        Reduction of Bacillus cereus Contamination in Biofilms on Stainless Steel Surfaces by Application of Sanitizers and Commercial Detergent

        We established the effectiveness of a washing, detergent and sanitizer treatments for reducing Bacillus cereus (B. cereus) contamination in biofilms on stainless steel surfaces. A water treatment did not affect biofilms and chlorine treatments at 200 ppm for 10 min and 300 ppm for 5 min, iodophor at 200 ppm for 10 min, and QAC at 400 ppm for 10 min were used to eliminate all B. cereus cells. B. cereus cells after detergent treatment were eliminated by chlorine at 100 ppm for 10 min, 200 ppm for 5 min, and 300 ppm for 3 min, and with iodophor at 100 ppm for 10 min and 200 ppm for 10 min, and QAC at 400 ppm for 10 min.

      • Visualization and Quantification of Radiochemical Purity by Cerenkov Luminescence Imaging

        <P>Determination of radiochemical purity is essential for characterization of all radioactive compounds, including clinical radiopharmaceuticals. Radio-thin layer chromatography (radio-TLC) has been used as the gold standard for measurement of radiochemical purity; however, this method has several limitations in terms of sensitivity, spatial resolution, two-dimensional scanning, and quantification accuracy. Here, we report a new analytical technique for determination of radiochemical purity based on Cerenkov luminescence imaging (CLI), whereby entire TLC plates are visualized by detection of Cerenkov radiation. Sixteen routinely used TLC plates were tested in combination with three different radioisotopes (<SUP>131</SUP>I, <SUP>124</SUP>I, and <SUP>32</SUP>P). All TLC plates doped with a fluorescent indicator showed excellent detection sensitivity with scanning times of less than 1 min. The new CLI method was superior to the traditional radio-TLC scanning method in terms of sensitivity, scanning time, spatial resolution, and two-dimensional scanning. The CLI method also showed better quantification features across a wider range of radioactivity values compared with radio-TLC and classical zonal analysis, especially for β<SUP>-</SUP>-emitters such as <SUP>131</SUP>I and <SUP>32</SUP>P.</P> [FIG OMISSION]</BR>

        Hop Extract Produces Antinociception by Acting on Opioid System in Mice

        In the present study, the antinociceptive profiles of hop extract were characterized in ICR mice. Hop extract administered orally (from 25 to 100 mg/kg) showed an antinociceptive effect in a dose-dependent manner as measured in the acetic acid-induced writhing test. Antinociceptive action of hop extract was maintained at least for 60 min. Moreover, cumulative response time of nociceptive behaviors induced with intraplantar formalin injection was reduced by hop extract treatment during the 2nd phases. Furthermore, the cumulative nociceptive response time for intrathecal injection of substance P ($0.7{\mu}g$) or glutamate ($20{\mu}g$) was diminished by hop extract. Intraperitoneal pretreatment with naloxone (an opioid receptor antagonist) attenuated antinociceptive effect induced by hop extract in the writhing test. However, methysergide (a 5-HT serotonergic receptor antagonist) or yohimbine (an ${\alpha}_2$-adrenergic receptor antagonist) did not affect antinociception induced by hop extract in the writhing test. Our results suggest that hop extract shows an antinociceptive property in various pain models. Furthermore, the antinociceptive effect of hop extract may be mediated by opioidergic receptors, but not serotonergic and ${\alpha}_2$-adrenergic receptors.

      • Antinociceptive profiles and mechanisms of orally administered coumarin in mice.

        <P>In the present study, the antinociceptive profiles of coumarin were examined in ICR mice. Coumarin administered orally (from 1 to 10 mg/kg) showed an antinociceptive effect in a dose-dependent manner as measured in the acetic acid-induced writhing test. Duration of antinociceptive action of coumarin maintained at least for 60 min. But, the cumulative response time of nociceptive behaviors induced by a subcutaneous (s.c.) formalin injection, intrathecal (i.t.) substance P (0.7 ?g) or glutamate (20 ?g) injection was not affected by coumarin. In addition, intracerebroventricular (i.c.v.) or intrathecal (i.t.) administration with coumarin (10-40 ?g) attenuated acetic acid-induced writhing response in a dose dependent manner. Intraperitoneal (i.p.) pretreatment with naloxone (an opioid receptor antagonist) attenuated antinociceptive effect induced by coumarin in the writhing test. Furthermore, i.c.v. or i.t. pretreatment with naloxone (5 ?g) reversed the decreased acetic acid-induced writhing response. However, methysergide (a 5-HT serotonergic receptor antagonist) or yohimbine (an α2-adrenergic receptor antagonist) did not affect antinociception induced by coumarin in the writhing test. Our results suggest that coumarin exerts a selective antinociceptive property in the acetic acid-induced visceral-derived pain model. Furthermore, the antinociceptive effect of coumarin may be mediated by activation of central opioid receptors, but not serotonergic and adrenergic receptors.</P>

        생물 오개념 논문의 분석을 통한 생물 오개념 편람의 제작 교육과정

        과학교육현장에서 구성주의 수업을 위해서 과학 오개념에 대한 연구 결과를 활용하는 것은 매우 중요하다. 현재까지 연구된 많은 생물 오개념 관련 연구 자료들을 활용하여 편찬한 '생물 오개념 편람'은 필요하다. 그래서 현재까지 연구된 생물 오개념 관련 연구 자료를 수집하여 연구된 학생들의 생물 오개념 자료를 발췌하였다. 그리고 발췌된 생물 오개념을 각 조사 연령별, 생물 영역별로 분류하여 분석하였다. 또한 '생물 오개념 편람'은 현장 교사들이 활용하기 때문에 '생물 오개념 편람'에 관하여 107명의 현장 교사들의 의견을 조사하였다. 연구 결과 총 764개의 생물 오개념이 발췌되었다. 764개의 오개념을 분석해 보면 저학년의 경우 동물과 식물 쪽의 생물 오개념이 많았으며 고학년으로 갈수록 유전, 진화, 생식, 발생 영역의 오개념이 증가하였다. 동물과 식물 영역은 거의 대부분이 생리 영역의 오개념이 많이 조사되었다. 학교 현장에서는 교사 중심의 수업이 많이 이루어졌고 학생들의 선개념을 조사하고 수업한다는 교사의 비율이 낮았다. 또한 현장 교사들은 '생물 오개념 편람'과 같은 자료가 수업 준비에 매우 효과적일 것이라고 하였다. 본 연구 결과를 바탕으로 수집된 764개의 생물 오개념 자료를 활용하여 '생물 오개념 편람'을 개발할 것이다. 이때는 '생물 오개념 편람'을 활용할 대상인 현장 교사들의 의견을 적극 반영해야 할 것이다. It is very valuable when many science misconceptions researched for constructivism are used at science educational field. Therefore it's necessary that development of the biological misconception handbook been able to use at science educational field by editing many researched result of biological misconceptions until now. So before producing 'Biological Misconception Handbook', research paper about biological misconception was collected and classify included biological misconception data in the research paper about biological misconception. And it's surveyed that instruction condition of science teacher and opinion of 'Biological Misconception Handbook' and 107 element, middle, high school science teachers send their opinions. There are 764 students' biology misconceptions in the research paper about biological misconception. As the result analyzed 764 students' biology misconceptions, there are many biology misconceptions about animal and plants in low grade students, there are many biology misconceptions about genetics, evolution, procreation, embryology in high grade students, At the result, of survey, the science teacher don't have constructivism education, use 'instructional experience of each teacher' or 'science instructional handbook' as method for making an investigation into students' preconception but don't use a research paper. And they predict that 'Biological Misconception Handbook' is used at planning instruction effectively. There are 'students' biological misconceptions', 'scientific concepts' in 'Biological Misconception Handbook' and they hope that it is distributed widely as book, CD, website. So 'Biological Misconception Handbook' will be developed based on 764 students' biology misconceptions in this research. And when 'Biological Misconception Handbook' is developed, opinion of biology teacher is reflected constructively.

        즉석섭취식품 도시락에서 Clostridium perfringens와 Bacillus cereus의 정량적 미생물 위해 및 관리 평가

        본 연구에서는 즉석섭취식품인 도시락에서 C. perfringens와 B. cereus의 정량적 리스크를 평가하고 안전관리 방안연구를 진행하였다. 국내 편의점에서 판매되고 있는 도시락 제품에서 C. perfringens는 검출되지 않았으나, B. cereus는 80개의 제품 중 23개 제품에서 검출되었다. 즉석섭취식품 도시락에서 C. perfringens는 포장방법에 상관없이 4°C에서 25°C 사이의 온도 범위에서 모두 사멸하였고, B. cereus는 15°C에서 사멸을, 17°C에서는 증식하였다. @RISK 프로그램을 이용한 정량적 위해평가분석 결과에 따르면 1일 1회 즉석섭취식품 도시락 섭취로 C. perfringens로 인한 식중독 발생 확률은 호기/진공 포장 각각 3.67×10<SUP>-13</SUP>, 2.15×10<SUP>-11</SUP>으로 매우 낮은 것으로 확인되어 즉석섭취식품 도시락에서 미생물학적 기준설정에 대한 제고가 필요한 것으로 판단된다. 그러나 B. cereus의 경우 본 연구에서는 설사형 B. cereus로 연구가 진행되었기 때문에 다른 유전형에 대한 연구가 추가로 필요할 것으로 사료된다. 본 연구에서는 또한 즉석섭취식품 도시락을 구입 후 B. cereus가 증식할 위험이 있는 상온에 보관한 경우를 고려하여 섭취하기까지 허용 가능한 수준(FSO)으로 줄이기 위해 전자레인지 가열을 통한 저감화 효과를 평가하였다. 섭취 전 20°C 이상의 상온에 보관 후 전자레인지 가열시간의 효과는 3분으로 가열하는 것이 2분, 2분 30초 가열하는 것보다 FSO를 만족시키는 보관시간을 연장할 수 있음을 확인하였다. 본 연구 결과는 현재 즉석섭취식품 도시락에서 C. perfringens와 B. cereus의 미생물학적 기준설정 제고와 안전관리 방안설정에 대한 과학적 근거 자료로 활용할 수 있을 것으로 사료된다. Quantitative risk assessment of Clostridium perfringens and diarrheal Bacillus cereus in RTE lunch boxes from convenience stores was performed. The efficacy of control measures using a microwave for B. cereus in RTE lunch boxes was also evaluated using the stochastic food safety objective (FSO) tool. The risk of C. perfringens and B. cereus due to consumption of RTE lunch boxes was estimated with @RISK. The results of the worst-case exposure assessment indicate that the probability of foodborne illness caused by C. perfringens was 1.22×10<SUP>-10</SUP> per one serving per day from a vacuum-packaged lunch box, while the probability of foodborne illness caused by B. cereus was zero. Storage time for lunch boxes could be extended up to 6 h at 20°C, 4 h at 25°C, and 2 h at 30°C with microwave heating for 3 min.

        The problem of selecting optimal target values for the mean of the quality characteristic of interest for a production process in which an item is sold in one of two market with different profit / cost structures or reworked. Two profit models are constructed which involve four profit / cost components: profit, production, inspection, and rework costs. Assumed that the quality characteristic of interest is normally distributed, methods of finding the most profitable process mean are presented and a numerical example is given.

        As the semiconductor wafer is required to be thinner for better compactness and higher integrity of the electronic products, mechanical dicing saw technique using the diamond blade is facing serious problems such as low cutting speed, tool wear, side wall crack and back side chipping. Recently, UV laser dicing process has emerged as an alternative methods to the blade dicing especially for the very thin wafers below 100㎛. In this study 355 nm UV laser dicing system has been developed in order to improve the productivity and reliability of ultra thin wafer dicing process. For the quality improvement of the developed dicing system various laser parameters are investigated such as power, speed and frequency of the laser beam. Moreover various measurement techniques are employed to evaluate the dicing quality using optical microscope, SEM and focused ion beam.

        영구치 치수 기질세포를 이용한 연골 분화 및 분화 시기에 따른 형태학적 변화

        Objectives: The aim was to confirm the stem cell-like properties of the dental pulp stromal cells and to evaluate the morphologic changes during in vitro chondrogenesis. Materials and Methods: Stromal cells were outgrown from the dental pulp tissue of the premolars. Surface markers were investigated and cell proliferation rate was compared to other mesenchymal stem cells. Multipotency of the pulp cells was confirmed by inducing osteogenesis, adipogenesis and chondrogenesis. The morphologic changes in the chondrogenic pellet during the 21 day of induction were evaluated under light microscope and transmission electron microscope. TUNEL assay was used to evaluate apoptosis within the chondrogenic pellets. Results: Pulp cells were CD90, 105 positive and CD31, 34 negative. They showed similar proliferation rate to other stem cells. Pulp cells differentiated to osteogenic, adipogenic and chondrogenic tissues. During chondrogenesis, 3-dimensional pellet was created with multi-layers, hypertrophic chondrocyte-like cells and cartilage-like extracellular matrix. However, cell morphology became irregular and apoptotic cells were increased after 7 day of chondrogenic induction. Conclusions: Pulp cells indicated mesenchymal stem cell-like characteristics. During the in vitro chondrogenesis, cellular activity was superior during the earlier phase (within 7 day) of differentiation.
