In this study we assessed the influences of ultraviolet (UV) light B radiation on epidermal ATPase-positive dendritic cell (DC) and the effect of green tea treatment in ICR mouse. The extent of changes following 200 mJ/㎠ (0.5 mW/sec) was studied at 0, 6, 12, 18, 24, 30 or 36 hours after exposure. SBCs were decreased by 6 hours after irradiation. There was tendency to decrease from 6 hours to 24 hours and had little further change from then to 36 hours after irradiation. The mice that received 0, 50, 100, 200, 300 or 400 mJ/㎠ of UVB were examined 24 hours after irradiation. The DCs were decreased as the radiation dose increases from 100 to 400 mJ/㎠. The frequency of UVB (200 mJ/㎠)-induced DC decrease was reduced by treatment of green tea (i.p. and topical application, p<0.01).

Cytogenetic and hematological analysis was performed in peripheral blood of pig in the high background radiation area (HBRA) (Goesan-gun) and control area. The frequency of micronuclei (MN) in peripheral blood lymphocytes from pig was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. When analysed by Iinear-quadratic model the line of best fit was y=0.0102D²+0.0250D+0.011[y=number of MN/cytokinesis-blocked (CB) cells and D=irradiation dose in Gy]. MN rates per 1,000 CB lymphocytes of pig from HBRA and control area were 9.20±3.88 and 11.00±2.98, respectively. There were no significant differences in MN frequencies and hematological values in pig between HBRA and control area.

This study was performed to determine the effect of Phyllostachys nigra var. henenis Strapf leaf extract on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of gamma-radiation. Phyllostachys nigra var. henenis Strapf administration before irradiation (I.P.: 125 mg/kg of body weight, at 24 hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.01). The frequency of radiation-induced apoptosis was also reduced by pretreatment of Phyllostachys nigra var. henenis Strapf (I.P.: 280 mg/kg or 28 mg/kg of body weight, at 24 hours before irradiation, p<0.01). These results indicated that Phyllostachys nigra var. henenis Strapf might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of Phyllostachys nigra var. henenis Strapf and its components.

We induced the activation of melanocytes in the epidermis of C57BL/6 mice by ultraviolet B (UVB) irradiation and observed the effect of green tea extract (GTE) on the formation, and decrease of UVB-induced epidermal melanocytes. C57BL/6 mice were irradiated by UVB 80 mJ/㎠ (0.5 mW/sec) daily for 7 days, and GTE was intraperitoneally or topically applied before or after irradiation. To estimate change of epidermal melanocytes, light microscopic observation with dihydroxyphenylalanine (DOPA) stain was performed. Split epidermal sheets prepared from the ear of untreated mice exhibited 11-13 melanocytes/㎟, and one week after UV irradiation, the applied areas were increased number of strong DOPA-positive melanocytes with stout dendrites. But intraperitoneal or topical treatment with GTE before each irradiation interrupted UVB-induced pigmentation and resulted in a marked reduction in the number of epidermal melanocytes as compared to the number found in UVB-irradiated and untreated skin. The number and size of DOPA-positive epidermal melanocytes of intraperitoneally or topically applicated GTE group were also significantly decreased as compared to that of only UV irradiated group at 3rd and 6th weeks after irradiation. The present study concludes GTP has a potential of inhibitor on UVB-induced pigmentation and depigmenting agent.

방사선 및 각종 독성물질에 의한 고환 정세관세포의 사멸은 apoptosis와 관련이 있다고 알려져 있으나 정세관상피주기에 따른 apoptosis 발생에 대한 변화연구는 미진하다. 본 연구에서는 감마선을 조사한 ICR 마우스의 고환에서 apoptosis 발생을 transferase-mediated end labeling (TUNEL) 과 periodic acid-Schiff (PAS) 염색을 동시에 실시하여 관찰하였다. Apptosis는 TUNEL 양성으로 나타났으며 특징적 형태변화를 보였다. 2 Gy (분당 2 Gy의 선량률)의 방사선을 조사하고 24시간동안의 변화를 관찰한바 방사선조사 후 12시간에 가장 높은 apoptosis 발생을 보였고 이후 감소하였다. 8 Gy까지의 방사선을 조사하고 8시간에 변화를 관찰한 결과 모든 정세관상피 주기에서 방사선 용량에 비례한 apoptosis의 발생이 관찰되었다. 방사선 용량-반응은 linear-quadratic 곡선[y=(-0.014±0.009)D²+(0.31±0.697)D+0.3575. y는 정세관 당 TUNEL 양성세포의 수, D는 방사선 용량(Gy), r²=0.9]에 가장 일치 하였다. 최대 반응은 8 Gy에서 관찰되었으며, 0.5 Gy조사군에서도 변화가 나타났다. 이러한 변화는 정세관상피주기 V에서 B정조세포와 정세관상피주기 XII의 분열기 정자세포에서 가장 현저하였다. The killing of male germ cells by radiation and other toxicants has recently been attributed to apoptosis, but a critical evaluation of the presence of the different features of apoptosis in each epithelial stage has not been performed. In this study, mouse testes exposed to radiation were examined by light microscopy and terminal transferase-mediated end labeling (TUNEL) with periodic acid-Schiff (PAS) stains to determine whether the cells were apoptotic according to several criteria. Apoptosis was easily recognized by the presence of peroxidase-stained, entirely apoptotic bodies. In the TUNEL-positive cells or bodies, the stained products correlated precisely with the typical morphologic characteristics of apoptosis as seen at the light microscopic level. The changes that occurred from 0 to 24 hours after exposing the mice to 2 Gy of gamma-rays (2 Gy/min) were examined. The numbers of apoptotic cells reached a peak at 12 hours after irradiation and then declined. The mice that received 0-8 Gy of gamma-rays were examined 8 hours after irradiation. Dose-response relationships were generated for each stage of the epithelial cycle by counting TUNEL-positive cells. The dose-response curves were linear-quadratic [y=(-0.014±0.009)D²+(0.31±0.697)D+0.3575. Where y=the number of apoptotic cells per seminiferous tubule, and D=the irradiation dose in Gy, r²=0.9] and there was a significant relationship between the frequency of apoptotic cells and the radiation dose. Although the maximum response was produced by 8 Gy, even 0.5 Gy induced marked changes. These changes were most pronounced in B spermatogonia of stage Ⅴ and the spermatocyte at the mitotic cells of stage ?.

In this study we assessed the influences of ultraviolet (UV) light B radiation on epidermal cells by apoptotic sunburn cell (SBC) and the effect of green tea treatment on the inhibition of SBC formation in SKH1-hr mouse. The extent of changes following $200mJ/cm^2$ (0.5 mW/sec) was studied at 0, 3, 6, 12, 18, 24, 30 or 36 hours after exposure. SBCs were recognized by 3 hours after irradiation. There was tendency to increase from 3 hours to 24 hours and decrease from then to 36 hours after irradiation. The mice that received 0, 25, 50, 100, 200, 400 or $800mJ/cm^2$ of UVB were examined 24 hours after irradiation. The SBCs were induced as the radiation dose increases from 0 to $200mJ/cm^2$. A further increase of radiation dose has little further effect. The frequency of UVB ($200mJ/cm^2$)-induced SBC formation was reduced by intraperitoneal injection of green tea extract (p<0.01).

Transgenic mouse models have been introduced and accepted by regulatory bodies as an alternative to carcinogenicity assay models to predict and evaluate chemical carcinogens. The recent research outcomes in transgenic mouse models have made progressive advances in the understanding of chemical carcinogenesis and the evaluation of potential human carcinogens. However, these models still remain to be insufficient assay systems although the insufficiencies have been recognised and are being resolved. Based on up to date information from literature, this review article intends to understand currently accepted transgenic mouse models, issues arising from study design, interpretation of the study, results of validation project and their cancer prediction rate, and further perspectives of cancer assay models from the regulatory view point.
