      • Corroded and loosened bolt detection of steel bolted joint based on improved you only look once network and line segment detector

        Steel bolted joint is an important part of steel structure, and its damage directly affects the bearing capacity and durability of steel structure. Currently, the existing research mainly focuses on the identification of corroded bolts and corroded bolts respectively, and there are few studies on multiple states. A detection framework of corroded and loosened bolts is proposed in this study, and the innovations can be summarized as follows: (i) Vision Transformer (ViT) is introduced to replace the third and fourth C3 module of you-only-look-once version 5s (YOLOv5s) algorithm, which increases the attention weights of feature channels and the feature extraction capability. (ii) Three states of the steel bolts are considered, including corroded bolt, bolt missing and clean bolt. (iii) Line segment detector (LSD) is introduced for bolt rotation angle calculation, which realizes bolt looseness detection. The improved YOLOv5s model was validated on the dataset, and the mean average precision (mAP) was increased from 0.902 to 0.952. In terms of a lab-scale joint, the performance of the LSD algorithm and the Hough transform was compared from different perspective angles. The error value of bolt loosening angle of the LSD algorithm is controlled within 1.09%, less than 8.91% of the Hough transform. Furthermore, the proposed framework was applied to fullscale joints of a steel bridge in China. Synthetic images of loosened bolts were successfully identified and the multiple states were well detected. Therefore, the proposed framework can be alternative of monitoring steel bolted joints for management department.

        A New Boiler-turbine-heating Coordinated Control Strategy to Improve the Operating Flexibility of CHP Units

        The operating flexibility of the power units is getting increasing attention from power systems especially those with large-scale fluctuating renewable energies. However, the combined heat and power (CHP) units are getting a bottleneck because their electricity productions are restricted by heat productions. This study aims to develop an electric-heat coordinated control strategy to make the CHP units more flexible. First of all, the dynamic model for a 300 MW CHP unit is set up, and its linear state-space description is obtained. A control strategy based on linear quadratic regulator (LQR) is then developed to satisfy different heat-power demands in various operating conditions. The control weights Q and R are optimized by particle swarm optimization. Moreover, the improved coordinated control strategy based on precise energy balance is put forward to increase the CHP power ramp rate considering electricity priority strategy and recovery control of the heat source. Finally, the simulation results show that the improved strategy is suitable for various CHP operating scenarios, and the case for electricity priority and heat recovery control significantly improves the unit power rate on the premise of stable heat supply. This work provides a reliable and flexible control mode for CHP units, which can support the power system stability and renewable energy integration.

        Ginsenoside Rb1 Inhibits Doxorubicin-Triggered H9C2 Cell Apoptosis via Aryl Hydrocarbon Receptor

        Doxorubicin (DOX) is a highly effective chemotherapeutic agent; however, the dose-dependent cardiotoxicity associated with DOX significantly limits its clinical application. In the present study, we investigated whether Rb1 could prevent DOX-induced apoptosis in H9C2 cells via aryl hydrocarbon receptor (AhR). H9C2 cells were treated with various concentrations ($-{\mu}M$) of Rb1. AhR, CYP1A protein and mRNA expression were quantified with Western blot and real-time PCR analyses. We also evaluated the expression levels of caspase-3 to assess the anti-apoptotic effects of Rb1. Our results showed that Rb1 attenuated DOX-induced cardiomyocytes injury and apoptosis and reduced caspase-3 and caspase-8, but not caspase-9 activity in DOX-treated H9C2 cells. Meanwhile, pre-treatment with Rb1 decreased the expression of caspase-3 and PARP in the protein levels, with no effects on cytochrome c, Bax, and Bcl-2 in DOX-stimulated cells. Rb1 markedly decreased the CYP1A1 and CYP1A2 expression induced by DOX. Furthermore, transfection with AhR siRNA or pre-treatment with AhR antagonist CH-223191 significantly inhibited the ability of Rb1 to decrease the induction of CYP1A, as well as caspase-3 protein levels following stimulation with DOX. In conclusion, these findings indicate that AhR plays an important role in the protection of Ginsenoside Rb1 against DOX-triggered apoptosis of H9C2 cells.

        Analysis of transcriptomic profiles and physiological traits of exogenous 24-epibrassinolide alleviating salt stress in Atractylodes macrocephala Koidz.

        Exogenous 24-epibrassinolide (24-epiBL) application is a preferred method for improving salt tolerance in plants. However, information about the effects of exogenous 24-epiBL application related to salt response in Atractylodes macrocephala Koidz. (AMK) has rarely been reported. Here, we aimed to investigate the effects of exogenous 24-epiBL (0.01 mg L−1) on enhancing salt tolerance in AMK seedlings under salt stress (5 g L−1 NaCl) based on seed germination experiments. Our results indicated that NaCl treatment suppresses AMK seed germination, while 24-epiBL treatment under nonstress and stress conditions had positive effects on plant seedling growth. Treatment with NaCl significantly inhibited radicle elongation and root activity to endure salt stress, and 24-epiBL treatment promoted growth and alleviated the damage to osmotic stress, ion stress sub- stances, and oxidative stress by regulating free proline (Pro) and soluble protein (SP) contents and enhancing soluble sugar (SS) conversion. Moreover, the expression of normal development process genes was affected, and differentially expressed genes (DEGs) in multiple pathways associated with amino acid metabolism, signal transduction, carbohydrate metabolism, and lipid metabolism were regulated under NaCl treatment. Further analysis of the AMK transcriptome profile revealed that exogenous 24-epiBL enhanced salt tolerance through hormone regulation and carbohydrate metabolism promotion, salt- responsive gene expression inhibition, and induction of pathways related to terpenoid and polyketide metabolism as well as pentose and glucuronate interconversions for plant growth promotion. Collectively, our findings successfully demonstrate that exogenous 24-epiBL plays a positive role in regulating the AMK response to salt stress, which provides potential genetic resources to improve salt tolerance in plants.

        Tanshinone IIA Protects Endothelial Cells from H<sub>2</sub>O<sub>2</sub>-Induced Injuries via PXR Activation

        Tanshinone IIA (Tan IIA) is a pharmacologically active substance extracted from the rhizome of Salvia miltiorrhiza Bunge (also known as the Chinese herb Danshen), and is widely used to treat atherosclerosis. The pregnane X receptor (PXR) is a nuclear receptor that is a key regulator of xenobiotic and endobiotic detoxification. Tan IIA is an efficacious PXR agonist that has a potential protective effect on endothelial injuries induced by xenobiotics and endobiotics via PXR activation. Previously numerous studies have demonstrated the possible effects of Tan IIA on human umbilical vein endothelial cells, but the further mechanism for its exerts the protective effect is not well established. To study the protective effects of Tan IIA against hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) in human umbilical vein endothelial cells (HUVECs), we pretreated cells with or without different concentrations of Tan IIA for 24 h, then exposed the cells to 400 μM H<sub>2</sub>O<sub>2</sub> for another 3 h. Therefore, our data strongly suggests that Tan IIA may lead to increased regeneration of glutathione (GSH) from the glutathione disulfide (GSSG) produced during the GSH peroxidase-catalyzed decomposition of H<sub>2</sub>O<sub>2</sub> in HUVECs, and the PXR plays a significant role in this process. Tan IIA may also exert protective effects against H<sub>2</sub>O<sub>2</sub>-induced apoptosis through the mitochondrial apoptosis pathway associated with the participation of PXR. Tan IIA protected HUVECs from inflammatory mediators triggered by H<sub>2</sub>O<sub>2</sub> via PXR activation. In conclusion, Tan IIA protected HUVECs against H<sub>2</sub>O<sub>2</sub>-induced cell injury through PXR-dependent mechanisms.
