      • 玄蔘의 Saponin 成分 檢索 : on the Saponin of the Radix

        Crude saponin(100g) were obtained by extracting the radix(5kg) of Scrophalariae koraiensis Nakai. Crude saponin were positive in the Liebermann-Burchard test. We observed 5 sports by TLC using BuOH saturated with H_2O : ethyl acetate : water(4:1:5, upper phase) as solvent and 1% Ce(SO_4)_2 in 10% H_2SO_4 as a color former. We divided into 3 fractions by common column chromatography using BuOH saturated with H_2O : ethylacetate : water(4:1:5, upper phase) as solvent.

        영병에 關한 文獻的 考察

        영병은 頸部의 結候兩側에 腫瘍이 발생되는 질병을 總稱하는 것으로 包括的인 면으로 볼 때 西洋醫學적으로 볼 때 甲狀腺腫大와 같은 것으로 본다. 이러한 병에 對하여 이미 오래전부터 地理적 環境과의 相關關係를 言及하였을 뿐만 아니라 좀더 나아가서는 情志失調도 이병을 일으키는 主要原因이 된다고 보았다. 現代醫學에 있어서는 甲狀腺腫大는 갑상선기능항진증에서 볼수 있는 것으로 보아 영병과 갑상선기능항진증과의 密接한 關係가 있음을 暗示해 주고 있다. 이와같이 古典적인 病名과 現代적인 症狀과의 比較分析을 위해 文獻考察方法도 韓醫學發展에 寄與할것으로 思慮된다. Goiter means all diseases causing tumor on the both sides of Adam's apple, and in the western medicine it is similar to tumefaction in thyroid gland. In the past the correlation between geographical features of deficiency of the kidney Yin as an important cause. In the present medicine, they suggest there should be close relation between goiter and hyperthyroidism. Therefore the comparative literature study on the ancient names of disease and present symptoms will contribute to the development of the Korean oriental medicine. The results of this study are summarized as follows: 1. In the past, the stagnation of qi and phlegm due to soil and water-borne diseases, and damage on being the response of the mind to the environmental stimuli was considered as the greatest causes of goiter, whereas in the present they give attention to goiter's symptoms caused by deficiency of Yin and think of deficiency of the kidney yin as important cause. 2. The ancient medicine books described the symptoms centering on the outer signs having tumefaction of diffuse on the neck, however modern oriental medicine writes symtoms of the whole body caused hyperactivity of yang due to yin deficiency inqi goiter addition to the outer symtoms and they are very similar to hyperthyroidism. 3. For the treatment, according to the treatment for the differentiation of symptoms and signs, they mainly use smoothing the liver and regulating the circulation of qi for the stagnation of qi; promoting blood circulation to remove blood stasis for the blood stasis; resolving sputum and softening hard masses for the lumps of phlegm; removing heat phlegm for the stagnation of phlegm fire, however in recent books we can find many ways for the nourishing Yin and prescription.

        Chlorpromazine의 유전적 안전성에 관한 연구

        연구목적 ; 항 정신성약물로 사용빈도가 높은 chlorpromazine을 사용하여 이 약물의 돌연변이원성과 유전독성을 알아보았다. 방 법 : Bacillus subtilis을 사용한 DNA손상성검토(rec assay), Salmonella typhimurium을 이용한 돌연변이원성실험(Ames test 및 SOS umu test) 그리고 생쥐에 chlorpromazine를 직접 주입하여 대사 후에 돌연변이원성을 확인하는 host-mediated assay법을 각각 사용하였다. 결 과 ; Rec assay에서 chlorpromazine은 Bacillus subtilis의 DNA에는 별다른 영향을 미치지 않았으며, salmonella typhomurium TA 98 및 TA100을 이용한 돌연별이원성 실험에서도 chlorpromazine은 어느 균에도 돌연변이원성을 나타내지 않았으며, S-9 mixture의 첨가에 의해 대사가 된 후에도 돌연변이원성을 나타내지 않았다. 또한 SOS umu test의 경우에도 β-galactosidase활성에는 별다른 영향을 미치지 않는 것으로 보아 chlorpromazine은 돌연별이원성을 일으키지 않는 것으로 판정되었으며 S-9 mixture 처리 후에도 이와 유사한 결과를 얻었다. 한편 생쥐를 이용한 host mediated assay에서도 Ames test에서 S-9 mixture를 첨가 한 경우와 같이 돌연변이원성이 없는 것으로 판정되었다. 결 론 : 이상을 종합해 볼 때 chlorpromazine은 본 실험에 사용한 4가지 방법에서는 돌연병이원성과 유전독성을 일으키지 않는 것으로 나타났다. Object : The aim of this study is to determine whether exposure to chlorpromazine causes mutagenicity and genetic disorders. Method : Ames(Salmonella typhimurium) test and Rec assay(Bacillus subtilis) were used as indicators for DNA damage. Furthermore, the levels of umu operon expression by measuring the β-galactosidase activity were monitered with the SOS umu test using S. typhimurium 1535 containing plasmid pSK1002. And the host-mediated assay was used to investigate the mutagenicity of chlorpromazine after the activation with in vivo metabolic systems. Results : From the results, chlorpromazine did not affect DNA of S. typhimurium and B.subtilis strains and showed no mutagenicity at the all concentrations tested. These phenomena was also similar to that after metabolic activation of chlorpromazine in in vivo system. Conclusion : These results suggested that chlorpromazine did not show the mutagenicity and genotoxicity by four different methods used in this study.

      • Phosphamidon 분해세균의 분리동정 및 생분해능

        토양으로부터 유기인계 살충제인 phosphamidon을 분해하는 세균들을 분리하고 Biolog system을 이용하여 동정하였다. 그람양성 세균들은 모두 Bacillus 속에 속하는 종들이었으며 그람음성 세균들은 토양에서 우점하지 않는 세균들이 많았다. 이들중 phosphamidon 함유배지에서 생장률이 높은 균주들을 선택하여 phosphamidon 분해능을 조사한 결과 Capnocytophaga gingivalis로 동정된 YD-17 균주가 가장 높은 생분해능을 나타내어 1000 ppm의 phosphamidon이 배양 21일 후 9.4%의 잔류량을 보였으며 이는 대조구에 비해 제거율이 52% 증가된 결과였다. 이 때 균주의 생장을 단백질량으로 측정하였는데 분해균주들이 고농도의 phosphamidon에 의해 저해되지 않고 지속적인 생장을 하여 phosphamidon을 탄소원으로 이용하는 생분해가 일어난 것을 확인할 수 있었다. Organphosphorus insecticide phosphamidon-degrading bacteria were isolated from agricultural soils and identified using Biolog microtiter assay. All Gram-ositive degrading bacterial strains belong to genus Bacillus and many Gram-negative bacteria were rare soil species. Among them fast growing strains on phospamidon-containing minimal medium were selected and their biodegrading capability were measured. YD-17 which was identified as Capnocytophaga gingivalis showed the highest biodegradation rate. It could increase the removal of phosphamidon up to 52%. During the biodegradation continuous increase of amount of cell protein was observed, which, indicated that phosphamidon was utilized as a carbon source for phosphamidon-degrading bacteria.

      • 염분이 함유된 폐수의 생물학적 처리시 염분농도가 오염물질의 제거에 미치는 영향

        Effects of salt concentration and C/N ratio on pollutants removal efficiencies were investigated in treatment of seafood wastewater by sequence batch reactor(SBR). The synthetic seafood wastewater containing salinity from 3,000 to 10,000 mg/L NaCI and C/N ratio of 2 and 4 was treated by SBR. Removal efficiencies of COD_(Mn), T-N, and T-P decreased with increasing salt concentration, but eventually reached a pollutants removal a platrau. As systems adapted in salts until salts concentration of 8,000mg/L, the time required to reach the pollutants removal efficiencies at steady state was reduced. However, the removal efficiencies of pollutants sharply decreased and the time require to reach steady state was increased as salt concentration rose to 10,000mg/L, Removal efficiencies of COD_(Mn) were higher for C/N ratio of 4 than for 2. Effects of salt on the nitrogen removal were not faster than the organic removal but the time required to the nitrogen removal at steady state was ncreased. The removal efficiencies of phosphorus also were redueed as salt concentration increased, which inhibited the release of phosphorus than the uptake.

      • 통풍성 관절염의 임상적 고찰

        통풍은 Purine 대사의 이상으로 발생하는 질환으로, 고뇨산혈증인 사람의 전부가 통풍으로 발현되지 않고 증상의 출현 양상이 다양하여 진단과 치료에 주의가 필요하며, 조기에 적절한 조치를 한다면 충분히 조절이 가능한 질환이다. 본대학 내과학교실에서는 통풍으로 치료한 32명의 환자에서 임상양상, 병력과 검사소견을 분석하여 다음과 같음 결과를 얻었다. 1. 32명 모두 남자이며, 최초 발병 시기는 24세에서 72세로 평균 43.8±11.9세이었으며, 30대에서 40대까지가 19명(59.4%)으로 대부분을 차지하였다. 내원 당시 나이는 27세에서 75세까지로 평균 52.3±10.4세였으며, 내원시 까지 평균 유병기간은 8.5±6.8년으로 나타났다. 2. 동반 질환으로는 고지혈증 12례, 신장질환 10례, 고혈압 12례, 비만 8례, 당뇨 2례 등이었다. 3. 이환된 관절은 단관절 침범이 19례(59.4%), 다관절 침범이 13례 이었으며, 최초 이한된 관절은 족무지 중족골지골 관절로 19례(59.4%)로 가장 많았고, 통풍 결절은 20례(62.5%)에서 관찰되었으며, 유병기간이 10년 이상된 12례중 11례 (91.7%)에서 결절이 관찰되었다. 4. 평균 혈중 요산치는 9.17±1.75 ㎎/dl이었으며, 8.0 ㎎/dl에서 9.9 ㎎/dl 사이가 19명으로 전체의 59.4%를 차지하였다. Objective: The gout is a heterogeneous group of diseases resulting from tissue deposition of monosodium urate or uric acid crystals from extracellular fluids supersaturated with respect to this end product of human purine metabolism. The clinical manifestations are such as hyperuricemia, gouty arthritis, gouty nephropathy, uric acid nephrolithiasis. We analyze of clinical manifestations and associated factors in gout. Method: We have reviewed the medical records, radiologic findings and clinical results of thirty-two patients admitted at our department from April 1996 to July 1997. Result: 1) All patients were male. The mean age at initial attack was 43.8 years old, ranging from 24 to 72 years old. 2) The mean level of serum uric acid was 9.17mg/dl on admission. 3) The first metatarsopharyngeal joint was involved in 19 cases (59.4%). Tophus was observed in 20 cases (62.5%). 4) Hyperuricemia was associated with hypertension, obesity, nephrolithiasis and hyperlipidemia.

      • 동종 골수이식 후 만성 이식편대숙주질환과 동반되어 신증후군으로 발현된 IgA 신병증 1예

        동종 골수이식 후 발생하는 신기능부전은 원인이 다양하여 감별에 어려움이 있다. 만성 GVHD는 흉선의 기능저하에 따른 면역관용의 실패로 발생되는 자가면역현상에 의해 발생되는 것으로 알려져 있으며 드물게 신장을 침범하여 신증후군을 일으키기도 한다. CaA의 용량을 줄이거나 투약중지 후 발생한 만성 GVHD에 동반된 신증후군의 경우 조기에 신조직검사를 시행하여 감별진단하고 CsA의 용량을 증량하거나 재투약하여 좋은 치료 결과를 보일 것으로 기대된다. 저자들은 문헌고찰을 통해 동종 골수이식 후 발생된 신증후군이 유사한 임상양상을 보이며 발생기전에 만성 GVHD와 자가면역현상이 연관되어 있음을 확인하였으며 동종 골수이식 후 신증후군의 임상양상을 보이는 환자에서 병리조직학적으로 lgA 신병증을 진단하고 이러한 lgA 신병증이 발생기전에 만성 GVHD와 자가 면역현상이 연관되어 있을 가능성을 최초로 확인하였으며 CsA를 이용하여 성공적으로 치료한 경험을 하였기에 보고하는 바이다. Renal insufficiency is occasionally encountered in allogeneic bone marrow transplantation and its cause is difficult to ascertain. Chronic graft-versus-host disease (GVHD) related to thymic dysfunction is immune-mediated and involves autoreactivity of T-lymphocytes derived from donor marrow to recipient's major histocompatibility complex(MHC) minor antigens. The clinical mainifestations of chronic GVHD are similar to those of autoimmune disease but kidney involvement is rare. Few cases of nephrotic syndrome with membranous nephropathy or minimal change nephrotic syndrome have been reported to be associated with chronic GVHD in allogeneic bone marrow transplant recipient especially after cyclosporine A(CsA) withdrawal, and these cases have responded well to CsA. Therefore, it is prudent to differentiate the cause of post-transplant renal insufficiency using renal biopsy and to start CsA in a case of nephrotic syndrome associated with chronic GVHD as early as possible. We report a case who had a massive proteinuria during the post-allogeneic marrow transplantation period. The cause of nephrotic syndrome was diagnosed as IgA nephropathy by renal biopsy. The patient was successfully treated with reintroduction of CsA.

        Lipid Peroxidation and Its Toxicological Implications

        Lipid peroxidation is a free radical oxidation of polyunsaturated fatty acids such as linoleic acid or arachidonic acid. This process has been related with various pathologies and disease status mainly because of the oxidation products formed during the process. The oxidation products include reactive aldehydes such as malondialdehyde and 4-hydroxynonenal. These reactive aldehydes can form adducts with DNAs and proteins, leading to the alterations in their functions to cause various diseases. This review will provide a short summary on the implication of lipid peroxidation on cancer, atherosclerosis, and neurodegeneration as well as chemical and biochemical mechanisms by which these adducts affect the pathological conditions. In addition, select examples will be presented where antioxidants were used to counteract oxidative damage caused by lipid peroxidation. At the end, isoprostanes are discussed as a gold standard for the assessment of oxidative damages.

        Flavonoid accumulation in common buckwheat (Fagopyrum esculentum) sprout tissues in response to light

        <P>The content and accumulation patterns of flavonoids in response to light were investigated in common buckwheat (Fagopyrum esculentum) sprouts over the course of 9 days. Buckwheat sprouts were grown under fluorescent (FL), red (RL), and blue (BL) light sources, as well as in darkness, and their extracts were analyzed. Sprout elongation and chlorophyll content were high in the presence of red light. The contents of the major buckwheat sprout flavonoids, orientin, isoorientin, vetexin, isovetexin, and rutin all peaked 5 days after germination under all four growth conditions, then gradually decreased. Sprouting cotyledons accumulated around 1.5-fold more orientin, isoorientin, quercetin-3-O-robinobioside, and rutin in the presence of red light, including from the RL and FL light sources, than in blue light alone. The C-glycosyl flavones, orientin, isoorientin, vetexin, and isovetexin, were not detected in the stem tissues of the hypocotyls, but C-glycosyl flavonols, such as quercetin-3-O-robinobioside and rutin, were present, and were twice as abundant under BL and FL than in the RL and darkness conditions. In the root tissues, a small amount of C-glycosyl flavonols was detected after BL exposure only; however, the other flavonoids were not detected at all. These results indicated that red light induces flavonoid biosynthesis in the cotyledons of buckwheat seedlings, but blue light has a greater effect on the accumulation of C-glycosyl flavonols in other seedling tissues.</P>
