        유산균 Lactobacillus 종간의 분류를 위한 RAPD 분석법의 매개변수에 관한 연구

        유산균 Lactobacillus의 종들을 사용하여 random amplified polymorphic DNA(RAPD) 분석법에 영향을 미치는 여러 매개변수들을 조사하였다. 그람 양성균인 Lacrobacillus의 chromosomal DNA를 가장 온전한 형태로 분리하기 위하여, 세포벽 분해효소인 mutanolysin(250 U/ml)을 1시간 처리한 후 lysozyme(30,000 U/ml)을 추가로 1시간 더 처리했을 때 다량의 온전한 chromosomal DNA를 얻을 수 있었다. 이 분리된 chromosomal DNA를 template로 하여 RAPD 분석법의 몇 가지 매개변수를 조사한 결과, 사용한 시료 DNA와 Taq DNA polymerase의 양에 따라 특정한 RAPD 밴드의 농도가 증가되는 것을 알 수 있었다. 또한 primer의 양을 증가시켰을 때 역시 새로운 RAPD 밴드가 증가되었으며, 특히 2가지 이상의 매개변수를 적용하였을 경우 나타나는 RAPD 밴드의 수는 크게 차이가 났다. 한편 사용한 primer의 종류에 따라 나타나는 RAPD 밴드의 수가 변화하였는데 이는 primer의 G/C양과 무관하였다. A study was carried out to understand some parameters affecting on RAPD analysis with Lactobacillus species. From the results, we found that appearance of specific DNA bands were very influenced by the concentration of $MgCl_2$ but it was overcome by applying enough amount of Taq DNA polymerase. Other parameters such as concentrations of template DNA, random primers and Taq DNA polymerase have enhanced the production of specific DNA bands by increasing their concentration applied. However, we noticed that G/C contents of random primers did not show any correlations with number of specific RAPD bands generated but the RAPD results were heavily influenced by the characteristics of the random primers, that is, the sequences of the oli.

        Currently, majority of the companies and public institutions outsource information processing and system developing business that used to be managed by internal employees on the grounds of cost-saving, efficiency and improvement of the quality of the service. Internally, companies adopt various security solutions such as DLP to strengthen information protection, DRM, DB encryption, server access control, and network access control, to construct information protection management system. From the perspective of information processing life cycle, security level has improved in a stage of harvesting, saving and discarding. However, there are still security threats from the use and delivery stage This tendency makes it essential in law for consignment company to have supervision of management about the personal informations process of the trust company, and for companies, way to enhance security has come into request. By checking the entire internal information process managing system of the trust company and deriving a method to enhance the information protection about the outsourcing process, security for outsourcing business can be enhanced and eventually, make it possible for the safe outsourcing execution.

      • Identification of Leuconostoc mesenteroides BL-1A Isolated from Traditional Baechu Kimchi

        Isolation of lactic acid bacteria (LAB) from Baechu kimchi was attempted to study characteristics of Leuconostoc strains. All isolates, BL-1A, BL-5 and BS-1 could not grow at 36˚C but Leuc. mesenteroides subsp. mesenteroides KCTC 3722 still grew very well at the mesophilic temperature. It was found that all isolated strains except BL-1A could grow in MRS media containing 4% NaCl (w/v). All isolates as well as Leuc. mesenteroides subsp. mesenteroides KCTC 3722 showed similar patterns of hexose fermentation that they all had the same hexose fermentation pathways. BL-1A could not ferment cellobiose among tested disaccharides as same as Leuc. mesenteroides subsp. mesenteroides KCTC 3722 resulting in final acidity of pH 5.8±0.05. This kind of fermentable activity was also found in trisaccharides fermentation. The BL-1A could not ferment melezitose at all as same as Leuc. mesenteroides subsp. mesenteroides KCTC 3722 (pH 5.7±0.01) but could ferment raffinose (pH 4.7±0.1). These unique patterns of fermentation of trisaccharides are usually found in Leuconostoc species. These kinds of similarities were also found in fermentation of 7 carbohydrate derivatives. Therefore the BL-1A from Baechu kimchi can be easily identified as another subspecies of Leuc. mesenteroides. Comparative studies of growth characteristics in the presence of NaCl revealed different growth pattern for BL-1A, compared to the Leuc. mesenteroides subsp. mesenteroides KCTC 3722. That is, the BL-1A can not sustain its growth under the higher saline condition (i. e., 4% NaCl) comparing to the Leuc. mesenteroides subsp. mesenteroides KCTC 3722 which showes strong fermentable activity in the same saline condition.

        과학적 추리에서 결정 불가능의 두 유형과 그 발달

        본 연구는 추론, 문제해결, 인과추론을 비롯하여 과학적 사고에서 흔히 있는 결정불가능(indeterminacy)이 상이한 두 유형 - 경합성 결정불가능과 정체성 결정 불가능-으로 구분된다는 것을 제안하고 각각의 발달과정과 이에 관계되는 요인들을 요약하였다. 경합성 결정불가능(competitive indeterminacy)은 가능한 결론들이 동등한 자질을 갖기 때문에 야기되는 것으로서 경합되는 대안들이 무엇인지를 명시하고 반응편중효과를 적절히 통제하면 4-5세 경의 아동도 결정불가능을 잘 지각한다. 반면에 정체성 결정불가능(identity indeterminacy)은 미확인사례의 존재에서 기인하는 것으로서, 경합성 결정불가능을 잘 지각하는 아동도 정적 사례가 오직 하나만 포함되는 특정 유형의 정체성 결정불가능 상황에서는 높은 오류를 보인다. 정적 포착(positive capture)이라 불리는 이 오류는 정체성 결정불가능이 경합성 결정불가능과는 개념적으로 구분될 뿐 아니라 다른 발달기제가 있음을 시사하고 있어서 선행연구들을 개관하고 이에 대한 연구방향을 제안하였다.

      • Biochemical Characteristics of Psychrotrophs Isolated

        Isolation of psychrotrophic lactic acid bacteria (LAB) from Chonggak kimchi was attempted in order to study characteristics of LAB growing at the low temperature. Among isolated strains, CL-1 showed exactly the same fermentation characteristics of Leuc. mesenteroides subsp. mesenteroides KCTC 3722 in tested carbohydrates except melezitose and the CL-1 showed very wide range temperature profiles (16C-28C) for growth comparing to other isolates. Its lowest temperature for growth was 16C which was much lower than other isolates. However all isolates couldn’t grow at 30C. In order to identify sudden inhibition of cell growth occurred at the transition temperature (16C → 14C), the CL-1 was tested for fermentation ability of hexoses and disaccharides at the lowered temperature. Among 5 tested hexoses, significant change was resulted from mannose fermentation. The CL-1 fermented mannose very well at 24C that gave a final pH 4.7 after 48 hr incubation but at 16C its final pH was only 5.7. This means that the CL-1 could not ferment mannose for its proper cell growth at 16C and inhibition of the cell growth was to begin. Similar tendency was found in fermentation of disaccharides at the lowered temperature. Among 6 tested disaccharides, inhibition of fermentation occurred only in melibiose. The CL-1 showed decreasing fermentation ability with the melibiose from pH 4.8 at 24C to pH 5.2 at 16 C. Fermentation of maltose, sucrose and trehalose was not affected by the lowered temperature at all. Thus, such growth inhibition of the psychrotrophs by lowered temperatures is related to occurrence of inability of fermenting the sugars and it leads to sudden growth inhibition of the psychrotrophic CL-1. From the test for bile salt tolerance, the CL-1 showed strong tolerance for glycocholate as much as Leuc. mesenteroides subsp. mesenteroides KCTC 3722 but both strains were strongly inhibited by 0.5% taurocholate. In case of taurocholate, the CL-1 could overcome the bile salt effect on cell growth with 0.2% taurocholate as incubation time goes by from 12 hr to 24 hr. The taurocholate tolerance of CL-1 was much better than that of Leuc. mesenteroides subsp. mesenteroides KCTC 3722.
