        당뇨 합병증과 군령탕 구성성분의 네트워크 약리학 분석 및 효능 예측

        Objectives : GunRyeong-Tang(GRT) is a traditional herbal prescription that combines Oryeongsan and Sagunja-tang. This study employed network analysis methods on the components of GRT and target genes related to diabetes complications to predict the improvement effects of GRT on diabetes complications. Methods : The collection of active compounds of GRT and related target genes involved the utilization of public databases and the PubChem database. We selected diabetes complication-related genes using GeneCards and confirmed their correlation through comparative analysis with the target genes of GRT. We constructed a network using Cytoscape 3.9.1 and conducted topological analysis. To predict the mechanism, we performed functional enrichment analysis based on Gene Ontology (GO) biological processes and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Results : Through network analysis, 234 active compounds and 1361 related genes were collected from GRT. A total of 9,136 genes related to diabetes complications were collected, and 1,039 target genes overlapping with the components of GRT were identified. The core genes of this network were TP53, INS, AKT1, ALB, and EGFR. In addition, GRT significantly reduced the H9c2 cell size and the expression of myocardial hypertrophy biomarkers (ANP, BNP), which were increased by high glucose (HG). Conclusions : Through this study, we were able to predict the activity and mechanism of action of GRT on diabetes and diabetic complications, and confirmed the potential of GRT as a treatment for diabetes complications through the effect of GRT on improving myocardial hypertrophy for diabetic cardiomyopathy.

        제생신기환이 허혈-재관류로 유발된 급성 신부전 마우스에 미치는 효과

        Renal ischemia-reperfusion injury(IRI), an important cause of acute renal failure (ARF), cause increased renal tubular injury. Jesaeng-sinkihwan (JSH) was recorded in a traditional Chines medical book named “Bangyakhappyeon (方藥合編)”. JSH has been used for treatment of diabetes and glomerulonephritis with patients. Here we investigate the effects of Jesaeng-sinkihwan (JSH) in a mouse model of ischemic acute kidney injury. The animals model were divided into four groups at the age of 8 weeks; sham group: C57BL6 male mice (n=9), I/R group: C57BL6 male mice with I/R surgery (n=9), JSH Low group: C57BL6 male mice with surgery + JSH 100 mg/kg/day (n=9) and JSH High group: C57BL6 male mice with surgery + JSH 300 mg/kg/day (n=9). Ischemia was induced by clamping the both renal arteries during 25 min, and reperfusion was followed. Mouse were orally given with JSH (100 and 300 mg/kg/day during 3 days after surgery. Treatment with JSH significantly ameliorates creatinine clearance(Ccr), Creatinine (Cr) and blood urea nitrogen(BUN) in obtained plasma. . Treatment with JSH reduced kidney inflammation markers such as Neutrophil Gelatinase Associated Lipocalin (NGAL) and kidney injury molecule-1 (KIM-1). JSH also reduced the periodic acid schiff (PAS) staining intensity and picro sirius red staining intensity in kidney of I/R group. These findings suggest that JSH ameliorates tubular injury including renal dysfunction in I/R induced ARF mouse.

        SDV 패러다임을 통한 자동차 디자인 변화 및 제안 - 인테리어 디자인을 중심으로 -

        Background The mobility revolution is evolving as software and hardware are gradually fused in automobiles, a functional hardware transportation. It is necessary to examine in detail the concept of innovative components and the direction of arrangement change necessary for the convergence design born by this revolution.Methods Most automobile manufacturers study the functions and characteristics of LEVEL 4 autonomous driving level 4 and the unnecessary steering system.& It is showing a concept car with LEVEL 5's steering system as an option. This interior composition was divided into stages A &B, and studied through a review of papers and references on the spatial composition and arrangement required at each stage. In this study, we tried to define, analyze, and propose the components, focusing on stage B suitable for the user-centered design of autonomous driving.Result Through the relationship between the smartphone revolution and future mobility and case analysis, a new concept of interior design components was defined and the convergence of hardware and software was derived. The limitations and suggestions of this study were that software controls the functions and performance of internal and external hardware that constitutes future mobility, and that interactions between pedestrians and passengers should be supplemented.Conclusion The convergence design proposed in this study is expected to contribute to the efficient and innovative development of components for future mobility interior design. In order to change the value chain of the automobile industry, it is necessary to propose more innovative and destructive designs in the process of user-centered design and development with SDV-centered convergence design. A lot of discussion is needed in the future to develop the interior design and establish the direction of future mobility, and we hope that this study will serve as a cornerstone for contributing to this. 연구배경 모빌리티 혁명은 기능적인 하드웨어 이동수단인 자동차에서 점차 소프트웨어와 하드웨어가 융합되어 진화하고 있다. 이 혁명으로 태동한 융합디자인에 필요한 혁신적인 구성요소의 개념과 배열 변화의 방향성에 관하여 구체적으로 살펴볼 필요가 있다. 연구방법 대부분의 완성차 업체들은 LEVEL 4 & LEVEL 5의 조향장치를 옵션으로 하는 컨셉트 카를 선보이고 있다. 이러한 인테리어 구성을 A & B단계로 구분하고, 각 단계에서 필요로 하는 공간 구성과 배열에 관한 논문과 참고문헌 등의 고찰을 통하여 연구하였다. 본 연구에서는 자율주행의 사용자 중심 디자인에 적합한 B 단계에 초점을 두고 그 구성요소들을 정의하고 분석하여 제안하고자 하였다. 연구결과 스마트폰 혁명과 미래 모빌리티와의 연관성 및 사례 분석을 통해 인테리어 디자인 구성요소들의 새로운 개념을 정의하고 하드웨어와 소프트웨어의 융합을 도출하였다. 소프트웨어로 정의된 차량(Software Defined Vehicle,이하 SDV)이 미래 모빌리티를 구성하는 내외장 하드웨어의 기능과 성능을 소프트웨어가 제어한다는 설정과 보행자와 탑승자 간의 인터랙션 보완이 필요하다는 점이 본 연구의 제한점이자 제안이라는 결과를 얻었다. 결론 본 연구에서 제안한 융합디자인은 미래 모빌리티 인테리어 디자인에 효율적이고 혁신적인 구성요소 개발에 기여할 것으로 사료 된다. 자동차 산업의 가치 사슬을 변화시키기 위해서는 SDV 중심의 융합디자인으로 사용자 중심의 설계와 개발 과정에서 더욱 혁신적이고 파괴적인 디자인을 제안해야 한다. 이러한 미래 모빌리티의 인테리어 디자인 개발과 방향성 정립을 위해 앞으로도 많은 논의가 필요하며, 본 연구가 이에 기여하는 초석이 되길 기대해 본다.
