that cause skin barrier abnormalities and immune dysfunctions. Sophora flavescens (SF) has been used in traditional Chinese medicine, but little research has been conducted on its anti-AD efficacy. In this study, we evaluated the effect of SF extract (SFE) on improving skin barrier function and immune abnormalities, which are the main symptoms of AD. SFE has the capacity to enhance the formation of cornified envelope (CE) that plays an important role in the skin barrier function. In addition, it was confirmed that SFE increased the expression of hyaluronic acid related to skin moisture. The effect of SFE against Staphylococcus aureus (S. aureus), which increases specifically in AD lesions, confirmed that SFE inhibited the production of pro-inflammatory cytokines induced by S. aureus. Furthermore, SFE was shown to inhibit the expression of pro-inflammatory cytokines induced by substance P (SP), the cause of skin neurogenic inflammation. These results demonstrate that SFE could be one of potential candidate agent for the treatment of AD by improving the skin barrier function and immune responses.

        The objective of this study is establishing of an alternative method using bovine cornea to rabbit eye irritation test. The bovine corneal opacity and permeability assay is checking opacity and permeability of bovine cornea after exposure of chemical irritants. In this study, we measured the opacity of the bovine cornea by quantifying the ability of light to pass through it using opacitometer. In addition, we optimized the basal opacity level before treatment of irritants to bovine corneas. With these results, we established the standard operating procedure of bovine corneal opacity assay.

        녹차와 밤송이로 구성된 복합추출물의 in vitro 항여드름 효능연구

        This study aims to investigate the anti-acne efficacy of the plant extract complex (PEC) consisting of Camellia sinensis L. leaf and Castanea crenata bur extract in vitro. The effect of PEC on anti-acne target including sebum production, Cutibacterium acnes (C. acnes)-induced inflammation, 5-alpha reductase activity and free radical scavenging activity was observed. The level of lipid production was measured through nile red staining in sebocytes to confirm the effectiveness of inhibiting sebum production, and C. acnes cultured medium (C. acnes CM) were treated on HEK293-hTLR2 cells to observe TLR2 activity. In addition, regulation of 5-alpha reductase activity was confirmed using rat liver microsomes, and antioxidant capacity was measured by DPPH assay. As a result, PEC showed the inhibitory effect on sebum production up to 59.02±5.13% at 100 μg/mL and the C. acnes CM with PEC treated group dramatically inhibited the TLR2 activity as much as the control level. At 100 μg/mL of the PEC, the activity of 5-alpha reductase enzyme activity was suppressed by about 20% compared to the group treated with rat microsomes, and the radical scavenging capacity was measured at 86.83±10.19. These results suggest that PEC, a complex extract of Camellia sinensis L. leaf and Castanea crenata bur, can be used as natural cosmetic raw materials that can relieve acne through inhibition of excessive sebum production, reduction of 5-alpha reductase activity, anti-inflammatory, and antioxidant properties.

        사회공포증 환자의 성격요인과 방어기제에 대한 연구

        연구목적: 사회공포 환자의 성격요인과 방어기제의 특징을 알아보고, 비가해형 및 가해형에 따라 유의미한 차이가 있는지 알아보고자 본 연구를 시행하였다. 방 법: 강북삼성병원 외래치료 환자중 DSM-Ⅳ에 근거하여 사회공포로 진단된 환자 28명을 대상으로 성격요인검사(Sixteen Personality Factor Questionnaire 한국판) 및 이화방어기제검사를 시행하였고 통계처리는 각 검사에 대한 빈도분석과 비가해형과 가해형에 대한 Independent-Sample t-test를 시행하였다. 결 과: 사회공포 환자에서 성격요인(평균점수) 중 shyness/boldness 요인이 정상보다 낮은 소견을 보였으며 방어기제에서는 유우머 항목이 정상보다 낮은 소견을 보였다. 성격요인과 방어기제 모두에서 비가해형과 가해형간에 유의미한 차이는 보이지 않았다. 결 론: 성격요인의 결과상 사회공포 환자들은 감정 표현을 억제하며 비사교적이다. 또한 자기주장을 못하여 타인에게 순종적, 동조적이며 정서적 불안정감이 심하다. 이러한 성격요인들은 모두 자율신경계의 약화, 불안의 증가, 공포증의 유발가능성의 증가등으로 사회공포와 관련되어 있다. 또한 방어기제의 결과상 사회공포 환자들은 유우머가 부족하고 지나치게 긴장, 경직되어 있어 사회공포 증세를 유발하기 쉬운 것으로 판단되었다. 사회공포의 두 유형인 비가해형과 가해형간에는 성격요인, 방어기제 모두에서 유의미한 차이가 없는 것으로 나타났다. Objectives: The purpose of this study was to evaluate the characteristics of personality factor and defense mechanism and to compare the difference between non-offensive and offensive types of social phobia patients. Methods: Study subjects consisted of 28 social phobia patients diagnosed with the criteria of DSM-Ⅳ. Data were collected through Sixteen Personality Factor Questionnaire and Ehwa Diagnostic Test of Defense Mechanism. We employed was the independent sample t-test for the analysis. Result: Among the personality factor, the shyness/boldness factor scored lower than normal in social phobia patients. Humor defense scored lower than normal among defense mechanisms. There was no significant correlation between non-offensive and offensive types of social phobia in personality factors and defense mechanisms. Conclusions: Social phobia patients were shier and less frequently use humor defense than the ordinary people. There was no significant correlation between non-offensive and offensive types of social phobia patients in personality factors and defense mechanisms.

        Andrographolide의 Extracellular Signal-regulated Kinase Pathway (ERK)를 통한 상피 세포 줄기세포능 향상

        Andrographolide, the main compound of Andrographis paniculata (A. paniculata), shows various biological properties including anti-viral, anti-inflammatory, anti-diabetic, and hepatoprotective effects. Our previous study has shown that A. paniculata extract exerts antiaging effects by activation of stemness in epidermal stem cells (EpSCs). In this study, we investigated the effect of andrographolide as a main compound of A. paniculata on EpSCs and its mechnism of action using several in vitro assays. Andrographolide increased the proliferation of EpSCs and induced cell cycle progression. Additionally, andrographolide increased VEGF production and the expression of stem cell markers integrin β1 and p63. Furthermore, phosphorylation levels of extracellular signal-regulated kinase 1/2 (ERK1/2), S6 ribosomal protein (S6RP) and Akt were increased by andrographolide. Taken together, these results indicate that andrographolide-induced proliferation of EpSCs is mediated by the ERK1/2, Akt-dependent pathway with increased production of VEGF and upregulated stemness through integrin β1 and p63.

        Anti-acne Properties of Artemisia annua Extract In Vitro

        여드름은 털피지샘단위(pilosebaceous unit)와 관련된 만성 염증 피부 질환으로, 여드름 병변에서는 피지 과다분비(hyperseborrhea)나 이상분비(dysseborrhea), 염증반응, 그리고 다른 피부상재균들에 비해 증가된 Cutibacterium acnes (C. acnes)로 인한 피부 미생물 균총의 불균형이 관찰된다. 이 연구는 개똥쑥 추출물(Artemisia annua extract: AAE)의 항여드름 효과를 확인한 것으로, 피부 미생물 균총의 균형유지와 관련된 항균효과와 염증반응의 억제, 과도한 피지 분비의 감소 측면에서 실험을 진행하였다. C. acnes와 Staphylococcus epidermidis(S. epidermidis)를 AAE와 공동배양 하였을 때, S. epidermidis의 성장률은 저해되지 않았지만 C. acnes의 성장률은 저해된 것을 확인하였다. 또한 AAE를 처리하여 배양한 C. acnes 배양배지를 세포에 처리하였을 때, 인터루킨-1β(IL-1β), 종양괴사인자-α(TNF-α)와 인터루킨-6(IL-6) 같은 사이토카인 분비의 감소와 TLR2 활성 억제도 확인하였다. 마지막으로 피지세포에 AAE를 처리한 결과, 팔미트산에 의해 유도된 피지형성을 감소시키는 것을 확인하였다. 이 결과들은 AAE가 다양한 타깃을 지닌 천연추출물로써 여드름의 주요 원인들인 C. acnes의 선택적 성장저해와 C. acnes로부터 유도되는 염증반응을 억제할 수 있으며, 과도한 피지형성을 감소시켜 결과적으로 여드름을 완화시키는 물질로 사용될 수 있다는 것을 제시한다. Acne vulgaris is a chronic inflammatory skin disease related to pilosebaceous unit. In acne lesions, hyperseborrhea, dysseborrhea, inflammatory event, and an imbalance in skin microflora, particularly an increase in Cutibacterium acnes (C. acnes) colonization comparing to other bacteria, have been observed. The objective of this study was to evaluate anti-acne effects of Artemisia annua extract (AAE) on antibacterial activity related to preservation of the balance in skin microbiome, inhibition of inflammation, and reduction of excessive sebum production. When C. acnes and Staphylococcus epidermidis (S. epidermidis) were co-cultured in the presence of AAE, the reduction of C. acnes growth by AAE was greater than that of S. epidermidis. In addition, when C. acnes was cultured in a medium containing AAE (C. acnes AAE), levels of cytokines such as interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) and IL-6 and toll-like receptors-2 activity were decreased in comparison with C. acnes cultured in a medium without AAE (C. acnes CM). Moreover, AAE significantly inhibited excessive sebum production induced by palmitic acid. These results suggest that AAE, as a natural extract with various targets, can inhibit selective growth of C. acnes and inflammatory reactions derived from C. acnes, which are the main causes of acne, and consequently can be used as a substance to alleviate acne by reducing excessive sebum formation.
