We carried out an epidemiological survey of infectious myonecrosis which first occurred in farmed whiteleg shrimps in May 2015. The virus was not detected in culture water, feeds, and surrounding organisms in the IMNV-infected farms. In 2015, 5 hatcheries of whiteleg shrimps were operated in Korea, so we carried out virus tests on shrimp broodstock, shrimp feed lug worms(imported), rearing water, and surrounding organisms sampled from each of the 5 hatcheries. The tests detected the virus in one of 6 tested adult shrimps. 25~40% of the imported lug worms were also found infected, which requires us to consider need for import quarantine. The rearing water as well as the 11 species present in the infected hatcheries were also tested positive. These results require us to consider need for further research on correlations between the virus and surrounding animals in infected shrimp farms.
Ferritin is an evolutionarily conserved protein that plays an important role in iron storage and detoxification. In this study, the gene encoding a ferritin H subunit homologue (RbFH) was cloned from rock bream (Oplegnathus fasciatus) and analyzed at the expression. The full-length ferritin H cDNA was 1162 bp long and contained an open reading frame (ORF) of 531 bp that encoded 177 amino acid residues with a predicted molecular mass of 20.8 kDa. The 5` UTR was 297 bp in length, and the 3` UTR 298 bp, and preceded by a 5`-untranslated region that contains a putative Iron Regulatory Element (IRE). The deduced amino acid sequence of RbFH shares extensive sequence identities with the H ferritins of a number of fish species and contains the ferroxidase center that is preserved in ferritin H subunits. Examination of tissue specific expression indicated that RbFH expression was most abundant in PBLs, RBC, liver and muscle.
Edwardsiella tarda, Streptococcus iniae and S. parauberis are main bacterial pathogens in aquaculture farms of olive flounder, Paralichthys olivaceus. We have discussed the efficacy and safety of 3 type-combined vaccines (A: S. iniae 1mg + S. parauberis 1mg + E. tarda 1mg, B: S. iniae 1mg + S. parauberis 1mg + E. tarda 0.5mg, C: S. iniae 1.5mg + S. parauberis 1.5mg + E. tarda 1mg) through intraperitoneal injections in olive flounder. None of the vaccines gave rise to any signigicant side effects on histopathology and blood chemistry. The antibody titers and lysozyme activities of A type were higher than those of B, C and control. Four weeks after vaccination, RPS (relative percent survival rates) was 62.5~75% (A type), 50~66.7% (B type) and 55.6~62.5% (C type) respectively. As the results, the combined vaccines are possible to prevent edwardsiellosis and streptococcosis, and A type : S. iniae 1mg + S. parauberis 1mg + E. tarda 1mg, is the most effective out of them.
