        Transmission electron microscopy (TEM) is a versatile and powerful technique that enables direct visualization of biological samples of sizes ranging from whole cell to near-atomic resolution details of a protein molecule. Thanks to numerous technical breakthroughs and monumental discoveries, 3D electron microscopy (3DEM) has become an indispensable tool in the field of structural biology. In particular, development of cryo-electron microscopy(cryo-EM) and computational image processing played pivotal role for the determination of 3D structures of complex biological systems at sub-molecular resolution. Here, basis of TEM and 3DEM will be introduced, especially focusing on technical advancements and practical applications. Also, future prospective of constantly evolving 3DEM field will be discussed, with an anticipation of great biological discoveries that were once considered impossible.

        Poxvirus under the eyes of electron microscope

        Zoonotic poxvirus infections pose significant threat to human health as we have witnessed recent spread of monkeypox. Therefore, insights into molecular mechanism behind poxvirus replication cycle are needed for the development of efficient antiviral strategies. Virion assembly is one of the key steps that determine the fate of replicating poxviruses. However, in-depth understanding of poxvirus assembly is challenging due to the complex nature of multi-step morphogenesis and heterogeneous virion structures. Despite these challenges, decades of research have revealed virion morphologies at various maturation stages, critical protein components and interactions with host cell compartments. Transmission electron microscopy has been employed as an indispensable tool for the examination of virion morphology, and more recently for the structure determination of protein complexes. In this review, we describe some of the major findings in poxvirus morphogenesis and the contributions of continuously advancing electron microscopy techniques.

      • 3D Electron Microscopy: A Versatile and Unique Tool for Structural Biology

Thanks to numeroustechnical breakthroughs and monumental discoveries, 3D electron microscopy (3DEM) has become an indispensabletool in the field of structural biology. In particular, development of cryo-electron microscopy and computational imageprocessing played pivotal role for the determination of 3D structures of complex biological systems at sub-molecularresolution. Analysis of crystalline object, single particle analysis and electron tomography are among the most widely used3DEM approaches, and each technique has unique advantages depending on the nature of the biological specimen. While3DEM analysis is now routinely performed using various resources that are publically available, the technique is constantlyevolving in order to understand biological insights that were once considered impossible.

      • 有機化合物의 Zone 精製法

        Bases of the theory of zone melting of organic compounds and discussed in relation to the thermodynamic equilibrium and phase diagram for solid solutions and eutectic mixtures of organic compounds. Then, some of the important theories and principles are discussed underlying the redistribution of solute concentration along the freezing solid-liquid interface during zone melting, and the equations are given for each case of complete mixing, partial mixing with boundary layer approach, and no mixing with diffusion only of the molten zone after single pass as well as multipass up to the ultimate distribution. Discussed next are some of the intrinsic problems and discrepancies that arise during the practical zone melting process such as the constitutional subcooling, absorption effect, gas and void formation, and effective distribution coefficient. The techniques and practical problems involved are then considered for the container, travel speed and direction of the zone, and the zone size together with the heating as well as mixing of the zone. Finally, some of the important applications and the related topic of zone melting of organics are mentioned. Since the main application so far has been the zone refining of organic compounds, special attention is given to some detail to the problems and techniques concerning the ultrapurification

      • 유전체 다층막 협대역 간섭 필터의 설계

        Design method for the Fabry-Perot type dielectric multilayer narrowband interference filter is presented. The filters are obtained by manipulating and matching quarterwave stacks of dielectric multilayers separated by one or two spacers. The stacks consist of proper numbers of alternate high and low refractive index films, each having a quarterwave optical thickness at design wavelength. Expressions for the transmittance of each filter system are presented in terms of the characteristic matrix. Design examples are given using ZnS and cryolite as the respective high and low refractive index film materials. Glass is used as the substrate material, and air is the incident medium. Computed performances of the spectral transmittance of the examples are compared at normal as well as at oblique incidence.

      • Zero Order Phase Retardation Waveplate Designed by Tilting the Optic Axis

        현재 관례적으로 널리 쓰이고 있는 유형의 복굴절 위상지연 파장판에서는 광축의 방위가 판면에 평행이 되도록 놓여 있으므로, 실용적인 두께의 파장판 한 장 만으로는 높은 위수의 위상지연만이 가능하며, 보다 성능이 좋은 영위수 위상지연을 얻기 위하여서는 두 장의 높은 위수의 파장판을 광축방위가 서로 수직이 되도록 접착하는 편법을 사용하고 있다. 본 논문에서는 영위수 파장판을 만드는 데 있어서 종래와 같은 제작과정의 번거로움을 피하고 비용의 절감을 위하여 관례적인 파장판과는 달리 광축 방위를 기울여 놓음으로써 한 장의 파장판 만으로 영위수 위상지연을 일으키는 새로운 형태의 파장판을 설계하였다. 이론적인 배경과 함께 설계과정을 제시하고, 설계결과를 실제 보기를 통하여 종래의 파장판과 비교 평가 하였다.

      • 水溶液에서 生長시킨 NaCl:Ni^+2 單結晶의 生長階段에 關한 硏究

        Experimental results indicate that the growth step of NaCl:Ni^+2 single crystal take a periodic distribution of step spacing and the step spacing is bigger for larger amount of Ni. This is explained in terms of the theory of step distribution by Schwoebel and Shipsey and by Gilmer and Bennema. It can be concluded for this crystal that k_??k+?? holds and impurity causes the increase in k_ value. The reason for the concave surface of the central portion of the crystal is also explained.
