      • Petunia Hybrida 의 엽내조직으로부터 원형질체 유리 및 배양후 Callus 형성에 미치는 요인

        The study was conducted to improve protoplast isolation and fusion efficiency of mesophyll of Petunia hybrids $quot;Cascade red.$quot; The protoplast culture was also attempted to determine 1-naphtalene acetic acid concentration added to basal medium, 8p-Kao and Michayluk(8p-KM), for colony and callus formation. Preplasmolysed mesophyll of Petunia hybrids was well digested in enzyme solution containing 0.5% macerozyme R-10, 2.0% cellulase Onozuka R-10, 0.01M MES buffer, and 0.7M mannitol adjusted pH to 5.8, and protoplasts with debris were washed with 0.6M mannitol solution once and purified with 0.7M sucrose solution twice. When PEG solution had been eluted with eluting solution which was adjusted pH to 9.0-10.5 after 30% PEG solution treatment, fusion frequency was highly developed. Callus was formed in 8p-KM medium containing 1.0ppm 2,4-D, 0.5ppm BA and 1.0ppm NAA, and 0.1M mannitol which was added as osmoticum.

        유전자총을 이용한 상추 내로의 배추 Glutathione Reductase (GR)유전자의 도입

        상추에 particle bombardment를 이용한 배추 Glutathione Reductase (GR)유전자의 형질전환효율 향상 및 형질전환체의 유전분석에 관한 실험을 수행하여 얻은 결과를 요약하면 다음과 같다. Particle bombardment 이용시 사출 전 4시간, 사출 후 16시간 동안 삼투조절제인 0.6M serbitolfmannitol 을 처리하여 형질전환시킨 상추 자엽조직에서 GUS 반응 spot이 가장 많았다. 0.3, 0.6, 1.5 kbp로 나타나는 3가지 종류의 primer를 사용하여 PCR한 결과 각각의 primer에 homology를 타나내는 band를 확인하여 355 promoter와 BcGRl유전자가 삽입이 된 것을 확인하였다. 형질전환된 상추의 자식 종자를 채종하여 kanamycin 200mg/L이 첨가된 MS배지에 파종하여 형질전환시키지 않은 상추의 자식 종자와 비교해 본 결과, 형질전환된 상추의 종자에서 발아된 유묘의 약 70%정도가 kanamycin에 대해 내성을 나타내었다. The cytosolic glutathione reductase(GR) gene of chinese cabbage was introduced into Lettuce (Lactuca sativa L.) with particle bombardment method. When cotyledon explants were treated with osmoticum-conditioning medium (0.6 M sorbitol/mannitol) 4 hours prior to and 16 hours after bombardment, it was identified by GUS assay that this condition was the most efficient one for introduction of foreign genes into cotyledon tissue of lettuce with particle bombardment. The stable integration of a GR gene was confirmed by the PCR analysis. 0.3, 0.6, 1.5 kbp PCR fragments of transgenes were obtained by three types of primers designed on the basis of GR sequence. To know whether the expression of the GR gene of pBKs-GR 1 can be stably maintained in the next generation, T$_2$selfing seeds obtained from the transformed mother plants were sowed on MS medium supplemented with 200 mg/L kanamycin sulfate. 70% of seedlings showed resistance to kanamycin.

        등온가열 수평원관내 융해과정동안의 열적 불안정성

        The constrained melting inside an isothermally heated horizontal cylinder has been repeatedly investigated in many studies only for the moderate Rayleigh numbers. This study extends the range of Rayleigh numbers to systematically investigate the transition during melting processes, especially focusing on the complex multi-cellular flow pattern and thermal instability. The enthalpy-porosity formulation, with appropriate source terms to account for the phase change, is employed. For low Rayleigh numbers, initially developed single-cell base flow keeps the flow stable. For moderate Rayleigh numbers, even small disturbances in balance between thermal buoyance force and viscous force result in branched flow structure. For high Rayleight numbers, Benard type convection is found to develop within a narrow gap between thee wall and the unmelted solid. The marginal Rayleigh number and the corresponding wave number are in excellent agreement with those from linear stability theory.

      • Agrobacterium tumefaciens 를 이용한 Petunia hybrida 엽조직내로의 GUS (β-Glucronidase) 유전자의 도입 가능성

        Petuina kybrida was transformed with GUS gene by cocultivation of leaf disc with Agrobacterium tumefacines. Petunia leaf showed resistant response to the concentration of 100 ㎎/L kanamycin. From the petunia leaf dises(about 5×5 ㎜) cocultivated with Agrobacterium suspension cultures for 48 hours in MS medium containing 1.0 ㎎/L 2,4-D and 0.2 ㎎/L BAP, kanamycin resistant callus were more effectively induce. Shoots regenerated from the callus when cultured on MS medium containing 1.0 ㎎/L BAP, 0.1 ㎎/L NAA, 200 ㎎/L kanmycin, 500 ㎎/L carbenicillin, 30 g/L sucrose and 8 g/L agar. 59% of regenerants showed positive color response when reacted with X-glu.

        캡슐 형상이 축열에 미치는 영향

        A numerical investigation of the constrained melting of phase change materials within spherical-like capsule is presented. A single-domain enthalpy formulation is used for simulation of the phase change phenomenon. The solution methodology is verified with the melting process inside an isothermal spherical capsule. Especially, the effect of capsule shape on the heat storage is emphasized. Two shape parameters are considered from the real capsule shape showing good characteristics of heat storage and the effect of these parameters is examined. Early during the melting process, the conduction mode of heat transfer is dominant. Thus the capsule shape with large surface area is desirable. However, the capsule shape with large surface area plays negative role on the strength of buoyancy-driven convection that becomes more important as melting continues.

        성층화된 축열조의 1차원모델에 대한 적분 근사해

        paper deals with approximate integral solutions to the one-dimensional model describing the charging process of stratified thermal storage tanks. Temperature is assumed to be the form of Fermi-Dirac distribution function, which can be separated to two sets of cubic polynomials for each hot and cold side of thermal boundary layers. Proposed approximate integral solutions are compared to the previous works of the approximate analytic solutions and show reasonable agreement. The approach, however, has benefits in mathematical difficulties, complicated solution form and unstable convergence of series solution founded in the previous analytic solutions. Solutions for a semi-infinite region, which have simple closed form solutions, give close agreement to those for a finite region. Thermocline thickness is obtained in closed form and shows proportional behavior to the square root of time and inverse proportional behavior to the square root of flow rate.

        성층축열조의 성능에 대한 설계인자의 영향

        This study is to systematically analyze the effect of various kinds of design factors on the performance of stratified thermal storage tank. Taguchi method, known as a very reasonable tool in the parametric study, is employed in the present work. Three dimensional unsteady numerical experiment is conducted for 4 design parameters of stratified thermal storage tank: inlet Reynolds number, Froude number, diffuser size with 3 levels (Re=400, 800, 1200, Fr=0.5, 1.0, 2.0 and150mm, 200mm, 300mm) and diffuser shape with 2 levels. Orthogonal array L18(2×37) is adopted for the analysis of variance. The result gives quantitative estimation of the various design parameters affecting the performance and helps to select the main factors for the optimum design of stratified thermal storage tank. Reynolds number is found to be the most dominant parameter and the diffuser shape plays significant role on the performance of stratified thermal storage tank. Based on this finding, the prior questions on the contribution of the diffuser shape proposed by the authors become clear. The optimum condition for the performance is a set of300mm, Re=800, and radial regulated plate diffuser. Conformation test shows the repeatability in the analysis and 1.3% difference between the estimated thermocline thickness and that of numerical result.
