        햄스터 난소세포에서 Daidzein이 항산화효소활성과 발현에 미치는 영향

        Reactive oxygen species (ROS) are produced in the metabolic process of oxygen in cells. The superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in cells systemize the antioxidative enzymes to control the oxidative stress. Daidzein is one of the isoflavonoids, and its role in controlling cellular oxidative stress is presently the active issue at question. In this study, we analyzed daidzein-induced survival rates of the CHO-K1 cells, activities of antioxidative enzymes, ROS levels, and expression levels of antioxidative enzyme genes in order to investigate the effect of daidzein on cellular ROS production and antioxidative systems. As results, the survival rate of cells was decreased as the dose of daidzein increases (12.5~200μM). Daidzein increased cellular ROS levels in a dose-dependent manner, while it reduced total SOD activities and the expression of CuZnSOD. In conclusion, we suggest that daidzein induces oxidative stress via down relation of SOD-related mechanisms. (Cancer Prev Res 11, 321-328, 2006)

        Polypenol oxidase activity in the petals of Chysanthemum indicum var. indica Max. (f.sin-dong-a) and the leaves of Artemisia vulgaris var. indica Max was estimated by means of Warburg's manometric method. The browning reaction in owing to the oxidation of chlorogenic acid with polyphenol oxidase.

        Polyphenol oxidase plays major rele in the browning of plant tissues. The enzyme, polyphenol oxidase was partially purified from an extract of Lotus root by ammonium sulfate fractionation, which resulted in a 148 fold increase in specific activities. The enzyme was relatively heat stable, The substrate specificity of the Lotus root polyphenol oxidase showed high affinity toward pyrogallol, gallic acid and catechol, but not towards monophenol. Potassium cyanide, cysteine, potassium metabisulfite, sodium diethyldithiocarbamate were potent inhibitors. It seems that the enzyme was partially purified from Lotus root is polyphenol oxidase containing copper.

      • Adrenal Chromaffin Cell을 이용한 남용약물의 의존성 발현에 관한 연구

        This study was attempted to investigate the effects of alprazolam and narcotic opiates on catecholamine release from bovine adrenal chromaffin cell (BACC). An extensive experiment was conducted to examine the acute and chronic effects of morphine as a model of opiates on catecholamine release. In order to begin the characterization on receptor type, concentration response curve of morphine was compared with naloxone treatment. The effect of chronic exposure to inhibitory concentrations of morphine on the number of binding sites for [?H] nitrendipine binding on BACC membrane was carried out to see the development of tolerance and dependence in these cells. As a result, following findings were found; Alprazolam and morphine caused a dose.dependent inhibition on carbachol induced catecholamine release from BACC. Naloxone, opiate antagonist, enhanced the inhibition of carbachol . induced catecholamine released caused by morphine, but there was no different effect on the various concentrations. Chronic exposure to inhibitory concentration of morphine inhibited the catechoamine release induced by carbachol between the concentration of 10^(-6) and 10^(-3). The effect of chronic exposure of 4 days to 10^(-4) morphine on [?H]dihydropyridine binding in BACC showed a marked increase in the number of binding sites for [?H] nitrendipine with no significant effect on binding affinity.

      • 쑥잎 폴리페놀 산화효소의 특성

        PPO was purified from the extract of Artemisia princeps var. orientalis leaves by ammonium sulfate fractionation and sephadex G-150 column gel filtration chromatography, which resulted in a 90-fold, increase relative to homogenate without inhibitors in specific activity. The optimum temperature was 25 °C and the optimum pH was 7.0 in enzymic reaction. The best substrates were catechol and 4-methylcatechol with Km values of 3.26mM, 1.03mM, respectively. The enzyme possesses high catecholase activity but little cresolase activity. The enzyme was inhibited in the presence of sod. diethyldithiocarbamate, KCN, thiourea and EDTA. Among the divalent cations(Cu^(2+), Zn^(2+), Mn^(2+), Mg^(2+), Co^(2+), Ni^(2+), Cu^(2+) and Zn ^(2+) were activators on PPO, whereas Mg^(2+) and Co^(2+) were inhibitors on PPO.

      • 쑥잎 성장에 따른 Polyphenol 산화 효소의 활성 변화

        Polyphenol oxidases of plants are copper metalloproteins which catalize the oxidation of mono- and o-diphenols to oquinones. In this work we show total polyphenol oxidase(PPO) activity and the ratio of active form PPO to proPPO activity in the developmental stage of Artemisia princeps var. orientalis (P_(AMPAN) H_(ARA)leaf. The latent form PPO was partially purified by using Triton X -l14. The latant enzyme was activated by treatment with trypsin. Total enzyme activity in Artemisia princeps var. orientalis (P_(AMPAN)) H_(ARA) leaves polyphenol oxidase was almost constant throughout development, whereas in aged leaves declined. But the ratio of active form PPO to proPPO was increased throughout subsequent development and aging.

        This study was carried out to know the effect of tetrachloro[bis (2-chloroethyl)ethylenedi amine-N,N'] plat inum(IV) (PtCl₄(2-CEen)) on a-amylase and pepsin activity. The comparison of PtCl₄(2-CEen) with cisplatin showed at each reaction. After the PtCl₄(2-CEen) reacted with a-amylase, its antitumor activity was examined. The effect of PtCl₄(2-CEen) on a-amylase activity showed inhibition. At high concentration relatively, it appears that the effect of inhibition was high. On the contrary, the effect of cisplatin on a-amylase activity showed activation. The relative activity was decreased, according as the concentration of cisplatin increases. The reaction mixture with PtCl₄(2-CEen) showed increased activity of pepsin than control. The activity of pepsin increased with increasing concentration of PtCl₄(2-CEen). On the contrary, cisplatin showed the inhibition of pepsin activity. The ratio of inhibition was decreased gradually, according as the concentration of cisplatin increases. After the PtCl₄(2-CEen) reacted with a-amylase, its antitumor activity was almost unchanged.

      • Magnolia denudata에서 분리한 polyphenol oxidase의 정제 및 특성

        Polyphenol oxidase(PPO) was purfied from an extract of Magnolia denudata by ammonium sulfate precipitation, followed by sephadex G-150 column, which resulted in a 30.3-fold increase in specific activity. The purified preparation contained no monphenolase activity. The enzyme was most active at pH 6.0 and the temperature optimum for the PPO catechol oxidation reaction was 30℃. The enzyme was completely inactivated after heat treatment at 70℃ for 10min. Km values of the PPO for (+)-catechin, 4-methylcatechol, chlorogenic acid and dopamine derived from Lineweaver-Burk plots were 3.45×10^-3M, 1.25×10^-3M, 1.02×10^-3M, 1.96×10^-3M respectively. The enzyme more quickly oxidised (+)-catechin than other substrates used. Cysteine, glutathione, potassium metabisulfite and sodium diethyldithiocarbamate were potent inhibitors.

        We investigated the effect of pH on the polyphenol oxidase (PPO) activity in the cell wall, plastid, mitochondrial and soluble fractions of the leaves from Nicotiana tabacum L. PPO-catalized oxidation of fert-buthylcatechol were performed in the pH range 3-9. The optimal pH of PPO in,the cell wall and mitochondrial fraction are 5.0 and 6.5 respectively, the optimal pH of PPO in both plastid and soluble fractions are 7.0.

        Flavonoid was isolated from the petals of Mahnolia demudata Desr. by Sephadex LH-20 column chromatography. The flavonlid compound was identified as rutin by UV, IR, GC and NMR spectra. Aglycon, rhamnose and glucose were obtained from hydrolysis of rutin with 1% sulfuric acid. The aglycon was identified as quercetin by UV, IR, GC, NMR and MS spectra
