Genetic Polymorphisms of t-PA and PAI-1 Genes in the Korean Population

Abnormalities in fibrinolysis system is associated with risk of hypertension. In this report, the Alu repeat insertion / deletion (I / D) polymorphism of tissue plasminogen activator (t-PA) and the Hind III RFLP of plasminogen activator inhibitor-1 (PAI-1) genes were investigated in 115 normotensives and 83 patients with hypertension, and their association with anthropometrical data and plasma biochemical parameters were analyzed. There were no significant differences in the gene frequencies of the two candidate genes between normotensives and hypertensives, respectively. Our results indicate lack of associations between the two polymorphisms in t-PA and PAI-1 genes and risk of hypertension in the population under study. However, the Hind III RFLP of PAI-1 gene was significantly associated with plasma glucose level, suggesting its role in glucose metabolism. It needs to be tested whether this RFLP of PAI-1 gene is associated with insulin resistance syndrome or non-insulin dependent diabetes mellitus (NIDDM) in the Korean population.

본 연구에서는 빈경하중과 축회전 속도를 변화시켜 가며 하중으로 인한 베어링의 부하-변위량 특성을 Harris 이론과 비교분석하였다. 실험결과 결함이 없는 경우 하중이 적을 때는 Harris이론과 비교적 잘 일치하나, 하중이 증가할 수록 구동축의 변곡, 볼베어링 하중의 불균형 등의 영향을 받아 이론값과 차이를 보였다. 반경방향 하중이 있는 단일결함과 다중결함의 경우 하중이 증가함에 따라 변위량의 변화는 축심변위량 보다 반경변위량이 더욱 커지게 된다는 것을 알았다. 베어링의 결함상태에 따라 하중과 변위량의 기울기인 부하-변위량 특성을 구함으로써 볼베어링의 결함유무를 검출하였다. In this study, comparison between Harris-theoretical values and experimental data of load-deflection characteristics in bearing was made. The experiments are conducted under the conditions of the various radial loads and speed of shaft. In the case of non-defective ball bearing, the experimental data agreed well with the Harris-theoretical values for the small steady radial load but not for the large steady radial load. For the radial load bearing, the experimental results show that the stiffness of bearing at the single and multiple defective bearing are bigger in the radial defectiion than in the axial deflection. Load-deflection characteristics for the bearing defect part make it possible to detect the presence of a defect in bearing.
Linkang Disequilibrium of Dopamine D2 Receptor Gene in the Korean Population

The genetic basis of hypertension is complex, and has been considered to be associated with the dopamine D2 receptor gene (DD2R). Because association studies using the candidate gene approach may provide important clues regarding the pathogenesis of hypertension and establish basis for further study, we performed the association study on the relationship between genetic polymorphisms in the DD2R gene and hypertension in Koreans. Eighty nine patients with hypertension and 86 age-matched subjects with normal blood pressure were enrolled. Genomic DNA was extracted from peripheral blood leukocytes. PCR-RFLP analysis was performed to detect the three polymorphic Taq I sites in the DD2R gene. There were no significant differences in genotype, allele and haplotype distributions of any polymorphisms in the DD2R gene between two groups, respectively (P>0.05), although significant linkage disequilibriums among these polymorphic sites were detected by pair-wise analysis (P<0.05). Therefore, our negative result suggest that the three Taq I RFLPs in the DD2R gene were not significantly associated with hypertension in Koreans.
Genetic Analysis of Kallikrein-Kinin System in the Korean Hypertensives

The kallikrein-kinin system affects regulation of blood pressure, and genes encoding for the components of this system have been considered as good candidates for hypertension. To evaluate the relationship between genetic polymorphisms of candidate genes involved in this system and hypertension, we performed case-control studies using genetic markers in Korean normotensives and hypertensives, respectively. By association study, there was a marginal association with hypertension in AA genotype distribution of A1789G polymorphism in the hKLK1 gene (P = 0.0754). Thus, this genetic polymorphism may weakly contribute to the susceptibility to hypertension in Koreans. We also observed that significant linkage disequilibrium exists among three polymorphic sites in the hKLK1 gene studied, suggesting that the three genetic polymorphisms can be useful as genetic markers in clinical association studies. Further studies using larger sample sizes and more genetic markers will be needed to clarify genetic influence of kallikrein-kinin system for hypertension.
식사대용 식품(Ready-to-eat meals) 중 병원성 세균의 분포와 항생제 감수성 양상

본 연구에서는 2005년 11월부터 2006년 3월 초까지 497개의 초밥, 김밥, 샌드위치 등의 식사대용 식품 검체를 수거하여 식품공전을 토대로 식사대용 식품에서 유래되는 미생물학적 인 오염도를 조사하고, 현대 사회에서 항생제 사용의 오남용으로 항생제 내성이 심각한 상황을고려하여 분리된 균에 대한 항생제 감수성 양상에 대하여 알아보고자 하였다. 수거된 검체를 실험한 결과 김밥에서 4개(1.4%)의 Escherichia coli가 분리되었고, 초밥, 샌드위치 등에서는 분리되지 앓았다. 또한 김밥에서 12개(4.4%), 초밥에서 8개(5.4%), 샌드위치에서 2개(4.3%)로 총22개(4.4%)의 Staphylococcus aureus가 분리되었다. 식사대용 식품에서 분리된 26개의 균주로 National Committee for Clinical Laboratory Standards (NCCLS)의 기준에 의거하여 vancomycin, teicoplanin, gentamycin, cipro-floxacin, synercid, cefotaxime, lincomycin, meropenem, cefazolin, amoxicillin-clavulanic acid으로 총 10개의 항생제를 사용하여 E. coli와 S. aureus의 항생제 감수성 양상을 파악해 보고자 하였다. 실험 결과 식사대용 식품에서 분리된 E. coli 4균주의 항생제 감수성 양상은 그람 음성균 치료에 주로 사용되는 항생제인 gentamycin, ciprofloxacin, cefotaxine, meropenem, cefazolin에 대해서는 감수성이 높다고 판단되었으나 amoxicillin-clavulanic acid에 대해서는 내성을 나타내었고, S. aureus의 항생제 감수성은 비교적 높다고 판단되었다. This study was performed in order to measure the level of food-borne pathogenic bacteria and antibiotic resistance pattern of found ready to eat meals such as Him-bap, Cho-bap, Hamburger, Sandwich and packed lunch boxes. A total of 497 samples were collected from supermarket and department of Seoul, Kyung-ki, Inctleon, Kang-won, Chung-Cheong from November, 2005 to March, 2006. The contaminated microorganisms were in most cases tract relative strain like E. coli and S. aureus. Result have shown E. coli was detected 4 strains and S. aureus was detected 22 strains. 26 strains were also tested the antibiotic resistance pattern. 26 strains were shown to be relatively susceptible to synercid, vancomycin, teicoplanin, ciprofloxacin, gentamycin, lincomycn, cefotaxime, meropenem, cephalosporin, amoxicillin-clavulanic acid by the MIC dilution method, but E. coli 1 strain was resistant to amoxicillin-clavulanic acid.
9주 동안에 걸친 함초섭취 및 복합운동이 비만 여대생에 대한 신체조성, 혈중 생화학적 변인 및 피부미용에 미치는 효과

This study tried to investigate the effects of jointed grasswort(Salicornia herbacea L.) consumption and combined exercise during 9 weeks on the body composition, blood biochemical parameters and skin function in women of 20 years. A total of 40 female subjects were participated in this study, and they were randomly divided into 4 gropus including placebo group(n=10), jointed grasswort group(n=10), placebo + exercise group(n=10) and jointed grasswort + exercise group(n=10), respectively. In the case of the body composition, jointed grasswort + exercise group indicated higher increase in fat-free mass, muscle mass and skeletal muscle mass than other groups after experiment during 9 weeks(p<.05). However, jointed grasswort consumption and/or combined exercise program during 9 weeks did not show any beneficial effects for blood lipid profiles or C-reactive protein(CRP) level. With respect to skin function, only jointed grasswort consumption show significanly higher skin moisturing effect than other groups(P<.05). In summary, data from our study suggest that although jointed grasswort consumption with combined exercise may be useful as the body composition of participants, it likely that only grasswort consumption are beneficial to attain the skin moisturing effect. Further studies using larger sample size and longer study period will be required.

Blood pressure, obesity and bone mineral density are multifactorial traits influenced by genetic and environmental factors. The purpose of this study was to clarify the associations among three genetic polymorphisms in the ER-α gene and several clinical phenotypes in college athletes and major students in physical education. Seventy two healthy young men including 34 athletes and 38 controls were recruited in the sudy. Pheripheral blood samples were collected from each individuals and total genomic DNA were isolated from buffy coat Experiments were performed by anthropometric and genetic nalyses. Statistical analysis were carried out using unpaired t-test or one-way ANOVA test for quantitative data such as anthropometrical and biochemical parameters, and x2-test for categorical data such as genotype and allele distributions. The results were produced as follows. Firstly, for PouII RFLP in 5-UTR of ER-α gene, this site was monomorphic in our subjects, and further analysis could not performed. Secondly, in the case of PoaII RFLP in the intron 1 of this gene, this polymorphism was significantly associated with SBP and DBP values in controls (p<.05). Therefore, this polymorphism might be useful as a genetic marker for inter-individual susceptibility in blood pressure regulation. Lastly, XbaI RFLP in the intron 1 of this gene was significantly associated with SBP, DBP, height, weight, BMR and spinal BMC values in young men (p<.05). Therefore, it is likely that this polymorphism is useful as a predictive marker for inter-individual susceptibility to several clinical phenotypes such as blood pressure, obesity and bone mineral density in korean college athletes and major students in physical education.
