      • 참취(Aster scaber Thunberg) 정단배양에서 대량증식에 미치는 Thidiazuron의 효과

        Shoot tips of Aster scaber Thunberg were cultured on Murashige and Skoog's(1962) medium containing various concentrations of growth regulators. Friable callus were effectively initiated on 0.5 ㎎/L 2,4 - D and 0.1 mg/L TDZ(thidiazuron) or BA than 0.5 - 2.0 ㎎/L 2,4 - D only. TDZ stimulated shoot proliferation in shoot tip explants of A. scaber. Shoot numbers treated with TDZ were greater than the number produced on the medium with 0.1 ㎎/L BA, but root formation and shoot elongation were inhibited by TDZ during the culture period. Multiple shoots were differentiated from the callus on medium supplemented with 0.1 ㎎/L TDZ. Multiple shoots were formed from greenish compact callus, these shoots had narrow and distorted leaves, secondary shoots directly developed from leaves of regenerated shoots, rosette type shoots had short internode. To establish in vitro propagation system of Aster, shoots constantly maintained when these shoots were transferred to 0.2 ㎎/L NAA plus 0.5 ㎎/L TDZ, and normal plants were obtained on the MS basal medium after 2 to 3 subcultures.

      • Hedera helix의 組織培養에 關한 硏究 : Ⅰ. Callus 形成 및 器官分化에 미치는 生長調節物質의 影響 Ⅰ. Effects of Growth Regulators on the Callus Formation and Organogenesis

        Hedera helix의 葉柄과 Leaf disc를 切取하여 生長調節物質을 달리한 Blayde's 基本培地에 培養하였던바, 그 結果를 要約하면 다음과 같다. 1. NAA 2㎎/ℓ+Kinetin 2㎎/ℓ에서 Callus의 形成이 가장 旺盛하였다. 2. 誘起된 Callus로부터 不定根의 發生은 NAA 5㎎/ℓ單獨處理에서 가장 효과가 있었다. 3. 不定根의 發生樣相은 Leaf disc에서는 多束狀으로 葉柄에서는 산발적으로 나타났다. 4. 2,4-D가 組合된 培地에서 Callus의 褐變現象이 나타났으며, 2,4-D의 濃度가 높을수록 현저했다. The leaf petioles and discs of Hedera helix were cultured on the Blayde's basic medium supplemented with various combinations of growth regulators. The results obtained are summarized as follows : 1. The calluses were induced vigorously from the Blayde's medium supplemented with 2㎎/1 NAA and 2㎎/1 kinetin. 2. The best growth of adventitous roots appeared on the calluses that had formed on the media cantaining only 5㎎/1 NAA. 3. Multiple roots were formed on the calluses of leaf disc, whereas single roots on the those of leaf petiole. 4. Brownish calluses were observed on the media with 2, 4-D.

      • 스테비아 잎과 마디 배양에 의한 부정신초 형성 및 식물체 재분화

        고빈도 스테비아 식물체를 재분화 시키기 위해 잎과 마디를 2,4-D, NAA, IBA와 BA를 혼용처리한 MS 배지에 배양하였다. 절편체 종류 및 식물생장조절물질의 종류와 농도가 캘러스 유도, 신초분화 및 식물체 재분화에 미치는 효과를 조사하였다. 잎 절편체에서는 캘러스형성이, 마디 절편체에서는 액아의 분화 및 신장이 양호하였으며 잎절편 배양결과 0.5mg/L 2,4-D와 0.5mg/L BA 혼용처리시 3.5%, 2.0mg/L NAA와 2.0mg/LBA 혼용처리시 24%, 3.0mg/L IBA와 3.0mg/LBA 혼용처리구에서는 36.2%신초가 분화되어 가장 효과적이었다. 마디배양의 경우 마디의 위치 및 치상방향에 따라 캘러스형성, 액아 분화 및 신장에 영향을 미쳤으며 캘러스 형성에는 정단부위쪽 마디가, 그리고 액아의 분화 및 신장은 정단부위 보다는 중앙 및 하단 부위가 양호하였다. 마디의 수직 치상이 수평치상에 비해 액아 분화 및 신장에 효과적이었고, 잎 절편배양에서와 같이 2,4-D와 BA, NAA와 BA 혼용처리보다는 IBA와 BA 혼용처리가 캘러스형성이 양호하였으며, 특히 3.0mg/L IBA와 3.0mg/L BA 혼용처리에서 배양 개체 당 6개의 식물체의 재분화 되어 가장 효과적이었다. In order to produce the high frequency regenerated plants, leaves and stem node segments of Stevia rebaudiana Bertoni M. were cultured on MS medium containing 2,4-D, NAA, IBA and BA. We investigated the effects of kinds of explants, kinds and concentration of plant growth regulators on callus induction, shoot differentiation and plant regeneration. Callus induced easily in the leaf segment and axillary bud or shoot are ready to differentiate or elongate in the stem node segment. In the leaf segment culture, percentages of shoot differentiation were 3.5%, 24% and 36.2% through on MS medium with 0.5 mg/L 2,4-D plus 0.5mg/L BA, 2.0mg/L NAA plus 2.0mg/L BA and 3.0mng/L IBA plus 3.0mg/L BA, respectively. The part of stem node and direction of inoculation affected significantly on callus induction, axillary bud differentiation and shoot elongation in the culture of stem node. The upper part of stem node with apical meristem is effective on the callus formation and the bottom part of that is apt to differentiate the axillary bud or elongate the shoot. Effective differentiated axillary bud could be recognized on the vertical inoculation than horizontal inoculation of stem node. The combination of IBA plus BA is the best plant growth regulators among the that of 2,4-D plus BA and NAA plus BA as like leaf segment culture. Eespecially, six plants per explant regenerated on the MS medium with 3.0mg/L IBA plus 3.0mg/L BA.

      • Hydroculture에 의한 觀葉植物의 發根 및 生長

        The objectives of this study were to determine the effect of rooting and sprouting of cuttings were taken the forth kinds of foliage plant ill hydroculture. Forth kinds of foliage plant, dracaena sanderiana, Euphorea milii, Hedera canarienis, Scindapsus aureus were placed in 1200, 800, 500 lux light intensity greenhouse for 12 weeks. Rooting from cuttings was not influenced by Hydroball and Zeolite media. However, root weight was the highest in Scindapsus plants. The media affected root development when foliage plants were placed under water deficiency. Scindapsus grown under water deficiency was occurred root heavily having root hair in Hydroball. Origin tissue of root was significantly differed by plant species. Hedera occurred from the upper epidermis of cut sites. Especilly Euphorbia formed the callus from cuttings but this callus inhibited rooting of cuttings. Shoot growth from cuttings was influenced by media and light intensity. Number of new leaves was maximized under 1200 lux light intensity and number of leaves and bud were increased in Zeolite than Hydroball.

        대규모 온라인 게임을 위한 XML 기반의 메시지 정의 시스템

        대규모 온라인 롤-플레잉 게임(MMORPG : Massively Multiplayer Role-Playing Game)에서 클라이언트와 서버 간의 상호작용은 대개 네트워크를 통한 메시지 전달을 통해 행해진다. 이런 MMORPG들에서는 게임의 내용이나 구성요소들이 지속적으로 진화해 가므로, 그에 따른 메시지들의 추가, 삭제, 또는 변경이 필요하다. 그러므로 게임 프로그래머들은 반복적으로 새로운 프로그램을 작성하거나 현재 있는 프로그램들을 수정해야 한다. 본 논문에서 소개하는 XML 기반의 메시지 정의 시스템은 게임의 상호작용을 위한 메시지들의 추가 및 변경을 용이하게 해준다. 이 시스템은 프로그래머들이 기존 메시지들을 변경하거나 새로운 메시지들을 추가하는데 드는 시간을 상당히 줄여 줄 뿐만 아니라 프로그래밍 버그 발생의 부담을 상당히 줄여준다. 또한 이 시스템은 일반 클라이언트-서버 응용 프로그램들에서도 사용이 가능하다. Client- server interaction in a massive multi-player online role-playing game(MMORPG) is usually made via network-message Passing. Since the game contents and elements in such MMORPGs are consistently evolved, messages need to be added, deleted, or modified accordingly. Therefore, game programmers are repeatedly required to write new programs or at least modify existing programs. The XML-based message definition system introduced in this paper facilitates the addition and modification of messages for game interaction ; it not only allows programmers to save a considerable amount of time for modifying existing messages or adding new messages but also relieves them of a serious burden of programming bugs. In addition, this system can be used for general client-server applications.

        This studies were performed to establish in vitro clonal multiplication of cordyline terminalis CV. Ti through ovary culture in Murashige and Skoog's media containing various levels of NAA(α-naphthalene acetic acid) and BA (benzyl adenine The calluses were induced vigorousely from the medium supplemented with 1mg/L NAA and 1-6mg/L BA. The differentiation of multiple-shoots was promoted on the medium containing 1mg/L NAA and 4mg/L BA. The numerous adventitious roots were initiated from the proximal part of multiple-shoots. After 8 months of callus growth and differentiation, each ovary tissue yielded 80 to 100 plantlets.

      • 딸기의 組織培養에 관한 硏究 : I.pH가 caullus誘起에 미치는 影響 I.Effect of pH on formation of calli

        This study was carried out to determine the optimum pH of medium for callus culture of strawberry(Fragaria grndiflora EHRH). Shoot apices and stolons of strawberry were cultured on the Murashige and Skoog's medium supplemented with 2,4-D 4mg/l plus on 0.2mg/l of kinetin, and in different pH levels(pH5.0-7.5). The results obtained were as follows: 1. Liquid callus was observed on stolen and shoot apex culture, but friable callus was only induced on stolen culture. 2. The optimum pH for the best callus growth and rapid multiplication of stolon was pH6.0, and that of shoot apex was pH 5.5. 3. With regards to relation between growth of adventitious roots and pH of medium, low levels of pH in medium caused rapid rooting from callus and abundant callus formation.
