To elucidate the action of $P_2$-purinoceptor, effects of adenosine triphosphate(ATP) and perivascular nerve stimulation were investigated from polygraph in the isolated renal artery of rabbit 1. ATP caused the relaxation on the precontraction with noradrenaline$(10{\mu}M)$ on the presene and absence of endothelium in the isolated renal artery of rabbit, the relaxative response was increased between 0.1 and $30{\mu}M$ on dose-dependent manner. 2. The relaxative response induced by ATP$(10{\mu}M)$ on precontraction with noradrenaline$(10{\mu}M)$ was blocked by the pretreatment with reactive blue 2$(10{\mu}M)$. 3. ATP inhibited the contractile response by perivascular nerve stimulation(0.3ms, 80V, 50Hz, 1 sec), the inhibitory action was blocked by the pretreatment with 8-phenyltheophylline$(10{\mu}M)$.
Ethanol 이 고양이 신장 Na-K-ATPase 활성에 미치는 영향

The effects of ethanol on Na-K-ATPase activity were investigated with cat kidney homogenate. The results were summarized as follows: 1. Na-K-ATPase activity was inhibited with dose-dependent manner by ethanol of higher concentration than 1%, and showed an estimated $I_{50}$ (the inhibitor concentration to cause 50% inhibition) of 7.5%. 2. Hydrolysis of ATP was linear with the incubation time in the absence and presence of 8% ethanol, whereas it was different with preincubation time in the presence of 15% ethanol. 3. Inhibition of Na-K-ATPase activity by ethanol was not affected by increased enzyme concentration, and showed the reversibility of the inhibitory pattern. 4. Kinetic studies of cationic-substrate activation of Na-K-ATPase showed that ethanol had both properties of classical competitive inhibition for $Mg^{{+}{+}}$ or $K^+ and non-competitive inhibition for ATP or $Na^+$. 5. Arrhenius plot yield two break point at $21^{\circ}$ and $30^{\circ}C$ in the absence of ethanol, whereas showing only one break point at $18^{\circ}C$ in the presence of 8% ethanol. These results suggested that ethanol inhibited Na-K-ATPase activity reversible through a disturbance of microenvironment of lipids associated with the enzyme.

Tuberculous lymphadenitis is not uncommon in Korea. Therefore, an inexpensive, safe and rapid method is needed to diagnose the tuberculous lymphadenitis. Flne needle aspiration cytology Is a good method for this purpose, but has several limitations in the diagnosis of tuberculous lymphadenitis, especially when the presence of acid-fast bacilli is not proved. To evaluation the usefulness of the polymerase chain reaction with enzyme immunoassay technique in the detection of Mycobacterium tuberculosis (M. tuberculosis) In the cervical Iymph node asplrates, the authors performed fine needle aspiration cytology and M. tuberculosis PCR with enzyme immunoassay for mycobacterial DNA sequences from 15 cases of the fine needle aspirates. Cytomorphologically, the cases were categorized into three types: predominantly necrotic materials; typical epithelioid cell granulomas with or without slant cells and caseous necrosis; and non-tuberculous lesions, such as reactive lymphadenitis, abscess, metastatic carcinoma and malignant lymphoma. M. tuberculosis DNA was found in 8 of 15 cases by PCR with enzyme immunoassay. Negative findings on PCR were achieved in 7 cases, which revealed non-tuberculous tymphadenopathy. In conclusion, we suggest that M. tuberculosis PCR with enzyme immunoassay using the fine needle aspirates is a very useful tool for the diagnosis of tuberculous lymphadenitis.

To validate the physiological properties of the histamine receptors of ileal smooth muscle in dog, the effects of adrenergic-, cholineric-, and H-receptor antagonists on the responses of ileal smooth muscle strips to histamine were investigated. The results were summarized as follows; 1. Histamine caused the contraction of ileal smooth muscle and the contractile responses were increased between the concentration of histamine $10^{-7}M$ and $10^{-5}M$ with dose-dependent manner in dog. 2. The shorter the treatment interval of histamine, the lower the contractile activity until the treatment interval extended to 40 minutes. 3. The contractile response induced by histamine was completely blocked by the pre treatment with a $H_1$-receptor blocker, chlorpheniramine and not by the pretreatment with a $H_2$-receptor blockers cimetidine. 4. The contractile response induced by histamine was not blocked by the pretreatment with a cholinergic receptor blocker, atropine. 5. The contractile response induced by histamine was not blocked by the pretreatment with an ${\alpha}$-adrenergic receptor blocker, phenoxybenzamine, or a ${\beta}$-adrenergic receptor blocker, propranolol. From these results, it was suggested that the contraction induced by histamine was elicited through $H_1$-receptor on the ileal smooth muscle in dog.
돼지 심관상동맥의 이완작용에 대한 purinergic 신경의 효과

To elucidate the effects of purinergic nerve on relaxation of pig coronary artery, the effects of ATP, 2-methylthio ATP and electrical perivascular nerve stimulation were investigated from physiograph on the isolated coronary artery of pig. The results btained were as follows; 1. The relaxative responses induced by perivascular nerve stimulation(20V, 0.5msec, 10sec) were the frequency(1~8Hz) dependent manner with phentolamine($10^{-5}M$) and atropine($10^{-6}M$) on isolated coronary artery of pig. 2. The relaxative responses induced. by adenosine($10^{-7}{\sim}5{\times}10^{-3}M$) or ATP($10^{-7}{\sim}5{\times}10^{-5}M$) on precontraction with histamine($10^{-5}M$) were the dose-dependent manner, but the contractile responses were often induced by ATP($10^{-4}M$ and $10^{-3}M$). 3. The relaxative responses induced by 2-methylthio ATP($2.5{\times}10^{-8}{\sim}2.5{\times}10^{-6}M$) on precontraction with histamine($10^{-5}M$) were the dose-dependent manner. 4. The relaxative response induced by 2-methylthio ATP($10^{-7}M$) on precontraction with histamine($10^{-5}M$) was completely blocked by the pretreatment with $P_{2Y}$-purinoceptor blocker, reactive blue 2($10^{-4}M$). 5. The neurogenic relaxative response induced by perivascular nerve stimulation(20V, 8Hz, 0.5msec, 10sec) was weakly inhibited by the pretreatment with ${\beta}$-adrenoceptor blocker, propranolol($10^{-5}M$) and blocked by the addition with $P_{2Y}$-purinoceptor blocker, reactive blue 2($10^{-4}M$). The results suggest that the purinergic nerve is innervated, and its relaxative response was mediated by $P_{2Y}$-purinoceptor on isolated coronary artery in pig.
미성숙 돼지 자궁 평활근의 운동성에 대한 Adenosine Triphosphate와 Prostaglandin $F_{2{\alpha}}$ 작용의 관계

This study was carried out to investigate the action of adenosine triphosphate (ATP) on the motility of immature pig uterine smooth muscle. ATP appeared contractile responses in a dose-dependent manner, showing the maximal contraction at the concentration of $10^{-3}M$ in the uterine smooth muscle strip. The contractile responses by $ATP(10^{-4}M)$ were not affected by atropine $(10^{-6}M)$, phentolamine $(10^{-6}M)$, propranolol $(10^{-6}M)$, pyrilamine $(10^{-6}M)$, cimetidine $(10^{-6}M)$, and theophyulline $(5{\times}10^{-5}M)$, but were inhibited uncompetitively by quinidine. The effects of these drugs on the contractile responses by prostaglandin $F_{2{\alpha}}(PGF_{2{\alpha}})$ were also comparable to those observed with ATP. When muscle strips were pretreated with indomethacin $(5{\times}10^{-5}M)$ for 20 min., the contractile responses by $ATP(10^{-4}M)$ were completely inhibited. But the contractile responses by $PGF_{2{\alpha}}$ were not affected by indomethacin. These results suggest that ATP elicited the contraction through noncholinergic- and nonadrenergic-receptor mediated by prostaglandin $F_{2{\alpha}}$ in pig uterine smooth muscle.
