Monascus anka strains with higher pigment production were developed using UV mutation and natural selection. To obtain organic solvent soluble pigments from Manascus anka, the following culture conditions were compared; standing and shaking culture with Nishikawa's medium, and shaking culture with Lin's medium. Shaking culture in Lin's medium exhibited decrease in solvent.-soluble intracellular pigments after initial increase. The decrease was accompanied by the increase in water-soluble extracellular pigments. Manascus anka preferred sucrose and ethanol among 7 carbon sources tested. Treatment of sterol biosynthesis inhibitors, (±)-miconazole and chlorocholine chloride(CCC), directed carbon pool to the biosynthetic pathway leading to the pigments with CCC's more pronounced effect. Two dimensional TLC revealed at least 7 yellow pigments suggesting existence of hereto unreported pigment. One of the most abundant yellow pigments was isolated and found to be ankaflavin by NMR and MS analysis.
Artemisia annua L . 의 조직배양을 이용한 이차대사 산물의 생산

Artemisia annua contains the antimalarial principle, artemisinin. The possibility of the production of this compound through tissue culture technique was studied. The optimum combinations of hormones for the induction of callus were ρ-chlorophenoxyacetic acid(pcPA) and 6-benzylaminopurine(BAP) or pcPA and N-isopentenylaminopurine(2iP) in 0.05 ㎎/ℓ each. For the growth of callus, the same combination of pcPA and BAP was optimum in concentrations of 1.0μM and 0.5μM, respectively, and the optimal concentration of sucrose was also found to be 2%(w/v). Tissue culture from the crown gall grew faster than normal callus. In the suspension culture broth and the cells of normal callus or Agrobacterium-transformed tumors, arteannuic acid and 11,12-dihydroarteannuic acid were found together with common phyfosterols, whereas artemisinin was not found(Received February 20, 1992, accepted March 31, 1922).
Calibration of Detection System of Crack in Concrete Structure by Using Image Processing Technology

The investigation of concrete structure typically relies on visual inspection which is one of the basic inspection techniques. Image processing techniques play a crucial role in the growing field of automatic surface inspection technique. However, kinds of inspection equipment, environmental condition and detection algorithm have much influence on the reliability of inspection result. This paper proposes a verification method and testing procedure for the reliability of inspection results and surveys characteristics of image acquisition systems and crack inspection algorithms.
Simplified Model Predictive Control Method for Three-Phase Four-Leg Voltage Source Inverters

A simplified model predictive control method is presented in this paper. This method is based on a future reference voltage vector for a three-phase four-leg voltage source inverter (VSI). Compared with the three-leg VSIs, the four-leg VSI increases the possible switching states from 8 to 16 owing to a fourth leg. Among the possible states, this should be considered in the model predictive control method for selecting an optimal state. The increased number of candidate switching states and the corresponding voltage vectors increase the calculation burden. The proposed technique can preselect 5 among the 16 possible voltage vectors produced by the three-phase four-leg voltage source inverters, based on the position of the future reference voltage vector. The discrete-time model of the future reference voltage vector is built to predict the future movement of the load currents, and its position is used to choose five preselected vectors at every sampling period. As a result, the proposed method can reduce calculation load by decreasing the candidate voltage vectors used in the cost function for the four-leg VSIs, while exhibiting the same performance as the conventional method. The effectiveness of the proposed method is demonstrated with simulation and experiment results.

Claviceps, purpurea PRL 1980의 microsome에는 단백질 mg 당 40 pmole의 cytochrome P-450이 함유되어 있었다. 이 microsome으로부터 단백질 mg 당 420 pmole의 cytochrome P-450 및 387 pmole의 NADPH-cytochrome c reductase로 정제하였다. 부분 정제된 이 두 단백질로 재구성을 시도하였을 때 agroclavine hydroxylase 활성을 나타내지 않았으나 NADPH-cytochrome c reductase를 microsome에 첨가하였을 경우에는 그 활성이 80% 증가되었다. Claviceps sp. SD58의 microsome에 있는 agroclavine hydroxylase의 $V_{max}$와 $K_m$값은 agroclavine에 대하여 각각 3.3 pkat와 0.11 mM 이었다. 또 이 균으로부터 정제된 cytochrome P-450과 기질인 agroclavine의 복합체는 제2특성을 갖는 spectrum을 나타내었다. 또 위의 두 균주로 부터 분리한 cytochrome P-450이 이산화탄소와 결합할때 그 difference spectrum의 최대흡광은 449 nm 이었다. SD58 to give a type II spectrum. The absorbance maximum in the CO-reduced vs reduced difference spectrum of purified cytochrome P-450 from C. purpurea PRL 1980 and Claviceps sp. SD58 was at 449 nm.

Determination of Biosynthetic Pathway of Decursin in Hairy Root Culture of Angelica gigas

To establish the biosynthetic pathway of decursin in Angelica gigas Nakai, feeding experiment with stable isotope-labeled precursors were conducted. Umbelliferone and decursin were labeled with deuterium at C-3. The umbelliferone, the decursin, and other commercially available putative precursors, L-phenylalanine-ring-d5 and trans-cinnamic acid-d7, were fed to the hairy root culture of A. gigas. Each deuterated compound was incorporated into decursinol, decursinol angelate, and decursin as determined by mass spectrometric analysis. These findings confirmed the coumarin biosynthesis pathway sequence is composed of phenylalanine, cinnamic acid, umbelliferone, decursinol, and decursin.
