2LP-11 Nonviral genome editing CRISPR nanocomplexes for targeting bacterial infections

Gene therapeutics for infectious diseases introduce a target-specific treatment strategy which can overcome problems of current broad spectrum antibiotics. In this study, we apply the CRISPR system, a pioneering tool for efficient genome editing, to genetically modulate molecular targets in bacterial infections via nonviral delivery. Recombinant Cas9 was derivatized with a cationic polymer and packaged with single guide RNA targeting the infection. We showed that polymer derivatized CRISPR complexes (= Cr-Nanocomplex) were successfully formed while maintaining endonuclease function. We also demonstrate that the Cr-Nanocomplex can be efficiently delivered into both bacteria and mammalian cells. Furthermore, the Cr-Nanocomplex showed higher genome editing efficiency compared to using native or lipid-based formulations. The present study shows a therapeutic strategy which can be potentially applied in the clinic for treating infections as well as various other diseases.

많은 통계조사에서 사용되고 있는 산업/직업분류코드가 기존에는 사람에 의해 수동으로 부호화되어 왔는데, 이러한 작업은 시간과 인력면에서 고비용을 요구할 뿐 아니라 개인별 시각, 이해도의 차이 등으로 정확성에 많은 문제가 제기되어 왔다 본 논문에서는 이러한 수동코딩 작업의 문제점을 해결하기 위하여 자동코딩 시스템을 개발, 이를 인구주택총조사와 사업체기초통계조사에 시험 적용하여 본 바를 바탕으로 향후 자동시스템으로의 전환 방향 등을 제시하고 있다. Korean standard industrial/occupational classification has been the basis of producing accurate statistical data related with our industrial structure and distribution of industry and occupation since 1960. But coding over several million records not only requires high cost in the aspects of time and manpower but also has many problems in accuracy and consistency. Therefore, we got to develop the automatic coding system in order to work out these problems of manual coding. This paper shows the structure of our system and the result of experiment over survey data of 2,000 Census.

한국 전통음악의 아속(雅俗)미학 패러다임 변화 연구 - 조선시대 거문고[玄琴]의 아속적 심미를 중심으로 -

본 연구는 아악과 속악의 개념과 범주를 어떻게 규정할 것인가는 논외로 하고, 조선 전기에서부터 후기로 이어 지는 과정에서 궁중음악과 풍류방 중심의 민간음악에 일관되게 투영되어 있는 전통 음악의 아속적 심미 체계에 대해 구체적 사료를 통해 밝혀보았다. 본 연구를 통해, 조선전기 아속과 관련된 음악의 심미는 주로 궁중의 제례악과 연향 악에 투영되었고, 특히 거문고[玄琴]의 심미적 사유가 그 중심에 있었다. 조선 후기로 내려오면서 아속과 관련된 음악 의 심미는 민간으로 확장되어 풍류방 음악에 투영되어 있으며, 거문고[玄琴]의 심미적 사유가 그 중심에 있었다는 것 을 살펴볼 수 있다. This study excludes how to define the concepts and categories of the Ah-ak(雅樂) and Sok-ak(俗樂). In the process from the early Joseon Dynasty to the late Joseon Dynasty, the aesthetic appreciation system of the Ah-Sok(雅俗) in traditional music, which is consistently projected on court music and folk music centered on Pungryubang, was revealed through specific historical records. Through this study, we were able to examine as follow : In the early Joseon Dynasty, the aesthetic of music related to the the Ah-Sok(雅俗) was mainly projected on the court's ritual music and Yeonhyangak-music for korean court banquet-, and in particular, the aesthetic thoughts of Geomungo[玄琴] was at the center. As we descend to the late Joseon Dynasty, the aesthetic of music related to the Ah-Sok(雅俗) has been expanded to the private sector, reflecting the aesthetic of Pungryubang music, and it can be seen that the aesthetic thought of Geomungo[玄琴] was at the center.

본 논문의 연구 목적은 거문고가 지닌 음악 내ㆍ외면적인 위상을 구체화하고 체계화하여 거문고에 대한 인식 및 음악 본연의 목적과 정체적 특징을 살피는 것이다. 거문고는 시대와 음악의 담론이 되어왔던 악기이며 오늘날 실기위주로 치중되어가는 음악의 일면에서 오늘날의 음악에 대한 음악내ㆍ외의 적절한 균형을 제시할 수 있는 주목해야 할 악기이다. 그리고 선비가 벗으로 삼거나, 향유의 대상으로만 인식했던 것 뿐 아니라, 인재양성과 제자들이 인격함양을 위한 공부에 있어서도 거문고는 매우 훌륭한 교육자재로 활용되었으며 이러한 거문고의 정체성은 시대가 바뀌어도 변할 수 없는 것이다. 그것은 시대의 흐름과 함께 역사 속에서 면면히 이어져야 하는 것이며, 거문고는 음악현장에서 악기로서의 의미뿐 아니라, 전 사회에 걸쳐 인격완성을 위해 소통하고 함께 해야 할 우리의 향악기인 것이다.

Cas9 conjugate complex delivering donor DNA for efficient gene editing by homology-directed repair

Delivery of the CRISPR ribonucleoprotein (RNP) for homology-directed repair (HDR) has been challengingdue to the low efficiency. Herein, we developed a Cas9 conjugate complex system which can induce efficientHDR editing with the use of a minimal amount of carrier material. Cas9 from Streptococcus pyogeneswas purified and conjugated with low molecular weight polymer (LP). The Cas9-LP conjugates were complexedwith single guide RNA (sgRNA) and donor DNA, which showed greatly enhanced internalizationinto cells compared to native Cas9 complexes, as well as a high extent of co-localization of Cas9 withsgRNA. The cytotoxicity of Cas9-LP complexes was evaluated, demonstrating low cytotoxicity comparedto the conventional lipofectamine formulation. Finally, the treatment of Cas9-LP complexes to HEK293Treporter cell line expressing a mutant red fluorescent protein (RFP) results in efficient base correction ofthe RFP gene (up to 31%), leading to restoration of RFP expression and fluorescence. We anticipate thatthe current method can be widely used as a platform for efficient HDR editing via ‘minimal carrierassisted’delivery without the aid of any external physical stimuli, which can be potentially applied forin vivo and ex vivo editing of cellular targets.

일본의 ‘후루사토(故鄕)’ 관광에서 나타난 농촌의 이미지에 대한 고찰

観光で現れるイメージは、観光客に重要な要所だということができる。イメージというのは、一般的に心の中で描かれる事物の感覚的な形象(絵)、あるいは心象ということができる。観光の素材として、故郷というイメージは、物質的な側面より精神的な側面が強く現れることである。過去の観光というのは、見て来るだけに限定されて、現在は帰ってからその以後までも影響を及ぼしている。故郷のもつイメージは、いつも新鮮で温かくて身近なイメージをもたらしている。 この論文は、観光の形態のふるさと観光を通して、農村のイメージを検討しようとした。このような研究のため、日本の農村観光と地域活性化の方案に関する既存の論文を検討してみて、ふるさと観光で現れる特徴で、ふるさとについてのノスタルジアの側面と農村で見られる自然環境や伝統文化、住民の積極的な参加に関して論じた。そして、ふるさと観光の事例地域で代表的な岩手県遠野市をあげて、昔話の伝承とボランティア活動、遠野ふるさと村の生活体験、伝統文化の商品化について考察した.
