Streptomces sp. MET 0515의 균체외 Alginate lyase의 정제 및 특성

알긴산분해효소(EC 4. 2. 2. 3)를 균체외로 생산하는 새로운 방선균을 전라남도 완도의 연안토양으로부터 분리하고 이 균주를 Streptomyces sp. MET 0515라 명명하였다. 이 균주가 생산하는 균체외 알긴산분해효소를 아세톤 침전, 음이온 교환 크로마토그래피(Q-Sepharose and DEAE-Sepharose), 젤 크로마토그래피(Sephacryl S-200 HR gel filtration chromatography)를 이용하여 정제하였다. 이 효소의 최적 활성 온도과 pH는 각각 $70^{\circ}C$와 pH 7.5이고, 온도 안정성은 $0-50^{\circ}C$, pH 안정성은 pH 6.0-9.0였다. 이 효소는 $Mn^{2+}$ 이온 첨가시에는 활성이 증가하였으며 1mM $Fe^{3+}$, 1mM EDTA, 1mM $Zn^{2+}$ 이온 첨가시에는 활성이 억제되었다. 또한 이 효소는 poly-alpha 1,4-L-guluronate와 poly-beta 1,4-D-mannuronate두 종류의 기질에 대하여 활성을 나타냈다. 따라서 본 효소는 다른 미생물 기원의 효소와 비교해 볼 때 Streptomyces sp.가 생산하는 첫번째 효소로 인정되었다. We isolated a new extracellular alginate lyase-producing microorganism, which displayed alginate-depolymerizing activity in plate assays, from coastal soils in Wando, Jeollanam-do, Korea. This alginate-depolymerizing bacterium belonged to the genus Streptomyces and it was named Streptomyces sp. MET 0515. An extracellular alginate lyase(ALY1) secreted by Streptomyces sp. MET 0515, was purified to homogeneity by a combination of acetone precipitation, anion-exchange chromatography (Q-Sepharose and DEAE-Sepharose) and Sephacryl S-200 HR gel filtration chromatography. Its molecular mass was 26 kDa as determined by SDS-PACE analysis. The enzyme had an optimal temperature of $70^{\circ}C$ for its activity, and was most active at pH 7.5. The thermal and pH stability were $0-50^{\circ}C$, and pH 6.0-9.0, respectively. The enzyme activity was stimulated by 1mM $Mn^{2+}$, and inhibited by 1mM $Fe^{3+}$, 1mM EDTA and 1mM $Zn^{2+}$. Preliminary analysis of substrate specificity showed that this alginate lyase had activity on both poly-alpha 1,4-L-guluronate and poly-beta 1,4-D-mannuronate in the alginate molecule.
기능성 소화불량증 환자의 피로도와 기혈수 변증의 상관성 분석

Objectives: The aim of this study was to investigate the correlation between fatigue degree and Comprehensive diagnosis of Qui Xui Shui in patients with functional dyspepsia. There again, we investigated an association between scores obtained from Comprehensive diagnosis of Qui Xui Shui and state of skin sympathetic tone or cardiovascular reactivity. Methods: Subjects were 56 patients with Functional dyspepsia and 8 healthy People. Fatigue degree was assessed by questionnaires consisting of subjective complaints of fatigue. Comprehensive diagnoais of Qui Xue Shui was investigated by questionnaire and physical examination, total score and composition score were calculated from the symptom score. Skin sympathetic tone was measured by Ryodoraku Score(RS) and Cardiovascular Reactivity(CV Reactivity) was checked by Pulse diagnostic apparatus. All patients were divided into two groups by RS $40{\mu}A$(below and above) or by CV Reactivity(decreased and increased or not decreased). Results: There was highly significant correlation between Fatigue Degree and total score and each composition score of Comprehensive diagnosis of Qui Xui Shui. And total score of patients was significantly higher than that of health controls : in the patients, females than males. Significance related with the level of RS $40{\mu}Aor$ the state of CV Reactivity were not remarkable in both patients and health control group. Conclusions: Fatigue degree seems to have a special feature reflecting the state of Comprehensive Diagnosis of Qui Xui Shui in patients with functional dyspepsia.
생맥산(生脈散)의 방사선 보호효과 : 생쥐 소낭세포 재생과 Apoptosis에 미치는 영향

Backgrounds & Objects: The aim of this study was to investigate the radioprotective effect of Shengmai-san(SMS), a herbal medicine, on mice jejunal crypt cell survival and Apoptosis. Methods: Mice were devided into 4 groups according to radiation dose and SMS treatment: Normal was the group without irradiation. Control was the group treated with D.W before 10 Gy irradiation. SMS 2.9 was sample group treated with 2.9 mg/10 g of SMS extract before 10 Gy irradiation and SMS 29 was sample group treated with 29 mg/10 g of SMS extract before 10 Gy irradiation. And Each group were sacrificedat 24 hours and 72 hours after irradiation. To analyze the crypt survival, hematoxylin-eosin staining was used and to analyze the apoptosis, the TUNEL assay was done. Results: 1. From the microcolony survival assay, the SMS 2.9 and SMS 29 showed the radioprotective effect with a statistical significance compared to the control group at 24 hr (P < 0.01) and 72 hr (p < 0.001) after 10 Gy irradiatien. And the differences of radioprotective effect between SMS 2.9 and SMS 29 were net significant. 2. The results of the TUNEL assay showed that the apoptotic index in SMS 2.9 and SMS 29 was significantly decreased, as compared to the control group at both 24 hr ( p < 0.01) and 72 hr (SMS 2.9 : p < 0.001. SMS 29 : P < 0.01) after 10Gy irradiation And the differences of between SMS 2.9 and SMS 29 were not significant. Conclusions: It could be suggested that the Shengmai-san has a prominent Protective effect in mice intestines against the radiation damage. And the radieprotective effect seems to be related to inhibition of the apoptosis.
COVID-19의 유행으로 인한 스트레스가 대학생의 삶의 질에 미치는 영향: 가족건강성의 조절효과

Objectives: As COVID-19 drags on, university students experience more stress and feel more anxious about their studies and career plans. Against the backdrop, this study aims to analyze the moderating effect of family health on the relationship between the spread of COVID-19 and the quality of life in university students. Methods: In order to achieve the purpose of this study, the results of an online survey performed with 216 university students in cities K and D were analyzed using frequency analysis, factor analysis, correlation analysis, regression analysis, and moderating effect analysis. Results: It was found that COVID-19 stress had a significant negative effect on the quality of life in university students, while family health showed a significant positive effect. In addition, the moderating effect of family health was statistically significant on the relationship between COVID-19 stress and the quality of life in university students. Conclusions: This study found that family health plays an essential role as a safety net in reducing stress and improve the quality of life in university students amid the social crisis of the COVID-19 pandemic. In this regard, it is required to develop various programs for communication with parents, including parent-child communication techniques.
다문화·탈북 가정 학생의 과학 학업성취도 특성 및 화학 문항 분석

본 연구에서는 전수로 시행된 국가수준 학업성취도 평가의 중학생 과학 성취도를 다문화 탈북 집단별 성취특성, 문항별 집단별 정답률 및 변별도, 내용영역별, 정답률 차이가 큰 문항유형별 특성을 계량적, 질적 방법으로 분석하였다. 연구결과, 다문화 탈북 가정 학생의 과학과 학업성취도는 일반 학생에 비해 매우 낮았으며, 기초학력과 기초미달 학생의 비율이 매우 높았다. 또한 다문화 탈북 가정 학생의 하위집단별로 과학과 학업성취도가 다르게 나타났으며, 특히 중도입국 학생과 탈북 가정 학생의 학업성취도가 가장 낮았다. 일반 학생과의 정답률 차이가 큰 문항을 분석한 결과, 중도입국 학생과 탈북 가정 학생의 경우는 전 영역에서 고르게 취약한 반면, 외국인 가정 학생의 경우는 특정 영역에서 취약함을 보였다. 이로부터 다문화 탈북 가정 학생을 위한 교수 학습 및 교육 지원이 각각 집단의 상황을 고려하여 적합한 방식으로 이루어지도록 교육적 함의를 도출하였다. In this study, we analyzed the characteristics of science academic achievement by multiple culture and North Korean migrant students types, percentage of correct answer and discrimination per items per groups, and contents domains using quantitative and qualitative method in the National Assessment of Educational Achievement (NAEA). As a results of analysis, the degree of science academic achievement of the students from multiple culture and North Korean is much lower than that of the general students. Also, the science academic achievement of the students from multiple culture and North Korean migrant is different from the general students. Especially, science academic achievement of the immigrated students and the North Korean migrant students was lowest. The analysis of items that the difference of the percentage of correct answer was large showed that the students from multiple culture and North Korean migrant were equally weak to it in all contents domains, but that the students from international marriage family born in country were weak in some areas. Therefore, the appropriate the teaching and learning method and the educational support is needed considering the group situation, so the educational implications are discussed.
