1) Ca-alginate bead에 고정화된 간세포의 생존도는 single cell의 경우가 배양 6일째까지 15% 선으로 떨어진 데 반하여 spheroid의 경우에는 40%선을 유지하여 single cell보다 spheroid의 고정화가 유리한 것으로 나타났다. 그러나 spheroid의 경우 초기 고정화 단계에서 세포 멸실률이 높았다. 2) 고정화된 spheroid의 암모니아 제거속도는 1.7∼2.8㎍ ammonia/㎍ ATP/hr이고 albumin 생성속도는 0.2∼0.4㎍ albumin/㎍ ATP/hr로써 고정화되지 않은 경우에 비해 상당히 감소하였다. 이는 alginate bead로 인한 물질전달 속도의 감소 때문으로 보여진다. 3) 하이브리도마 세포 (초기농도:2×10?cells/4.5㎖)와 간세포 spheroid(2×10?cells/10㎖, 생존율 약60%)를 coculture하여본 결과 배양 7일째까지는 하이브리도마 세포에 의해 생산되는 암모니아가 거의 (암모니아 평균농도:2㎍/㎖이하로) 제거되고 urea가 생성됨을 확인 하였다. 4) Ca-alginate bead에 고정화된 간세포 spheroid 2㎖(cell number : 2×10?cells/㎖)을 rat의 복강에 주사하여 배양해 본 결과 간세포의 생존율은 12일째 되는 날까지 50% 이상이었고 albumin 생성속도도 초기값의 80% 선을 유지하였다. 이상의 결과로 보아 고정화된 간세포는 in-vitro에서보다 in-vivo에서 더 안정된 것으로 보인다. Microencapsulation of hepatic spheroid using Ca-alginate was carried out and the change of cell viability, ammonia metabolism and albumin synthesis was studied. The viability of encapsulated hepatic spheroid was poor with respect to th free cells, but the hepatic functions such as ammoinia metabolism, tested using a hybridoma culture system. were found to support high during a week. When the hepatic spheroid bead was injected into mouse peritoneal cavity, after a week, the cell viability was about 50% and albumin synthesis rate was kept up to 80% of the initial value.
하이브리도마 세포의 유가식 배양에서 배지첨가속도가 단일클론 항체 생산에 미치는 영향

유가식 배양에서 배지첨가속도가 하이브리도마 세포의 증식과 항체생성속도에 미치는 영향을 조사하였다. 항체 비생성속도 q_0값이 변화는, 회분식 배양의 경우 배양 초기에 가장 높았다가 계속 감소하는 경향을 보였으나, 배양 48시간 이후부터 배지의 첨가가 시작되는 유가식 배양에서는 q_0값이 배지첨가 직후부터 빠른 속도로 증가하기 시작해 배양이 끝날 때까지 높은 값으로 유지되었다. 그리고 배지의 비첨가속도 q_f가 증가되면 항체의 비생성속도 q_0도 민감하게 증가하는 경향을 보였다. 생성된 최종 항체농도 C_fimaf은 배지첨가속도에 큰 영향을 받지 않고 거의 비슷한 값을 보였지만 평균적으로 회분식 배양보다는 그 값이 3배 이상 증가되어 최고 220㎍/㎖ 까지 향상되었다. 유가식 배양에서, 첨가배지의 성분 중 혈청농도를 0.5%까지 낮추고 glucose와 glutamine의 농도를 2배로 강화시킨 배지를 사용했을 때는 q_0값이 더욱 더 높아져 최종 항체농도가 회분식 배양에서 보다 4배 이상 높은 270㎍/㎖ 까지 향상되었다. 이 때 항체 비생성속도 q_p는 glutamine의 소비속도와 밀접한 관계가 있는 곳으로 나타났다. The effect of media feeding rate on cell growth and monoclonal antibody production in the fed-batch culture of hybridoma A4W was studied. In the batch culture, the highest specific antibody production rate was observed at the begining of the culture period but its value tended to decrease rapidly with the culture time. The final antibody concentration and volumetric productivity was 65㎍/㎖ and 13 ㎎ Mab/l/day, respectively. In the fed-batch culture, the specific antibody production rate, q_u rebounded sharply within a few hours after the media feeding was started and it remained high until the end of culture if the media feeding was continued. The final antibody concentration was 220 ㎍/㎖ and the volumetric productivity was 45.1 ㎎/l/day. Further increase in final antibody concentration was achieved by applying a modified media of which component was fortified with glucose and glutamine, hence the final antibody concentration in this case was 270 ㎍/㎖ and the volumetric productivity was 51.8 ㎎/l/day, which is as four times as high comparing to that of batch culture.

This research observed the morphological change and ultrastructural change of hair after having straight permanent on the twenty years old of woman`s virgin hair through the scanning electron microscopy and transmission electron microscopy. The straight permed hair was observed that the cuticular cells of cuticle layer around the surface lifted off. Also, some cuticular cells were destructed to be off. The endocuticle of cuticular cell has formed 500 nm of holes toward the longitudinal line of cuticular cell, and some of these holes were united to have the diameter of 1 μm~1.5 μm. Cortex has formed lots of holes in line with membrane of cortical cell, especially there were the holes among the limited membrane of peripheral part of melanin granules and of macrofibrils. These holes were distributed mainly around the exocortex near the cuticle layer more than the central part of cortex, where much more holes than in the cortex of virgin hair were formed.
THE SELECTIVE DELIVERY OF ANTICANCER AGENTS USING TARGET - SENSITIVE LIPOSOMES

Target-sensitive (TG-S) liposomes having modified antibodies on their surface were employed to study the release of calcein and the selective delivery of the anticancer agents, doxorubicin (DOX) and methotrexate (MTX). The release of calcein from TG-S liposome occurred when the various target cells were contacted with liposomes and it was proportionally increased with the increase of antibody affinity to the target cells. Increasing the concentration of antigen molecules (major histocompatibility, MHC) on the surface of RMA-S, the release of calcein and drugs from TG-S liposomes contacting with RMA-S also rised. The destabilization of TG-S liposomes was only induced above a threshold density of surface antigen on the target cell membrane. The growth inhibition of specific target cells by the liposomal drugs was always stronger than that of the non-specific ones. For specific target cells, the IC_(50) of liposomal DOX was about 2 times greater than that of free DOX, on the while, for non-specific target cells, more than 5 times. This indicates that the liposomal drugs were transferred preferentially to the specific target cells than the non-specific ones. Based on this phenomenon, the TG-S liposomal MTX were also applied for the selective elimination of the specific target cells in the mixed culture of specific and non-specific target cells.

N/A About one-third of radiologically indeterminate solitary pulmonary nodules (SPN) are eventually turned out to be malignant. It is very important to noninvasively determine whether the SPN is malignant or not for the decision of its way of management. PET imaging is highlighted by its unique ability of imaging the function and metabolism of cells. Glucose metabolism is increased in malignant transformed cells. We performed FDG-PET studies in patients who ha radiologi- cally indeterminate SPN and compared the findings with histologic diagnoses to assess the diagnostic accuracy in the detection of malignancy and to decide which parameter is the most suitable for clinical practice among peak SUV (pSUV), average SUV (aSUV), 50/10 ratio, and time-activity curve (TAC). Thirty patients were included in this study and the most useful parameter was pSUV. The sensitivity and specificity in the detection of malignant SPN using 3.5 as a cut off pSUV were both 87%. Interestingly, all 2 false-negative cases were bronch- ioloalveolar carcinoma on histologic examination. If these cases, which could be strongly suspected by CT findings, were excluded, the sensitivity of pSUV was 100%. In conclusion, PET imaging is very helpful for determining malignancy in indeterminate SPN and pSUV is a conveniently measurable parameter which is valuable for interpretation.
