This study was conducted to find the optimum transformation condition using Agrobacterium harboring promoterless GUS gene. The optimal medium for shoot induction from leaves of Populus nigra×P. maximowiczii was MS medium supplemented with 0.1㎎/ℓ NAA, 0.5㎎/ℓ BAP(94% regeneration frequency and 11.5 average number of shoot) According to the test using pBI121, the concentration of antibiotics for selection marker gene was 100㎎/ℓ kanamycin or 60㎎/ℓ geneticin in the SIM(shoot inducing medium) 3. Two weeks later, callus was induced in the SIM 3 and this callus grew up to 0.5-1cm shoots after 6 weeks in the new SIM 3. And the treatment with methylation inhibitor(5-azacytidine) led to a dramatic increase in foreign gene expression rate from 5.7% to 26.7% . The vector systems showed. different transformation efficiencies based on the fluorometric and histochemical GUS assay. In this study the vector systems used for transformation seemed to affect transformation frequency, in which pEHA101 yielded more transformants(35.9%) than LBA4404/pBI121 did(5.7%). This result indicated that pEHA101 was effective to insert the promoterless foreign gene into a poplar genome.
노각나무 성숙 접합자 배로부터 부정아 유도에 미치는 BA 및 NAA 효과

Zygotic embryos of Stewartia koreana Nakai were cultured to determine the effects both of BA and NAA on adventitious shoot induction. Multiple shoots (about 11 per explant) were formed when the embryos were treated with 1.0 mg/L BA alone. On the other hand, NAA appeared to inhibit shoot induction when treated with BA. Adventitious shoots looked differ in form and color by the combinations of BA and NAA treated. When both BA and NAA were present, the shoots became short and developed dark color. The highest rooting was observed at 0.5 mg/L NAA. The results could be useful for the establishment of in vitro regeneration system for Stewartia koreana Nakai.
The Time for Collecting of Cryptomeria japonica Seeds

The time of seed collection is regarded as one of major concerns to obtain sound seeds. The physical and germinal aspects of Cryptomeria japonica D. Don (Taxodiaceae) seeds were analyzed to determine the optimum harvesting time in Korea. Cones were picked every 10 days from the 30th of July to the 30th of October in both 2005 and 2006. Seeds were collected from picked cones. Seed size and weight were not significant in two consecutive years. The 1,000-seed weight was 3.3 g for cones picked at the 18th of August and 5.3 g for cones picked at the 30th of September. The size of seeds was increased as the time of collection from the 18th of August to the 30th of September: from 19.3 mm to 21.3 mm in length and from 15.8 mm to 18.5 mm in width. Average germination rates in 2005 was 18.3% and 19.6% in 2006. The highest germination rate was 34.3% from seeds collected at the 30th of September in 2005. In 2006, the highest germination rate was 31.7% for seeds collected at the same date as the 2005 seeds. After the end of September, germination rate was decreased in both years. The results implied that the best cone picking time for Korean C. japonica seeds is around the end of September.
