        자궁내막증 병변에서의 Cytokine mRNA의 발현 양상

        목적: 자궁내막증은 최근에 그 빈도가 급격하게 증가되고 있으나 아직까지 병인과 치료법이 확실하게 정립되지 못하고 있는 부인과 내분비의 대표적인 질환이다. 최근 들어서 자궁내막증 환자의 불임의 원인으로 cytokine이 밀접하게 관여됨이 밝혀지고 있고 자궁내막증의 병인으로 복강내로 역류된 월경혈에 의한 복강내의 국소적 염증에 대한 개체의 반응정도와 면역체계의 변화가 중요하게 인정되고 있으나 아직까지 정확한 기전은 밝혀져 있지 않기 때문에 이에 저자는 자궁내막증 환자의 복수 내에서의 cytokine의 정량측정과 골반내 자궁내막증 병변조직에서의 cytokine mRNA의 발현 양상을 조사하여 자궁내막증 환자의 병인분석을 시도하였다. 연구방법: 자궁내막증 진단을 받고 복강경 혹은 개복 수술을 받은 30명의 환자에서 총 60개의 자궁내막증 병변이라고 의심되는 조직을 채취하여 이를 RT-PCR법을 이용하여 Cytokine 유전자의 발현양상을 조사하였다. 또한 7례의 정상대조군과 23례의 자궁내막증 환자의 복수를 ELISA법을 이용하여 정량분석을 시도하였다. 결과: 자궁내막증 환자의 복수내에서 IL-6와 IL-10의 농도는 자궁내막증의 임상적 중증도에 따라서 의미있게 증가되어 있었으나 IFN-γ, TNF-α, IL-1β, 그리고 IL-5의 농도는 정상 대조군에 비해서 변화가 없었다. 모든 예의 심부 자궁내막증 병변조직과 표재성 자궁내막증 병변조직에서 IL-1β cytokine mRNA의 발현을 볼 수 있었다. IL-5와 IL-6은 표재성 병변 12개 중에서 각각 2개의 black lesion에서만 발현을 볼 수 있었으며 IL-10은 표재성 병변에서는 12개중 2개에서 그리고 심부성 병변에서는 8개중 1개의 조직에서만 발현되었다. IFN-γ는 표재성 병변에서는 전혀 발현이 없었으나 심부성 병변에서는 8개중 4개의 조직에서 발현이 되었으며 TNF-α는 표재성 병변에서는 red 및 black 병변에서 각각 1개의 조직에서만 발현이 되었으나 심부성 병변에서는 역시 8개중 4개의 조직에서 발현이 되었다. 결론: 표재성 병변이 골반강내에 착상하여 염증성 반응이 일어날 수 있는 원인이 제공되면 IL-1β 혹은 TNF-α같은 염증성 cytokine이 분비가 되며 이로 인해 생성되는 단핵세포의 chemotactic factor에 의해 대식세포의 증가와 활성화가 이루어지고 이어서 복강내에 IL-6 등의 cytokine이 증가되며 마지막으로 여러 가지 증가된 cytokine에 대한 반대 반응으로 IL-10이 증가됨을 추정할 수가 있겠으며 이러한 가정은 앞으로 cytokine을 이용한 치료적 응용의 기초적인 연구로서 중요한 의미를 제공할 수 있다고 하겠다. Objective: The pathogenesis of endometriosis is generally accepted that retrograde menstruation and alterations in the local pelvic immune environment. This study was performed to help elucidate what kind of role various cytokines might play in the pathogenesis of endometriosis. Method: Concentrations of peritoneal fluid cytokines were compared in 7 women with normal pelvic finding and 23 women with endometriosis by enzyme-linked immunosorbent assay(ELISA). The patterns of cytokine mRNA expression in 8 ovarian endometrioma and 12 superficial pelvic endometriosis lesions were investigated by reverse transcription-polymerase chain reaction(RT-PCR) amplification method. Result: Both IL-6 and IL-10 levels in peritoneal fluid specimens with endometriosis tended to be higher than normal. However, there were no significant differences between peritoneal fluid concentrations of IFN-γ, TNF-α, IL-1β, and IL-5 of women with and without endometriosis. The levels of IL-6 and IL-10 were significantly higher in peritoneal fluid of women with severe endometriosis compared to women with mild endometriosis. IL-1β mRNA was expressed in all of 8 deep and 12 superficial endometriosis lesions. IL-5 and IL-6 mRNA were expressed in only two black lesions respectively, however, both were not expressed in the all deep lesions. Expressions of IL-10 mRNA occurred in one red and one black lesion while this was expressed in only one of the deep lesions. TNF-α mRNA was expressed in one red and one black lesion of 12 superficial lesions compared with four of the deep lesions. There was the difference between kinds of increased cytokines in the peritoneal fluid and those of expressed cytokines in the endometriotic lesions of patients with endometriosis. Conclusion: This study supports the concept that local immunologic factors may be important in the pathogenesis and pathophysiology of endometriosis. The pattern of cytokine mRNA expression of endometriotic lesions would seem to indicate that proinflammatory cytokines such as IL-1β and TNF-α are responsible for the development or progression of endometriosis.

        누에와 육계 복합 추출물의 in vivo 면역증진 기능성 연구

        Objectives : The aim of this study was to investigate the immunopotentiating activity of combine extract that Silkworm and Cinnamomum cassia . Recently, acute epidemic diseases such as cold and viral respiratory diseases have been emerging. So, interested in immunity enhancement has been increasing, and research on natural products to promote immunity activity has been actively conducted. Methods : To confirm the immunopotentiating activity effect, Silkworm (SW), Cinnamomum cassia (CC), and SWCC combined extracts were treated 14 days at 300 ㎎/kg/day. The changes of glutamic oxalacetic transaminase (GOT), glutamic pyruvate transaminase (GPT) in serum were analyzed after experiment. The changes in the total spleen cell number were measured. Immune cells in spleen were analyzed using fluorescence activated cell sorter (FACS). also, analyzed the expression of cytokines in spleen. Results : Total number of cells in the spleen and FACS analysis of T lymphocytes activated in the spleen showed that the SWCC combined treated group had much higher frequency of active cells than both single groups. The ratio of CD4+CD8+, CD4+CD69+ and CD4+CD25+ T cells in spleen, SWCC is higher than other groups except Nor in CD4+, CD4+CD69+, CD4+CD25+ T cells. The results of this study suggest that SWCC can help immune function via IL-2, IL-10, IL-12, IFN-γ cytokine production, increased T lymphocytes and splenocyte proliferation. Conclusion : Therefore, these results suggested that the SWCC combined extracts administration increase stronger immunity enhancement than when SW and CC adminstration.

        LC/MS/MS를 이용한 축산밀집지역 하천에서 테트라사이클린계 항생제 및 대사체 분석 연구

        The purpose of this study was to optimize an analytical method for the detection of tetracycline antibiotics and their metabolites in water and to determine their fate and movement in streams. To analyze three tetracycline antibiotics and eight of their metabolites, water quality samples were taken from nine sites, which were scattered across upstream areas, into which livestock manure was directly released from nearby livestock operations, to downstream areas, between November 2010 and October 2011. These samples were pretreated with hydrophilic-lipophilic balance (HLB; 200 mg, 6 cc) cartridges and analyzed using liquid chromatography/ tandem mass spectrometry (LC/MS/MS). Consequently, the limit of detection of the analysis method for the tetracycline antibiotics and their metabolites ranged from 3.7 to 18.2 ng/L. The recovery rates were 81-122%, and the relative standard deviation was less than 16%. In addition, the concentration of tetracycline antibiotics and their metabolites decreased or they were undetected when sampled from the upstream to downstream sites. Although the levels of tetracycline antibiotics and their metabolites detected were lower than their EC50, it is necessary to continue monitoring these substances in the environment.

        누에, 맥문동복합물의 흉선 림프절내 T세포 활성 유도 효능 연구

        Objectives : The purpose of this study is to investigate effects on the immune system of Bombyx mori L. and Liriopis seu Ophiopogonis Tuber mixture (BL) in Thymus, Lymph Nodes. Methods : Eight-week-old male Balb/c mice were divided into five groups : Group one included the normal mice (Nor). Positive control group two administrated with red ginseng (RG) 100 mg/kg . Group three administrated with Bombyx mori L. (BX) 300 ㎎/㎏. Group four administrated with Liriopis seu Ophiopogonis Tuber (LP) 300 ㎎/㎏. Group five administrated with the mixture of Bombyx mori L. and Liriopis seu Ophiopogonis Tuber (BL) 300 ㎎/㎏. After 2 weeks administration, mice were sacrified and antigen receptor in Thymus, Lymph Nodes was analyzed by using Fluorescence Activated Cellorter Sorting (FACS). we counted the total of Thymus and Lymph Nodes cells. GOT (glutamlc oxaloacetic transaminase), GPT (glutamlc pyruvic transamlnase) in serum were analyzed after experiment. Results : In Effects of Nor, RG, BX, LP, BL on the ratio of CD4+CD8+, CD4+CD69+ and CD4+CD25+ T cell in Thymus and Lymphnode, BL is higher than other groups except Nor in CD4+, CD4+CD69+, CD4+CD25+ T cell. The number of Thymus and Lymph Nodes increased in BL. In the level of GOT and GPT, BL decreased comparing to others group except Nor. Conclusions : BL may have effect on T cells in Thymus, Lymph Nodes. In addition, Bombyx mori could be immune functional material with others herb materials.

        연구논문 : 웅진기 백제양식 연대시론

        최근 공주지역에서 많은 고고자료가 조사되었다. 그럼에도 불구하고 기존의 설에 따르면 웅진기의 고고자료의 수는 아직 미미하다. 이러한 원인은 『三國史記』에 기록된 한성의 일시적 함락과 웅진천도를 백제 한성양식의 하한으로 설정한 것에서 기인한다. 본고에서 검토한 결과 한성지역에서 최소한 6세기1/4분기까지 백제 한성양식의 기조가 유지되었다. 또한 최근 조사 보고된 공주 수촌리유적Ⅱ지점의 2호 목곽묘를 제외한 나머지 분묘와 연기 송원리유적의 대표적인 분묘인 KM016, KM046 석실묘는 웅진기로 편년되었다. 이 편년에 근거하면 금동제 관모, 식리 등의 위세품은 개로왕대에 시행된 왕·후·태수제의 관작 또는 웅진기에 일반화된 담로제와 관련된 유물일 가능성이 높은 것으로 해석되었다. 이는 한성 중앙에서 지방 지배를 위해 사여된 물품이라는 기존의 설과는 다른 해석이다. 한성지역에서 백제 한성양식이 최소한 6세기1/4분기까지 유지되었고, 왕·왕족의 장례와 국가 의례적 목적으로 조성된 정지산유적이 520년경으로 편년되었다. 몽촌토성과 무령왕릉 호석에서 일본 고분시대의 토기인 TK23형식의 須惠器가, 정지산유적에서 TK47형식의 須惠器가 출토되었다. 이것을 감안하면 須惠器의 연대는 현재 일본에서 통용되는 연대보다는 일본의 고전적 연대와 영남지역의 도질토기와의 교차연대에 의한 연대관이 보다 타당한 것으로 판명되었다. Recently, many archeological relics were investigated in Gongju region. Nonetheless, the number of archeological relics at Woongjin period is still small according to the conventional theory. The reason is that the temporary surrender of Hanseong and the transfer of Capital to Woongjin recorded in 『Samguk Sagi(三國史記)』 is set as the lowest limit of Baekje Hanseong style. According to the results of study in the paper, the basis of Baekje Hanseong style has been maintained in Hanseong region at least until the first quarter of the 6th century. Besides, stone chamber tomb KM016 and KM046 that is the typical tomb of Yeongi Songwon-ri tombs, and other tombs except the outer coffin tomb no.2 of recently investigated and reported Gongju Suchon-ri Ⅱplace was dated at Woongjin period. On the basis of this chronology, it is highly possible that prestige goods such as gilt-bronze crown and gilt-bronze Copper Shoes were relics relating to the official rank of the king·peers·exarchal system put in force in the King Gaero era or Dam`ro system generalized in Woongjin period according to the analysis. This is an analysis different from the conventional theory that those were goods granted by the metropolis Hanseong for Regional Ruling Syst. Baekje Hanseong style has been maintained in Hanseong region at least until the first quarter of the 6th century. And Jeongjisan(艇止山) remains built for the funeral of king·royal family and national ceremony were dated approximately at the year 520. Given that Sueki(須惠器) of TK23 type, pottery at the period of ancient burial mounds in Japan, was unearthed at Mongchon earthen fortress and circular burial protection stone of the tomb of King Muryong, and Sueki(須惠器) of TK47 type was unearthed from Jeongjisan remains, it was proved that cross dating between Japanese classical chronology and stoneware of Yeongnam region is more valid than the chronology of Sueki(須惠器), which holds true in Japan at the present time.

        자궁경부 원추절제술후 잔류종양에 대한 임상병리학적 관련인자

        N/A Objective : To evaluate the clinicopathologic variables that are important for predicting residual disease after cervical conization in hysterectomy specimens. Methods : We performed retrospective review of 125 patients who had undergone cervical conization and endocervical curettage followed by subsequent hysterectomy. Pathologic findings of the cone margins, endocervical curettage samples, and residual lesions in the postcone hysterectomy specimens were recorded for analysis. Results : The prevalence rates of positive cone margins increased with higher severity of cervical neoplasia. There was a statistically significant difference in the prevalence of residual disease in hysterectomy specimens between patients with positive endocervical margins on cone biopsy(67.3%) and those with negative margins(11.8%) (p<0.01). Most residual lesions in the postcone hysterectomy specimens were similar to cone pathology, however the possibility of more advanced lesions in the uterus was present in patients with positive cone margins. Residual disease was significantly more frequently found in patients with positive endocervical curettage(82.4%)(p<0.01). Especially, when both the endocervical margin and endocervical curettage were positive, residual disease was present in 25 of 28 (89.3%) hysterectomy specimens. Conclusions : From these results, we concluded that the status of endocervical margin and endocervical curettage have significant predictive value with respect to the presence or absence of residual disease, and final decision in regard to subsequent management should be based on histologic finding of the cone margins and endocervical curettage as well as the patient's age and desire to retain reproductive capability.

        체지방 감소 고기능성 발현 떫은 감 소재 개발 연구

        Objectives : This study was conducted to evaluate the antioxidant activity and inhibitory activity of pancreatic lipase in vitro using natural products sangju dungsi, chungdo bansi, kabju baekmok three kind of young persimmon fruit extracts and to develop a high - functional anti - obesity substance with reduced body fat. Methods : The antioxidative effect of the three kind of young persimmon fruit extracts was determined by hot water extraction, 30% EtOH extraction and 70% EtOH extraction. The antioxidant activities were evaluated through radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6- sulphonic acid) (ABTS) radicals. Furthermore, we examined total phenol and flavonoids contents. And the pancreatic lipase inhibition was measured by selecting the sangju dungsi extract with antioxidant effect. Results : The yield of water, 30% and 70% EtOH extract from young persimmon fruit was sangju dungsi (water 8.3%, 30% EtOH 18.1%, 70% EtOH 16.9%), chungdo bansi (water 16.2%, 30% EtOH 16.1%, 70% EtOH 11.6%), kabju baekmok (water 16%, 30% EtOH 16.8%, 70% EtOH 11.5%). The sangju dungsi 30% EtOH extract eminent DPPH (IC<sub>50</sub>, 19.25 ± 2.94 ㎍/㎖) and ABTS (IC<sub>50</sub>, 19.94 ± 4.07 ㎍/㎖) radical scavenging activity, and total phenol 52.37 ± 0.78 ㎎/g and flavonoid 7.97 ± 0.13 ㎎/g contents were also excellent. It also showed excellent pancreatic lipase inhibition efficacy. Conclusion : The extracts from the three kind of young persimmon fruit extracts showed excellent antioxidant activity. sangju dungsi 30% EtOH extract may be a new anti-obesity material by reducing fat absorption through the inhibition of pancreatic lipase.
