      • Influence of Aeration Rate and Reactor Shape on Composting Poultry Manure and Sawdust

        We investigated various physicochemical properties, i.e., temperature, ammonia and carbon dioxide concentration, pH, electrical conductivity (EC), C/N ratio, organic matter content, moisture content, bacterial population, and seed germination index (GI), during the composting of poultry manure and sawdust for different aeration rates and reactor shapes. Three cylindrical-shaped and three rectangular-shaped laboratory-scale 60-L composting reactors were used in this study, with aeration rates of 0.3, 0.6, and 0.9 L min<sup>-1</sup>kg<sup>-1</sup> (drymatter)DM. Results showed that the low aeration rate (0.3 L min<sup>-1</sup>kg<sup>-1</sup>DM) corresponded to a higher and longer thermophilic phase than did the high aeration rate (0.9 L min<sup>-1</sup>kg<sup>-1</sup>DM). Ammonia and carbon dioxide volatilization were directly related to the temperature profile of the substrate, with significant differences between the low and high aeration rates during weeks 2 and 3 of composting but no significant difference observed during week 1. At the end of our study, the final values of pH, EC, moisture content, C/N ratio, and organic matter in all compost reactors were lower than those at the start. The growth of mesophilic and thermophilic bacteria was directly related to the thermophilic and mesophilic condition of the compost. The final GI of the cylindrical reactor with an airflow rate of 0.3 L min<sup>-1</sup>kg<sup>-1</sup>DM was 82.3%, whereas the GIs of the other compost reactors were below 80%. In this study, compost of a cylindrical reactor with a low aeration rate (0.3 L min<sup>-1</sup>kg<sup>-1</sup>DM) was more stable and mature than the other reactors.

        The purpose of this study was to evaluate effects of Cordyceps militaris on fat oxidation during submaximal exercise. Fifteen healthy male collegiate students(ingestion group, N=7; control group, N=8) participated as subjects after signing an informed consent following overnight, subjects completed maximal graded execise test on treadmill to determine exercise intensity(50%VO2max). To evaluate the fat oxidation using indirect calorimeter, it was performed submaximal treadmill to determine exercise lasting. 40min at 50%VO2max after 5day and consumed Cordyceps militaris on separate days. This study was used 2×4 two way repeated ANOVA to analyze physiological and metabolic variables between two groups and metabolic variables between two groups and four exercise periods(l0min, 20min, 30min 40min), and used Scheffe to post-hoc test. Significance was set at α=.05. The results of this study were as followed: First, there were significantly highest in ingestion group for V02, percentage of fat oxidetion(%fat), fat per kilocarolies(Fkcal), and total per kilocarolies(Tkcal)(p<.05), but respiratory exchange ration(RER) and heart rete(HR) were significantly lower in ingestion group(p<.05). Second, RER, %fat, Fkcal were significantly difference in 10, 20, 30, 40min of exercise period(p<.05). This results suggest that there is the effect of Cordyceps militaris on fat oxidation submaximal exercise. Additionally, to evaluate effect of Cordyceps militaris as ergogenic aid, it should be required the future study that included extend subjects, various dosage, and chronic consumption.

      • A study on the removal of sulfate and nitrate from the wet scrubber wastewater using electrocoagulation

        <P>Wet scrubber is widely used to remove pollutants from a furnace flue gas or from other gas streams. However, high cost for the treatment or exchange of contaminated washed water is one of the problems. Electrocoagulation (EC) is an electrochemical method for treating polluted water, whereby sacrificial anodes dissolve to produce active coagulant precursors in the solution. This technology can be used for the removal of color, anionic contaminants, and colloidal particles. The goal of the present study was evaluation of EC process for treatment of wet scrubber wastewater and optimization of the process. In this study, the influence of electrode material and electrode distance on removal efficiency (%) of nitrate, sulfate, and total suspended solids (TSS) was investigated with synthetic wastewater. Using an Al electrode, 60.6% of nitrate, 50.0% of sulfate, and 96.8% of TSS were removed by EC treatment within 30 min. In case of Fe electrode, 69.7% of nitrate, 75.0% of sulfate, and 98.8% of TSS were removed. The treatment efficiency for real wet scrubber wastewater was evaluated with Fe electrode. After 180 min of EC treatment, 50% of color, 40% of nitrate, 40% of sulfate, and 95% of TSS were removed, respectively. All these results give clear evidence that EC process can effectively reduce the TSS, nitrate, and sulfate ions from wet scrubber wastewater.</P>

        ATM-dependent chromatin remodeler Rsf-1 facilitates DNA damage checkpoints and homologous recombination repair.

        <P>As a member of imitation switch (ISWI) family in ATP-dependent chromatin remodeling factors, RSF complex consists of SNF2h ATPase and Rsf-1. Although it has been reported that SNF2h ATPase is recruited to DNA damage sites (DSBs) in a poly(ADP-ribosyl) polymerase 1 (PARP1)-dependent manner in DNA damage response (DDR), the function of Rsf-1 is still elusive. Here we show that Rsf-1 is recruited to DSBs confirmed by various cellular analyses. Moreover, the initial recruitment of Rsf-1 and SNF2h to DSBs shows faster kinetics than that of γH2AX after micro-irradiation. Signals of Rsf-1 and SNF2h are retained over 30 min after micro-irradiation, whereas γH2AX signals are gradually reduced at 10 min. In addition, Rsf-1 is accumulated at DSBs in ATM-dependent manner, and the putative pSQ motifs of Rsf-1 by ATM are required for its accumulation at DSBs. Furtheremore, depletion of Rsf-1 attenuates the activation of DNA damage checkpoint signals and cell survival upon DNA damage. Finally, we demonstrate that Rsf-1 promotes homologous recombination repair (HRR) by recruiting resection factors RPA32 and Rad51. Thus, these findings reveal a new function of chromatin remodeler Rsf-1 as a guard in DNA damage checkpoints and homologous recombination repair.</P>

        Purification and Characterization of Helicobacter pylori ${\gamma}$-Glutamyltranspeptidase

        Gamma-glutamyltranspeptidase (GGT) was purified to electrophoretic homogeneity from the cell extract of H. pylori. The purified enzyme consisted of heavy and light subunits with molecular weights of 38 kDa and 21 kDa, respectively. N-terminal amino acid sequence of heavy and light subunits revealed that H. pylori GGT was processed into 3 parts for a signal peptide of 27 amino acid residues, a heavy subunit of 352 residues, and a light subunit of 188 residues during translation. The reaction rate for hydrolysis of ${\gamma}$-GpNA was 84.4 ${\mu}mol/min$ per milligram of protein, and that for the ${\gamma}$-glutamyl transfer from ${\gamma}$-GpNA to gly-gly was 23.8 ${\mu}mol/min$ per milligram of protein. The apparent Km values of H. pylori GGT for ${\gamma}$-glutamyl compounds were on the order of $10^{-3}$ to $10^{-4}$ M and those for acceptor peptides and amino acids were on the order of $10^{-1}$ to $10^{-2}$ M. The GGT protein kept approximately 80% of the initial enzymatic activity on incubation at $60^{\circ}C$ for 15 min. The optimum temperature and pH for reactions of both hydrolysis and transpeptidation were $40^{\circ}C$ and 9.0, respectively. The transpeptidation and hydrolysis reactions catalyzed by H. pylori GGT were strongly inhibited by L-Gln and moderately inhibited by L-Ala, L-Ser, ${\beta}$-chloro-L-Ala, and L-Glu. These results demonstrated that the biochemical properties of H. pylori GGT are different from those of other bacterial GGTs. Further, H. pylori GGT might degrade glutathione in the gastric mucous layer of humans if the enzyme could be secreted in the bacterial niches.

        Effects of Ethanol Addition on the Efficiency of Subcritical Water Extraction of Proteins and Amino Acids from Porcine Placenta

        In a previous study, hydrolysates of porcine placenta were obtained and the extraction efficiency for proteins and amino acids was compared between sub- and super-critical water extraction systems; optimum efficiency was found to be achieved using subcritical water ($170^{\circ}C$, 10 bar). In this study, the effects of adding ethanol to the subcritical water system were investigated. The lowest-molecular-weight extraction product detected weighed 434 Da, and the efficiency of extraction for low-molecular-weight products was increased when either the concentration of ethanol was decreased, or the extraction time was lengthened from 10 min to 30 min. The highest concentration of free amino acids (approximately 8 mM) was observed following 30 min extraction using pure distilled water. The concentration of free amino acids was significantly lower when ethanol was added or a shorter extraction time was used (p<0.05). Color change of the solution following extraction was measured. There were no significant differences in color between lysates produced with different extraction times when using distilled water (p>0.05); however, using different extraction times produced significant differences in color when using 20% or 50% ethanol solution for subcritical extraction (p<0.05). The range of pH for the hydrolysate solutions was 6.4-7.5. In conclusion, the investigated extraction system was successful in the extraction of $\leq$ 500 Da hydrolysates from porcine placenta, but addition of ethanol did not yield higher production of low-molecular-weight hydrolysates than that achieved by DW alone.

        도심 수목원에서의 경관감상과 산책이 인체의 생리적 안정에 미치는 영향

        This research was conducted to investigate the impact of viewing scenery and walking in the urban forest onphysiological relaxation of humans. The experiment was conducted in Hanbat Arboretum located in Daejeon, and thecontrol experiment was conducted in front of Daejeon City Hall. The subjects that participated in the experimentcomprised 24 Korean male university students in their 20s (average age, 21.1±2.5 years), participated in the experimentas the subject, and these subjects were classified into three groups divided into three locations such as the pine forest inArboretum, the pond in Arboretum, and the city. The subjects sat down and viewed scenery for 10 min, and they thenwalked for 15 min. Further, physiological changes were measured using indicators such as heart rate variability (HRV),blood pressure, and pulse rate. As a result, when subjects viewed scenery and walked in the urban forest, a statisticallysignificant increase in the high frequency (HF) power of HRV and decrease in systolic pressure was observed comparedwith when subjects viewed scenery and walked in the city. Through this research, it was found that activities like viewingscenery or walking in Arboretum of the city are effective in increasing the physiological relaxation of the city residents.

        원인 불명의 실신 환자의 진단에 있어서 Head - up tilt Test 의 유용성과 혈관미주신경성 실신 환자의 임상적 특징

        N/A Background: Syncope, defined as a transient loss of consciousness, is a frequently encountered symptom, but despite thorough clinical and invasive diagnostic investigation, the cause of syncope remains unexplained. In recent years, head-up tilt test has been of increasing interest as a diagnostic aids in patients with unexplained syncope. We studied to define the usefulness of various diagnostic test in the evaluation of patients with recurrent syncope, to assess the usefulness of head-up tilt test in the working of syncope of unknown origin and to evaluate the clinical characteristics of subgroups of patients with syncope induced by head-up tilt test. Methods: Between June, 1991 and November, 1992 at St. Mary`s hospital, 21 patients with history of syncope and 24 control subjects without history of syncope underwent 60° head-up tild test for 60 min. Results: 1) The definite cause for recurrent syncope were diagnosed in 10 of patients (47,6%) after clinical and invasive tests. 2) During head-up tilt test, vasovagal responses were provoked in 5 of 21 patients (23.8%) with recurrent syncope and 1 of 24 patients (4.2%) without syncope (p<0,05). 3) The head-up tilt test induced symptomatic bradycardia or hypotension in 3 of 10 patients (30%.) with negative electrophysiologic results. 4) During tilt induced vasovagal response, a) mean heart rate decreased to 42±24 beat/min from supine control of 70±12 beat/min (p<0. 05), b) mean systolic blood pressure decreased to 95±12 mmHg from supine control of 120±S mmHg (p<0,05), c) mean diastolic blood pressure decreased to 60±21mmHg from supine control of 72±13 mmHg (p<0.05), d) the time interval to onset of vasovagal response was 22±12 min. Conclusions: 1) In a large proportion of patients with recurrent syncope, the diagnosis remains unexplained despite a neurologic and cardiologic investigation including an electrophysiologic study. 2) Vasovagal responses are frequent cause in patients with recurrent unexplained syncope. 3) The head-up tilt test may be a simple, safe and highly yielding provocative test in the investigation of syncope of unknown origin.

      • 무산소성 운동이 혈청 지질 농도 및 면역세포 반응에 미치는 영향

        The purpose of this study was to determine serum lipids and immune responses on the anaerobic exercise. Serum total cholesterol, HDL-C, LDL-C and plasma triglyceride, percentages of WBC and their subsets, natural killer cells were evaluated between before and after the anaerobic power. All subjects were high trained male athletes (19-24 years of age). Results were expressed as mean and standard deviations. One-way ANOVA repeated measure was used to compare the all parameters between before and after the anaerobic power test. All significant differences were set at α=.05. In this study, comparison of anaerobic power(Nm/%BW) was higher in soccer players than ssirum and judo players (p<.05). The percentage of WBC, monocyte, eosinophil and basophil were no significant differences between before and after anaerobic exercise. There were increased on neutrophil, lymphocyte after anaerobic exercise in three groups. (p<.01, p<.001, respectively). Also, there were significant differences on T cell, B cell, NK cell between before and after anaerobic exercise (p<.001).
