        바실러스 발효대사물이 함유된 신규 칼슘보충용식품의 성장기 쥐 장골 성장촉진효과

        기존 칼슘보충용식품의 성장촉진효능을 증진시키기 위하여 칼슘흡수 관여 물질인 γ-PGA를 다량 생산하는 Bacillus subtilis SE4 균주를 선발하였다. 선발된 B. subtilis SE4의 발효대사물을 기존 칼슘보충용식품에 첨가하여 성장기 수컷 흰쥐에 28일간 경구투여 한 결과 발효대사물이 첨가된 신규칼슘보충용식품군에서 혈청 내 IGF-Ⅰ 함량이 대조군이나 기존칼슘보충용식품군에 비해 유의적으로 높게 나타났다(p<0.05). 일일증체량, 일일사료섭취량, 장골길이, 장골 무게, 장골폭 등의 성장관련 지표에서도 장골의 길이와 일부 부위별 두께에서의 유의적 결과를 포함하여 전반적으로 신규칼슘보충용식품군이 대조군과 기존칼슘보충용식품군에 비해 양호한 성적을 나타내었다. 이러한 결과는 기존칼슘보충용식품에 발효대사물을 첨가함으로써 나타난 칼슘의 생체흡수율 증대 및 성장호르몬의 분비활성화 등 여러 성장관련요인들이 흰쥐 성장에 긍정적으로 작용한 때문으로 생각되어진다. This study was conducted to investigate the effect of administration of calcium supplementary food containing fermented product of Bacillus subtilis SE4 highly producing poly-γ-glutamic acid on the growthparameters of adolescent male rats. Four-week old male Sprague-Dawley (SD) rats were fed for 4 weeks and assigned to the following 4 groups: two groups administered orally with new calcium supplementary food (such as 150 ㎎/㎏ and 300 ㎎/㎏) containing fermented product of B. subtilis SE4, one group administered with conventional calcium supplementary food product (150 ㎎/㎏) and one saline group as control. Daily weight gain and daily food intake in the two new food product groups were higher than those of conventional food product group and control group. Especially, the content of serum IGF-Ⅰ in the two new food product groups were significantly higher than those in conventional food product group and in control group (p<0.05). In addition, length and weight of longitudinal bone in the two new food product groups were longer and heavier than those of conventional food product group and control group. Therefore, the addition of fermented food product of B. subtilis SE4 into the conventional calcium supplementary food increased all the parameters examined for the growth of the adolescent male rats.

      • 지하철역사 공간특성이 성범죄발생에 미치는 영향분석

        본 연구는 지하철역사에서의 범죄예방환경 구축을 위한 기초자료를 제공하기 위한 목적으로 범죄통계 분석, 전문가 면담, 실태조사, 그리고 컴퓨터 시뮬레이션 등의 방법을 활용하여 진행되었다. 실태조사는 역사의 주변환경은 유사하지만 범죄발생률에서만 차이를 보이는 6개의 비환승역을 대상으로 진행하였다. 또한 역사에서 승객이 승강장으로부터 개찰구까지 어떻게 이동하는지를 모사하고 정체와 혼잡이 발생하는 지점을 규명하기 위해 시뮬렉스 프로그램이 이용되었다. 분석결과, 높은 범죄발생률을 보이는 역사는 역사 내에서 잦은 정체와 혼잡이 일어나는 것을 확인할 수 있었다. 따라서 어떤 건축적 요소가 역사 내에서 승객의 정체와 혼잡을 야기시키는지에 대해 분석하였고 이러한 분석을 기반으로 지하철역사에서 범죄예방을 위한 건축디자인 가이드라인이 제시되었다. The purpose of this study was to deduce implications for the improvements in subway station for crime prevention based on the comprehensive analysis of the crime data, expert interview, field observation, and computer simulation. For field observation, 6 non-transfer stations which have similar circumstance characteristics and where there is a difference only in crime rate were selected and investigated. And for simulating how passengers move from platform to ticket gate in these stations, the Simulex was used. The results showed that the higher crime rate of the station is correlated with the place where the congestion and crowding frequently occurs. In conclusion, the architectural design elements of subway station which cause the passenger congestion were summarized and measures of architectural design for crime prevention in subway station were suggested.

        지하철역사 내 성폭력범죄 발생에 영향을 주는 건축계획특성 연구

        The purpose of this study was to deduce implications for the improvement in subway station for crime prevention based on the comprehensive analysis of the results of crime data analysis, expert interview, and computer simulation. To deduce characteristics of architectural design affecting the risk of sex offenses, passenger data, architectural documents, and crime data of seventeen non-transfer stations which have similar circumstance characteristics were investigated. And for simulating how passengers move from platform to ticket gate in stations, the Simulex was used. The results showed that the higher sexual crime rate of the station is correlated with the place where the congestion and crowding occurs. In conclusion, the architectural design elements of subway station which cause the passenger congestion were summarized and measures of architectural design for crime prevention in subway station were suggested.

        다중이용시설의 폭발물 테러위험도 평가를 위한 건축계획요소 체계화 연구

        As a preliminary step for developing vulnerability assessment model of terrorism, this study aims to deduce and classify architectural design elements of multi-use buildings to protect them from terrorism using explosives. For these objectives, eleven domestic and foreign guidelines of anti-terrorism, including RVS which is one of the commonly used tools for assessing vulnerability to terrorism, were analyzed. As results, 2 scenarios of explosive attack, 4 layers of defense, and 58 architectural design elements for risk assessment of terrorism were deduced. And the design elements were categorized into 18 groups based on their purpose and function to take into account the supplementary effects among them. Then, the design measures applicable for each element were classified into several grades on the basis of its protection or risk level. Lastly, 11 multi-use buildings were selected and investigated how the elements suggested in this study were applied to them.

        국내 고층 건축물의 차량폭발물테러 위험도 분석 연구

        The purpose of this study is to assess vulnerability of vehicle bomb attack of high rise buildings in Korea. For this objective, 4 layers of defense and 17 architectural design elements were deduced on the basis of domestic and foreign anti-terrorism guidelines. Weights of 4 layers and 17 elements were developed through the AHP survey of anti-terrorism specialists. The potential vulnerability of bomb attack of 10 high rise offices in Seoul were assessed based on the model. The result indicates that there were no significant difference of overall vulnerability among all buildings and the vulnerability level was found to be medium. Among 4 layers of defense, the vulnerability of 2nd layer was the highest and the vulnerability of 3rd layer showed more variation among the buildings assessed. In conclusion, several cost-effective architectural design options to mitigate potential vehicle bomb attack were suggested.

        노인을 고려한 아파트 욕실의 색채계획에 관한 연구

        The purpose of this study is to suggest the color scheme of bathroom which is more easily discernable for the elderly while it maintains the similar mood. Criteria for analysis of color scheme was developed based on the results of existing literature on the color perception of the elderly, and the discernabilities and image scales of color schemes of bathroom in seventeen model apartment houses were analyzed using the criteria. The most frequently used colors in the bathroom were the hues of Y, YR, high value and low chroma with Vp, Lgr tone and white, and these color schemes were considered to be difficult to discern for the elderly who is suffering yellowed eye sight. Therefore, this study suggests the color scheme which is more easily discernable for the elderly who is suffering yellowed eye sight while it maintains the similar color image scale.

      • 상업지역의 환경적 특성에 따른 범죄위험도 분석

        본 연구는 상업지역을 대상으로 범죄발생에 영향을 주는 인구사회, 경제, 토지지용과 관련된 내용뿐만 아니라 도시 및 건축 환경과 관련된 내용을 포함하는 평가지표를 도출하고, 행정동 단위로 환경적 특성에 따른 범죄위험도를 분석하여 범죄예방을 위한 정책적 함의를 도출하는 것을 목적으로 하였다. 연구에 사용될 평가지표를 도출하기 위해 선행연구 및 기존 위험도 평가모델의 사례분석을 통해 범죄발생에 영향을 줄 수 있는 인구사회학적 특성 변인과 물리적 특성 변인을 도출하였다. 분석대상을 선정하기 위해 지역의 상업화비율과 범죄발생밀도의 분포를 고려하여 서울시 6개 자치구의 106개 행정동을 분석대상으로 선정하고, 인구사회학적 특성, 물리적 특성, 범죄발생 특성에 관한 평가지표 데이터를 수집하였다. 수집된 데이터를 기반으로 평가지표 간의 상관관계를 분석하여 범죄발생에 영향을 주는 요인을 총 4개로 요약, 추출하였으며, 주성분 회귀분석을 통해 범죄유형별 주요 영향요인 및 영향력을 추출하였다. This study aims to identify the indexes of evaluation including sociodemographic, economic, land-use characteristics as well as urban and architectural environment which affect the occurrence of crime in the commercial area, and to assess the crime risk of administrative dong according to their environmental characteristics. Finally, this study aims to deduce some implications for the crime prevention policy. To identify the indexes of evaluation, existing studies and risk assessment model were analyzed. 6 administrative ku and 106 administrative dong were selected for the analysis considering the proportion of commercial land use and the frequency of crime occurrence. Collected data from 106 administrative dong were analyzed to find the correlations among the indexes of evaluation. The physical and socio-demographic variables which affect the occurrence of crime were summarized into 4 major factors through principle component regression analysis.

        1-Deoxynojirimycin 생산 균주 Bucillus subtilis MORI 3K-85의 단백질 분석

        In our previous study, we isolated and characterized a 1-deoxynojirimycin (DNJ)-producing bacterium, Bacillus subtilis MORI, from chungkookjang, a Korean traditional food. B. subtilis MORI was subjected to γ-irradiation and the resulting bacteria were screened for increased DNJ production. A mutant was identified that produced 7.6 times more DNJ and named B. subtilis MORI 3K-85. In this study, the protein profiles of both strains were compared by one-dimensional and two-dimensional gel electrophoresis (1-DE and 2-DE, respectively) under both native and denaturing conditions. The 1-DE native-PAGE and 1-DE SDS-PAGE analyses identified 5 and 7 bands, respectively, that were found at higher concentrations in B. subtilisMORI 3K-85 than in B. subtilis MORI. Similarly, 2-DE analyses identified 20 protein spots which were found at higher concentrations in B. subtilis MORI 3K-85. The peptide mass profiles of these 20 proteins were analyzed by MALDI-TOF and compared with peptide sequences of B. subtilis and B. amyloliquefaciens in the MASCOT database. This screening suggested that three dehydrogenases, an aldolase, a synthetase, an isomerase, a reductase, and a peroxidase are elevated in B. subtilis MORI 3K-85. Based on this data, one or more of the elevated 8 enzymes might be related to the DNJ biosynthetic pathway.

        알파글루코시다아제 저해제 1-deoxynojirimycin을 생산하는 Bacillus 균주의 분리 및 동정

        토양으로부터 분리한 Streptomyces sp. 30여 균주들과 한국전통식품에서 분리한 Bacillus sp. 200여 균주들로부터 α-glucosidase 의 활성을 저해하고 동시에 DNJ를 생산하는 유용균주를 선발하였다. 실험결과 한국전통식품인 청국장으로부터 α-glucosidase 저해능이 높고 DNJ 생산능이 우수한 한개의 균주를 선발하였다. 이 균주의 동정을 위하여 API kit에 의한 균의 당 이용능 분석, HPLC와 GC에 의한 균체의 quinone 및 지방산 분석 등과 함께 균의 16S rDNA 염기서열을 분석하였으며 주사전자현미경에 의해 균주의 형태적 특성을 관찰하였다. 그 결과 본 연구를 통해 선발한 균주는 건강기능성식품 개발 등에 적용할 수 있는 GRAS에 속하는 균주임을 확인하여 B. subtilis MORI로 명명하였다. Thirty Streptomyces sp. and 200 Bacillus sp. isolated from Korean soils and traditional foods were screened for their abilities to inhibit α-glucosidase and produce 1-deoxynojirimycin (DNJ). This screening identified a Bacillus sp. bacterium that strongly inhibited α-glucosidase and produced high levels of DNJ from Chungkookjang, a Korean traditional food. The bacterium was characterized in terms of its biochemical and molecular biological properties such as sugar utilization, cellular quinone composition, cell wall fatty acid composition, and 16S rDNA sequence. In addition, scanning electron microscopy was used to visualize the morphology of the bacterium. These analyses identified the bacterium as B. subtilis, a bacterium with Generally Recognized as Safe (GRAS) status. The selected strain was named B. subtilis MORI.
