누룩으로부터 ${\beta}-galactosidase$ 활성을 가지는 균주를 순수분리하여 분리된 균주(NY-15)의 ${\beta}-galactosidase$를 정제하였다. 다량배양하여 모은 균체를 8%(v/v) toluene 처리에 의한 추출액을 정제의 출발 물질로 하여 acetone 침전에 의해서 탈지처리를 한 후 40-60%포화의 ammonium sulfate 분획을 하고 DEAE-cellulose, DEAE-Sephadex A-50의 이온교환 chromatography를 한 후 Agarose-PAPT를 이용한 affinity chromatography에 의해서 정제하였다. 이 정제법에 의해서 ${\beta}-galactosidase$는 조효소 추출액으로부터 40배 정제되었으며 회수율은 7.7%였나. 누룩 yeast의 ${\beta}-galactosidase$는 2개의 subunit로 되어 있으며, 각 subunit 분자량은 각각 96,000과 130,000이었다. 효소활성에의 최적 pH는 7.5이었고, 최적온도는 $40^{\circ}C$이었다. A strain of Nuruk yeast No. IS (NY-15) which produced high activity of ${\beta}-galactosidase$ was isolated from Nuruk, and the crude enzyme was prepared by whey permeate culture of the microorganism. The crude enzyme was purified 40-fold with a 7.7% yield by acetone and ammonium sulfate fractionational precipitation, and chromatography on DEAE-cellulose, DEAE-Sephadex A-50 and Agarose-PAPT. Purified ${\beta}-galactosidase$ from Nuruk yeast showed two types of subunit patterns; a slow moving band and a fast moving deeply stained band, both anode·migrating at pH 7.5. The molecular weight of the former was estimated to be about 130,000 and that of the latter was 96,000 by SDS-polyacrylamide gel electrophoresis. The optimum pH of the enzyme activity was 7.5 and maximum activity appeared at $40^{\circ}C$.
감정노동이 직무소진을 매개로 반생산적과업행동에 미치는 영향에 관한 연구

Recently the importance of emotional labor has increased so this study aims to prove the effects of emotional labor(surface acting and deep acting) on counterproductive work behaviors(deviate from organization and individual deviation) using burnout(emotional exhaustion, personality disorder, decrease in sense of accomplishment) as a medium. 350 surveys were sent to employees such as workers from call centers, salesperson from department store, financial worker, hospital staff, flight attendant and workers from customer center who and worked with emotional labor and 300 surveys excluding insincere ones were used for analysis. The result is as follows. First, surface acting which is a type of emotional labor chosen by customer workers had a positive effect on burnout such as personality disorder and decrease in sense of accomplishment. Second, deep acting had a negative effect on personality disorder and decrease in sense of accomplishment but did not show any effect on emotional exhaustion. Third, personality disorder and decrease in sense of accomplishment among burnout had effect on counterproductive work behaviors(deviate from organization, individual deviation) but emotional exhaustion did not show any effect on work behaviors(deviate from organization, individual deviation). Based on the result of analysis, implication of this study and direction of future research were suggested.

Rice seeds of 4 cultivars including Whachung-giant embryonic rice and Nampung-giant embryonic rice, as a group of the non-waxy rice cultivars, and Shinsunchal-giant embryonic rice and Whachungchal-giant embryonic rice, as that of the waxy rice cultivars, were germinated at $27^{\circ}C$ for 3 days to compare the changes in some physicochemical properties of the starch granules and the starch-hydrolysing enzyme activities during germination, respectively. ${\alpha}-Amylase$ activity of rices germinated for 3 days found to be higher than that of malt. Especially, Whachung-giant embryonic rice and Shinsunchal-giant embryonic rice were greater in activity than other rice cultivars and possessed the activities double that of malt. In contrast, ${\beta}-amylase$ of germinated rice found to be considerably less active than malt, although the giant embryonic rice group showed prevalent activity as compared o the normal rice group. With the starch granules, the amount of long glucose chains from amylose molecules were reduced in the non-waxy type giant embryonic rices, while the chain length increase was found in the waxy type giant embryonic rices. For the distribution profile of the glucose chain length from amylopectin molecules, we could observed that the chain length with DP (degree of polymerization) ranged 33 to 66 and 14 to 32 increased with the decreasing rate of that above 67 and below 13 regardless of starch waxiness. With non-waxy type of giant embryonic rices, susceptibility for glucoamylase were found to reduce along with germination, however, increase in susceptibility was observed with waxy rice types. In addition, we found the reduction in both initiation and termination temperature, and enthalpy for gelatinization.

Seventy percent ethanolic extracts from commercially available 13 legumes were made to investigated their antioxidative activities by determining the reducing power, inhibitory effect on lipid peroxidation, scavenging activity against both superoxide radical and hydroxyl radical, together with inhibitory activity toward mitomycin C-induced oxidative damage of DNA. High level of reducing powers were detected in Yepat, Sokpiri, Yuweol-bean and Jeokdu. Inhibitory effects on lipid peroxidation were found ubiquitously in all extracts examined when employing the linoleic acid autoxidation system, whereas, only 3 legumes, Yepat, Namul-bean and Jeenuni-bean, were revealed marked inhibition in rabbit erythrocyte-ghost membrane lipid peroxidation system. Yepat, Namul-bean, Jeokdu and Jeenuni-bean showed great scavenging activities on superoxide radical, on the other hand, high level of hydroxyl radical scavenging activities were demonstrated in Sokpiri, Chungtae, Yepat and Jebi-bean. Ubiquitous inhibitory effects on mitomycin C-induced oxidative damage on DNA were found in all extracts tested, Among them, however, Yepat, Jeenuni-bean, Namul-bean, Nokdu and Jeokdu showed the higher level of inhibition. Taken together, we could assign Yepat, Jeokdu, Jeenuni-bean and Sokpiri, for the legume species highly functional on overall antioxidative activity.

In the conventional video file system, videos are stored in a high performance server which has mass storage hard disks or disk arrays. For 3D internet broadcasting, real time operations are required to transmit video files to many clients. This paper describes the design of the 3D internet broadcasting system which can provide realtime streaming service to many users in the 5G environment. In reality, unicast is used to transmit multimedia contents over the internet rather than IP multicast since IP multicast has its own drawbacks in deployment, security, maintenance and so on. In addition, multimedia broadcasting service system like VoD has difficulties in applying to 3D internet broadcasting system since it requires a large amount of system and network resources. In this work, we develop a 3D internet broadcasting system which can construct effective data delivery by minimizing performance-degrading factors.

Starches from 4 rice cultivars, including Whachungchalbyeo and Whachungbyeo, together with two correspondiug conversion mutants with giant embryonic phenotype, Whachungchal-giant embryonic rice and Whachung-giant embryonic rice, were isolated to compare its physico-chemical properties. The amylose content and the length of glucose chains in the fractionated amylose was reduced in response to conversion of the rice cultivars for developing giant embryonic phenotype. For the fine structure of amylopectin, conversion to giant embyonic rice cultivars accompanied with slight increase in the length of B chain plus decrease in the amount of A chain, and also a slight increase in average glucose chains length of amylopectin fractions could be observed. X-ray diffractogram revealed 4 cultivars to be the representative A types. We could also obtain the results that the hydrolysis rate by glucoamylase in the waxy rice cultivars decreased in response to conversion to the giant embryonic rice cultivars, while the rate increased in the non-waxy rice cultivas. Termination temperature for gelatinization was observed to decrease in response to conversion into the giant embryonic rice cultivars, however, the enthalpy for gelatinization was found to increase with the same conversion process.

The experiments of rice bread processing were carried out to compare the varietal difference in processing adaptability to rice bread between brown and milled rice flour, and to analyze the interrelations among chemical properties of rice grain and some characteristics relevant to rice bread processing and quality. There was varietal difference in adaptability to rice bread processing in both brown and milled rice, but there was not significant adaptability differrence between brown and milled rice flours to rice bread processing. However, there was remarkable adaptability difference between brown and milled rice flours to rice bread processing in some rice varieties. Three high-amylose rices AC 27, IR 44, Suweonjo showed high quality of milled rice bread among tested rice materials. Brown rice revealed better adaptability to rice bread processing compared with milled rice in all varieties except the above three varieties. Especially, the glutinous rice Hangangchalbyeo failed to normal formation of rice bread from milled rice flour, but it showed the successful formation of rice bread from brown rice flour. The interrelations among chemical components of rice grain and some charateristics relevant to rice bread processing and quality exhibited quite different tendency between brown and milled rices. In the case of rice bread processing by brown rice flour, the larger volume expansion of dough during fermentation made the more springy rice bread and the more moist rice bread showed the more soft and cohesive physical property. In the case of rice bread processing by milled rice flour, the lower protein of rice flour was closely associated with the more moistness of rice bread and the higher lipid led to the more uniformal air pore distribution, the smaller pore size and the lower springiness of rice bread. Also, the larger volume expansion of dough during fermentation made the better loaf formation and the larger pore size of rice bread. The better loaf formation of rice bread revealed the softer hardness and the lower chewiness, and the lower springiness was closely correlated with the more uniformal size distribution of air pore and the smaller pore size in rice bread.
에너지 빅데이터를 활용한 머신러닝 기반의 생산 예측 모형 연구

전력망의 역할은 안정적인 전력공급이 최우선이다. 예고 없는 불안정한 상황에 대한 여러 가지 대비에 대한 방안이 필요하다. 기상 데이터를 활용하여 탐구적 데이터 분석을 통한 피처 간의 관계를 파악하여 머신러닝 기반의 에너지 생산 예측 모형을 모델링한다. 본 연구에서는 주성분분석을 사용하여 에너지 생산 예측 시 영향을 미치는 피처를 추출하였으며 머신러닝 모델에 적용함으로써 예측 신뢰도를 높였다. 제안한 모형을 사용하여 특정 기간을 대상으로 생산 에너지를 예측하고 해당 시점의 실제 생산 값과 비교함으로써 주성분분석을 적용한 에너지 생산 예측에 대한 성능을 확인하였다. The role of the power grid is to ensure stable power supply. It is necessary to take various measures to prepare for unstable situations without notice. After identifying the relationship between features through exploratory data analysis using weather data, a machine learning based energy production prediction model is modeled. In this study, the prediction reliability was increased by extracting the features that affect energy production prediction using principal component analysis and then applying it to the machine learning model. By using the proposed model to predict the production energy for a specific period and compare it with the actual production value at that time, the performance of the energy production prediction applying the principal component analysis was confirmed.

본 연구의 목적은 유치원과 어린이집 중간관리자의 직무역량에 대한 인식 및 교육요구의 우선순위를 비교·분석하는 것이며, 이를 통해 유아교육기관 중간관리자의 역량강화를 위한 교육프로그램을 개발하는데 기초자료를 제공하는 것이다. 이를 위하여 부산ㆍ경남지역에 소재한 유치원과 어린이집의 중간관리자 218명을 대상으로 2016년 1월 13일부터 2월 5일까지 설문조사를 실시하였고, 직무역량에 대한 교육요구가 높은 최우선순위 직무역량을 도출하고자 대응표본 t-검증과 Borich 요구도를 분석하였다. 연구결과는 첫째, 유치원과 어린이집 중간관리자가 인식하는 현재 직무역량의 수행수준은 5점 척도에서 3.77, 3.74로 각 각 나타났다. 둘째, 유치원과 어린이집 중간관리자의 직무역량에 대한 교육요구를 분석한 결과, 공통적으로 유치원과 어린이집 중간관리자의 교육요구도가 높은 직무역량은 10개로 도출되었다. 이 중 교육요구도가 가장 높은 직무역량은 ‘특수아에 대한 발달특성 이해’와 ‘새로운 정보화 기술 이해’였고, ‘교직 인성 및 전문성 발달’ 범주의 역량내용이 가장 많았다. 이러한 결과를 기초로 유아교육기관 중간관리자의 직무역량을 강화하기 위한 교사교육 및 연수의 방안에 대하여 논의하였다. The purpose of this research study is to conduct a comparative analysis of the awareness of job competences and the educational needs priority of middle managers in kindergartens and childcare centers according to the types of institutions. Through this, this study is to offer preliminary data necessary for developing educational programs designed to strengthen the capabilities of middle managers in early childhood education institutions. To achieve the purpose, this research study conducted a survey of 218 middle managers in kindergartens and childcare centers. A paired t-test and the Borich Needs Assessment Model(1980) were carried out to present the priority of the job competencies with high educational needs. The analytic research findings are as follows. First, it is indicated that the current level of job competences awareness in middle managers of kindergartens and chidcare centers has been measured on a 5-point scale, 3,77 and 3.74, a general level, respectively in turn. Second, the analytic research about the educational needs of middle managers in kindergartens and childcare centers shows that 10 job competencies with high educational needs of middle managers in kindergartens and childcare centers have been put forward. For the job competencies with the highest need for education, Understanding the Developmental Characteristics of Children with Special Needs and Understanding New Information Technologies have commonly accounted for the highest rate. The category contents of the area of the development of teachers character and professionalism have assumed the greatest weight. Based on the research findings, this research study discussed the methods about education and training of teachers to strengthen job competences of middle managers in early education institutions.
