        효모의 전사억제 유전자인 YDR1 의 기능 연구를 위한 온도감수성 변이주의 분리

        The human Dr1 (hDr1) gene represses transcription by interacting with TATA-binding protein and by preventing formation of transcription preinitiation complexes. The YDR1 gene of the yeast Saccharomyces cerevisiae, which is the counterpart of hDr1, is an essential gene for viability and its overexpression is deleterious to the cells. Nine ydr1 mutants were isolated either by error-prone PCR or by hydroxylamine mutagenesis followed by plasmid shuffle. Four ydr1 mutants were FOA-sensitive, suggesting that the ydr1 alleles are not functional. The other five ydr1 mutants were conditional mutants at 16℃ or 37℃. DNA sequence analysis revealed the nature of the mutations. Three of the four FOA-sensitive ydr1 strains resulted from nonsense mutations, which are R34 (ydr1-4), S50 (ydr1-5), and R118 (ydr1-2). Among conditional mutants the ydr1-7 mutant (L48P) showed cs at 16℃ and the other four ydr1 alleles showed ts at 37℃ (E35K, ydr1-3 ; S40F, ydr1-1 ; E67L, ydr1-6 ; Q96L, ydr1-8). The five base substitutions were positioned within the histone-fold motif in the YDR1 gene, suggesting that the histone-fold motif is important for the Ydr1 function. The ydr1 alleles obtained in this project will be useful to generate revertants to uncover suppressor genes involved in transcription.

        비유가 수업에 대한 흥미와 세포기관의 기능 이해에 미치는 효과

        The structure and the function of the cell should be learned in the biology course of high school. Students have difficulty in studying these topics because terms of cell parts are strange to them and the function of each cell part was explained separately in the textbooks. The purpose of this study is to investigate the effects of systematic analogies on student motivation and understanding of the function of cell parts. For this study. 126 11th graders in 4 classes were sampled from a high school in the city of Jeonju in Korea. The students were divided into two groups for implementation of the program, one experimental group and one control group. In the experimental group the teaching material containing factory analogy was used, which was focused on the function of cell parts, while the contents of a science textbook were taught in the control group. The results of this study were very informative. Students in the experimental group showed higher records at the achievement test, and the differences between the two groups were statistically significant. And students in the experimental group showed higher interest about the teaching method and the learning content than those in the control group. Interestingly, students in the experimental group who presented their own analogies after 6 months showed remarkable understanding of the function of cell parts.

      • 유전 개념의 이해를 위한 염색체 모형 이용 수업의 효과

        Mendelian genetics is one of major themes in the secondary level of biology education. But students have difficulty in studying it because of the abstract and dynamic attributes of genetic concepts. The purpose of this study is to investigate the effects of dynamic model on student motivation and understanding of genetic concepts. For this study, 234 10th graders in 6 classes were sampled from a high school in Seoul, Korea. The students were divided into three groups for implementation of the program, two experimental groups and one control group. The model with dynamic and stochastic attributes was shown to students by a teacher in the experimental group 1, while it was used by students in experimental groupⅡ, In the control group, the contents of a science textbook were taught in the traditional way. The results of this study were very informative. Students in the experimental groupⅡ showed higher records at the achievement test, and the differences were statistically significant. And students in the experimental groupⅡ showed higher interest about the teaching method and the learning content than those in the experimental group I and the control group.

        호르몬 작용 이해를 위한 동적 비유 모형 수업의 효과

        Homeostatic control by hormone is a basic concept at the 'Stimulus and Response' part in Biology. But secondary school students have difficulty in studying this topic because conceptual relations are abstract and complex and there are too many things to memorize. Therefore the purpose of this study was to develop a dynamic analogical model with which students can simulate the control of blood sugar level and to examine the effects of conception learning by using it. For this study. 73 filth graders in 4 classes were sampled from a high school in the city of Jeonju in Korea. The students were divided into two groups for implementation of the program, one experimental group and one control group. In the experimental group the dynamic analogical model which was developed in this study was used, while the contents of a science textbook were taught in the control group. The results of this study were very informative. Students in the experimental group showed a higher record at the achievement test and attitude test, and the differences between the two groups being statistically significant. In addition. interviews with students in the experimental group provided evidence that the simulation activity was interesting and helpful in understanding hormonal control mechanism by changing students' mind from matter category to process category. The use of the model was found to be effective for improving the understanding of the concepts of hormonal control.
