Excitation-contraction coupling in skeletal muscle is process by which depolarization of the muscle fiber membrane, elicited by a nerve action potential, triggers the release of $Ca^{2+}$ from the sarcoplasmic reticulum(SR). The resulting rise in intracellular $Ca^{2+}$ concentration$([Ca^{2+}]_i)$ activates the troponin complex, thereby initiating the contraction of the muscle. The question remains as to what factors are involved in the inhibition of SR $Ca^{2+}$ release in fatigued muscle. The purpose of this study was determine whether ATP-sensitive $K^+(K_{ATP})$ channels are activated and contribute to decrease in $[Ca^{2+}]_i$ during fatigue development in the mouse skeletal muscle. To elucidate a role of $K_{ATP})$ in relation to ECC, I measured the modulation effects of $K_{ATP})$ channel blocker(glibenclamide) and opener(pinacidil) on $[Ca^{2+}]_i$ after fatiguing electrical field stimulation(FEFS). Intracellular $Ca^{2+}$ signals were recorded by conforcal laser microscopy(LSM 410) and monitored using the fluorescent $Ca^{2+}$-Sensitive indicator Fluo-3 AM. The results of this study were as followed: 1. The relative [Ca2'li after FEFS in the pre-glibenclamide-treated group was higher than the control. And relative $[Ca^{2+}]_i$ after FEFS in the pre-glibenclamide-treated group was lower than the control. 2. The relative $[Ca^{2+}]_i$ after FEFS for 3 min in the control, pre-glibenclamide-treated group and pre-pinacidil-treated group showed a similar pattern; the gradually significant decrease in $[Ca^{2+}]_i$. But, these decreasing pattern was most significant in the control. These findings suggest a tight relationship between $K_{ATP})$ and $Ca^{2+}$ in ECC during fatigue. Therefore, 1 thought that activation of $K_{ATP})$ channels may be one of mechanisms of the fatigue in skeletal muscle.

This study was performed to investigate the effect of ultrasound irradiation on epidermal growth factor(EGF) expression in rat wound tissue. Skin wounds were created below 5mm to both sides of scapular inferior angle. The right wound was used experimental side and left was used control side. Ultrasound was irradiated pulse rate $20\%$, frequency 1MHz, intensity $0.5W/cm^2$ for 5 minutes during 3days. After sonication during 3 days, rats were sacrified. The expression of epidermal growth factor evaluated immunohistochemistry on mouse anti-EGF. In the control side, a little expression of EGF was observed at epidermis and dermis. In the experimental side, A strong immunostaining was seen at epidermis and dermis. This study suggests that ultrasound irradiation is effective on EGF expression in wound tissue.

Neuromuscular junction consist of presynaptic membrane, synaptic cleft and postsynaptic membrane. In the neuromuscular junction, presynaptic membrane is the motor nerve terminal, have many synaptic vesicle. Postsynaptic membrane is the motor end plate of muscle fiber and the most striking structural features are the deep infolding of the sarcolemma. Between the nerve and muscle cells, there is a synaptic cleft of some 50-100nm. This review shows the ultrastructure and function of neuromuscular junction, summarizes the current knowledge.
흰쥐의 피부화상 후 저강도 레이저 조사가 Substance P의 발현에 미치는 영향

This study was performed to investigate the effect of low power laser irradiation on Substance P(SP) expression in the burned skin of the rats. Burns of about 3cm in diameter were created with $75^{\cric}C$ water on the back of the rats, and the lesion of experimental group were irradiated on days 1, 2, 3 and 4 postwounding. Control leasions were not irradiated. After burns, low power laser irradiation was applied by using 1000Hz, 830nm GaAlAs(Gallium-aluminum-arsenide) semiconductor diode laser. The expression of evaluated Substance P(SP) immunohistochemistry on rabbit anti-SP The results of this study wereas follows 1. The Substance P was expressed in the lamina I and II of dorsal horn of spinal cord. In expression of SP, the lesion of control group made SP to more induce significantly than experimental leasions. 2. SP immunoreactivity in burned leasion of spinal cord were decreased markedly 4 days after burns, and decreased gradually from 1 day to 2 days in burns which is laser irradiation These data suggest that low power laser have a pain release effect in the burned skin of the rats.
Plasma Matrix Metalloproteinase, Pulmonary Function, Bronchodilator Response, and Emphysema Severity

Objectives: Chronic obstructive pulmonary disease is characterized by chronic airway inflammation induced by noxious particles, and protease-antiprotease imbalance is suggested as possible pathogenesis. We evaluated the relationship between matrix metalloproteinases (MMPs) and COPD severity. Research Design and Methods: The MMP-1, MMP-8, MMP-9, and MMP-12 levels were measured for 93 subjects including COPD patients and controls. The relationship between the MMPs levels and lung function, emphysema index, bronchial wall thickness, pulmonary artery pressure, and quality of life were determined using general linear regression analysis. Results: There was significant association of MMP-1 with bronchodilator reversibility (both FVC and FEV1), MMP-8 with FVC, and MMP-9 with FEV1. MMP-1, MMP-8, and MMP-9 were correlated with emphysema index independent of lung function. However, MMP-12 was neither associated with lung function nor emphysema severity. Association of MMPs levels and bronchial wall thickness, pulmonary artery pressure, and quality of life were not significant. Conclusions: The MMP-1, MMP-8, and MMP-9 levels are associated with COPD severity, and can be used as biomarker to understand characteristics of COPD patients.

A role for ATP in nociception and pain induction was proposed. ATP-gated P2X ion channel receptors are localized throughout the nervous system and have been identified on neurons which participate in conduction of nociceptive information from the periphery to central nervous system. We consider the role of ATP as a peripheral activator of nociceptive sensory neuron via ATP-gated ion channels.

Low intensity laser irradiation is potential physical agent that triggers the muscle regeneration by previous study. In muscle regeneration, a number of growth factors also promotes that is triggered in response to muscle damage. The transforming growth factor(TGF)-$\beta$ is involved in the activation of cell proliferation and the inhibition of cell differentiation in muscle regeneration. This is secreted not only autocrine system but also paracrine and endocrine. Therefore, We investigated that effects of Gallium aluminum arsenide(GaAlAs) diode laser for the expression of TGF-$\beta$ on lumbar spinal cord after extensor digitorum muscle crush injury. After laser irradiation, the immunoreactivity of TGF-$\beta$ was increased bilaterally in gray mater of spinal cord. Especially, in 1 day, experimental group was highed than control, and in 3 day, lateral motor nucleus were storong immunoreactivy of TGF-$\beta$. Also, in 1 and 2 day, TGF-$\beta$ was showed in white mater as well as gray mater, but in 3 day, only showed in gray mater. These data may suggests to the establishment of laser irradiation on spinal cord for skeletal muscle injury.
