잣나무 약 배양에 의한 callus 유도

잣나무의 雄花를 5月 18日~25日에 採取하여 四分子期에서 未成熟花粉에 이르는 段階의 葯을 培養한 結果 callus를 誘導하였다. G.D 基本培地에 BAP 0.1ppm 및 NAA 0.1~1ppm을 組合으로 處理한 培地에서 callus發生이 좋았으며, ½濃度의 M.S培地(Sucrose 2%, Agar 0.8%)에 BAP 0.1ppm과 NAA 0.5ppm을 加한 경우에도 좋았다. callus의 chromosome數는 12인 haploid도 있었으나 대부분 24로서 diploid였다. 生長이 旺盛한 callus는 體細胞에서 起因된 것으로 생각되어 앞으로 chromosome의 變異 等을 더욱 調査하여 ploidy의 특성을 究明할 수 있으리라고 생각된다.

Effects of Spinal and Peripheral Injection of α1A or α1D Adrenoceptor Antagonists on Bladder Activity in Rat Models with or without Bladder Outlet Obstruction

Purpose: Antagonists of α1-adrenergic receptors (α1ARs) relax prostate smooth muscle and relieve voiding and storage symptoms. Recently, increased expression of α1ARs with change of its subtype expression has been proved in bladder outlet obstruction (BOO). To search for the evidence of changes in α1ARs subtype expression and activity in the peripheral and spinal routes, the effects of spinal and peripheral administration of tamsulosin (an α1A/D-selective AR), naftopidil (an α1A/D-selective AR), and doxazosin (non-selective AR) on bladder activity were investigated in a rat model with or without BOO. Methods: A total of 65 female Sprague-Dawley rats were divided into the BOO surgery group (n=47) and the sham surgery group (n=18). After 6 weeks, cystometry was assessed before and after intrathecal and intra-arterial administrations of tamsulosin,naftopidil, and doxazosin. Results: After intra-arterial administrations of all three drugs, bladder capacity (BC) was increased and maximal intravesical pressure (Pmax) was decreased in both BOO and the sham rat models (P<0.05). After intrathecal administration of all three drugs, BC was increased and Pmax was decreased in only the BOO group. The episodes of involuntary contraction in the BOO rat models were decreased by intra-arterial administration (P=0.031). The increase of BC after intrathercal and intra-arterial administrations of α1ARs was significantly greater in the BOO group than in the sham group (P=0.023, P=0.041). In the BOO group, the increase of BC and decrease in Pmax were greater by intra-arterial administration than by intrathecal administration (P=0.035). There were no significant differences of the degrees of changes in the cystometric parameters among the three different α1ARs. Conclusions: Up-regulations of the α1ARs in BOO were observed by the greater increases of BC after α1AR antagonist administrations in the BOO group than in the sham group. However, there were no subtype differences of the α1ARs in functional parameters of bladder activity. In addition, α1ARs also act on the lumbosacral cord which implies that the sensitivity of α1ARs is increased in pathologic models such as BOO. Further evaluation including differential expression of α1ARs in BOO models are need.

Streptomyces coelicolor A3(2)의 Acetyl Xylan Esterase를 발현하는 Escherichia coli의 과산화수소 저항성

Streptomyces coelicolor A3(2)의 acetyl xylan esterase (AxeA)가 Escherichia coli의 과산화수소 저항성에 미치는 영향을 알아보고자 하였다. AxeA 발현은 isopropyl-$\beta$-thiogalactoside로 유도되었고 생산된 AxeA는 SDS-polyacrylamide gel electrophoresis방법으로 확인하였다. AxeA 발현에 따른 과산화수소 저항성의 변화를 E. coli의 생장곡선과 생존율을 통하여 조사하였다. AxeA가 발현되지 않으면 모든 처리 농도 (1 mM, 2.5mM, 5mM)에서 균의 사멸이 일어났다. AxeA가 발현되는 조건에서는 5mM을 제외한 과산화수소 1mM와 2.5mM에서 E. coli의 사멸이 저지되었다. 또한 1.5mM의 과산화수소에 대한생존율이 59%에서 74%로 높다졌다. 동시에 E. coli의 최고생장온도에서에 근접한 $45^{\circ}C$에서의 생존율도 증가되는 결과를 얻었다. 그러므로 AxeA 단백질은 산화적 스트레스와 온도스트레스에 대해 교차 저항성을 나타내는 역할을 한다고 결론지었다.

Whang, Soon-Won's IL-WEOL is to be comprehended as follows in the analysis of the archetype criticism. First of all, it is caused by racial remembrance said by Karl Jung that In-Chul intuitively felt blood-relation with himself on seeing Bon-Dol grandfather whom he had never seen before, and he also felt the same blood as his on seeing the grandfather when he served as a chief governer in Kwang-Ju. And it being also because In-Moon raised a toad, a white rat, and a snake etc. in his own house, and In-Ju liked a play best in spite of herself among a lot of hobbies, the butcher in the olden time is symbolically expressed as a rogue role and a jester role. Accordingly it means that In-Moon and In-Ju remembered that their ancestors had also been butchers. It is classified as follows if it is criticised by the special archetype of Karl Jung which means persona, shadow, and the self. Sang-Jeon grandfather's persona is presented in the veil of president of Dae-Ryuk Company, which was just for his own living. And In-Ho's persona is only for his success in life and his won existence on earth keeping his social position as a chief governer in a district. In such conditions of mind, he easily abandoned even relationship between father and son. On the other hand Bondol grandfather's persona is presented in the veil a murderer for the sake of his son's future. Shadow is presented in In-Moon's character, who is not on good terms with others and so just raises a toad, a white rat, and a snake in his house. The self is presented in In-Chul, who recognized himself through the destruction of Bon-Dol grandfather, Gee-Ryong, and In-Chul's family. Whang, Soon-Won's IL-WEOL may be understood as heroic archetype ; recognition of 'the Self' of In-Chul and racial remembrane are quest and a series of pain that In-Chul's family experienced are 'Initiation' which a hero passed by, and it is only pretense for redemption with a view to taking 'the Self' that In-Chul's family was ruined. IL-WEOL is contrastive to each other in the constitution as shown in In-Chul's 'the Self' and Na-Mee's 'Construction of a house', Park, Hae-Yeon's 'Play', and In-Chul's 'Dream', and 'A knife, and the formation of T word'. In addition to those listed above, Mrs. Hong and In-Moon, Gee-Ryong and Ester are also contrastive to each other. In the last place analyzing it by virtue of 'N.Frye's archetype of four seasons', it is an archetype of spring when he was born that In-Chul was in search of everything to find out 'the Self', every occurrence and episode which In-Chul produed in order to find out 'the Self' is summer symbolizing that a hero comes into the paradise, it is autumn which is an archetype of tragedy that In-Chul's family was in catastrophe, and it is an archetype of winter that everything took to pieces and was annihilated in the last part. Therefore In-Chul acquired 'the Self' by the help of sacrifice of his family in the end. Accordingly IL-WEOL is a literary work which is composed of circular archetype of spring, summer, autumn, and winter. And red color is archetype of blood, sacrifice, and intensive passion. As a result Park, Hae-Yeon's 'Dream' and the color 'Red' which anybody else symbolizes altogether symbolize a butcher.

Voiding Dysfunction after Total Mesorectal Excision in Rectal Cancer

Purpose: The aim of this study was to assess the voiding dysfunction after rectal cancer surgery with total mesorectal excision (TME). Methods: This was part of a prospective study done in the rectal cancer patients who underwent surgery with TME between November 2006 and June 2008. Consecutive uroflowmetry, post-voided residual volume, and a voiding questionnaire were per¬formed at preoperatively and postoperatively. Results: A total of 50 patients were recruited in this study, including 28 male and 22 female. In the comparison of the preopera¬tive data with the postoperative 3-month data, a significant decrease in mean maximal flow rate, voided volume, and post-voided residual volume were found. In the comparison with the postoperative 6-month data, however only the maximal flow rate was decreased with statistical significance (P=0.02). In the comparison between surgical methods, abdominoperineal resection pa¬tients showed delayed recovery of maximal flow rate, voided volume, and post-voided residual volume. There was no significant difference in uroflowmetry parameters with advances in rectal cancer stage. Conclusions: Voiding dysfunction is common after rectal cancer surgery but can be recovered in 6 months after surgery or ear¬lier. Abdominoperineal resection was shown to be an unfavorable factor for postoperative voiding. Larger prospective study is needed to determine the long-term effect of rectal cancer surgery in relation to male and female baseline voiding condition.

산림토양으로 부터 내산성 streptomyces sp. 균주의 분리 및 동정

In this study, an acidoduric Streptomyces strain was isolated and identified from acidic forest soil around Dankook University, Cheonan Campus. This isolated strain had rod-shaped, smooth, non-motile spore and the shape of spore chain was compact spiral. This structure appeared similar to the sporangium of the genus Streptosporangium but this strain proved to be a Streptomyces strain by an electron microscopic study and cell wall analysis. This strain showed a best growth on neutral medium, was also able to grow on the acidic media of pH 4.0 and pH 5.0. The color determination of this strain on various agar media and other physiological tests were carried out by ISP-methods. From these results, the isolated strain was considered to be Streptomyces mirabilis. 본 실험에서는 숲속 토양으로부터 산성조건에서 생장이가능한 내산성 균주를 분리 동정하였다. 분리된 균주는 형태적 관찰과 박층 크로마토그래피에 의한 세포벽 분석과 ISP 분류 기준에 따른 생리적 실험을 통하여 동정하였으며, 이 균주의 포자는 형태가 간상형이고 표면구조는 매끈한 형태였다. 포자의 배열형태는 compact spiral 구조였으며 이 균주는 명확한 포자낭벽을 갖고 있지 않았고, 세포벽 분석결과에 따르면 Streptomyces 속에 속하는 것으로 밝혀졌다. 또한 중성조건에서 생장이 우수하나 산성조건에서도 생장이 가능한 내산성 균주로써, Nonomura의 분류 기준에 의하면 Streptomyces mirabilis인 것으로 동정되었다.
