Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

Hypoxia-Responsive MicroRNA-101 Promotes Angiogenesis <i>via</i> Heme Oxygenase-1/Vascular Endothelial Growth Factor Axis by Targeting Cullin 3

<P>Aims: Hypoxia induces expression of various genes and microRNAs (miRs) that regulate angiogenesis and vascular function. In this study, we investigated a new functional role of new hypoxia-responsive miR-101 in angiogenesis and its underlying mechanism for regulating heme oxygenase-1 (HO-1) and vascular endothelial growth factor (VEGF) expression. Results: We found that hypoxia induced miR-101, which binds to the 3 ' untranslated region of cullin 3 (Cul3) and stabilizes nuclear factor erythroid-derived 2-related factor 2 (Nrf2) via inhibition of the proteasomal degradation pathway. miR-101 overexpression promoted Nrf2 nuclear accumulation, which was accompanied with increases in HO-1 induction, VEGF expression, and endothelial nitric oxide synthase (eNOS)-derived nitric oxide (NO) production. The elevated NO-induced S-nitrosylation of Kelch-like ECH-associated protein 1 and subsequent induction of Nrf2-dependent HO-1 lead to further elevation of VEGF production via a positive feedback loop between the Nrf2/HO-1 and VEGF/eNOS axes. Moreover, miR-101 promoted angiogenic signals and angiogenesis both in vitro and in vivo, and these events were attenuated by inhibiting the biological activity of HO-1, VEGF, or eNOS. Moreover, these effects were also observed in aortic rings from HO-1(+/-) and eNOS(-/-) mice. Local overexpression of miR-101 improved therapeutic angiogenesis and perfusion recovery in the ischemic mouse hindlimb, whereas antagomiR-101 diminished regional blood flow. Innovation: Hypoxia-responsive miR-101 stimulates angiogenesis by activating the HO-1/VEGF/eNOS axis via Cul3 targeting. Thus, miR-101 is a novel angiomir. Conclusion: Our results provide new mechanistic insights into a functional role of miR-101 as a potential therapeutic target in angiogenesis and vascular remodeling. Antioxid. An Ac-type Plasma Display Panel (PDP) operating with Ne-Ar(0.1%) Penning mixture gas is fabicated. The characterics of the panel with electrodes covered with thin and thick dielectric layers are studied. The brightness of the Neon-orange light emitted by the panel measured as function of applied voltage and frequency. As an application, a graphic display system equipped with PDP showing still and moving pictures is made.

NGS_SNPAnalyzer: a desktop software supporting genome projects by identifying and visualizing sequence variations from next-generation sequencing data

Background Sequence variations such as single nucleotide polymorphisms are markers for genetic diseases and breeding. Therefore, identifying sequence variations is one of the main objectives of several genome projects. Although most genomeproject consortiums provide standard operation procedures for sequence variation detection methods, there may be differencesin the results because of human selection or error. Objective To standardize the procedure for sequence variation detection and help researchers who are not formally trainedin bioinformatics, we developed the NGS_SNPAnalyzer, a desktop software and fully automated graphical pipeline. Methods The NGS_SNPAnalyzer is implemented using JavaFX (version 1.8); therefore, it is not limited to any operatingsystem (OS). The tools employed in the NGS_SNPAnalyzer were compiled on Microsoft Windows (version 7, 10) andUbuntu Linux (version 16.04, 17.0.4). Results The NGS_SNPAnalyzer not only includes the functionalities for variant calling and annotation but also providesquality control, mapping, and filtering details to support all procedures from next-generation sequencing (NGS) data to variantvisualization. It can be executed using pre-set pipelines and options and customized via user-specified options. Additionally,the NGS_SNPAnalyzer provides a user-friendly graphical interface and can be installed on any OS that supports JAVA. Conclusions Although there are several pipelines and visualization tools available for NGS data analysis, we developedthe NGS_SNPAnalyzer to provide the user with an easy-to-use interface. The benchmark test results indicate that theNGS_SNPAnayzer achieves better performance than other open source tools.

Agrobacterium tumefaciens를 이용한 bialaphos 저항성 형질전환 벼의 개발

비선택성 제초제인 bialaphos는 고등식물에 있어서 glutamine 합성을 억제하여 식물체를 고사시킨다. Acetyltransferase에 의해 encoding된 bialaphos 저항성 유전자는 세균 Streptomyces hygroscopicus SF1239로부터 cloning된 것을 사용하였다. Bialaphose 저항성 유전자를 Agrobacterium 감염법을 이용하여 국내에서 재배되는 벼(동진)에 도입한 결과 약 30%정도의 형질전환 식물체를 얻을 수 있었다. T_1세대의 17개체는 hygromycin과 bialaphos에 대한 저항성 유전형질이 3 : 1로 분리되었다. 또한 Southern 분석을 실시한 결과 wild type의 식물체에서는 Bar 유전자의 검출을 볼 수 없었으나 형질전환 식물체의 경우 Bar 유전자의 검출이 가능하였다. T_3 세대의 형질전환 식물체와 wild type의 식물체를 포장상태에서 비선택성 제초제인 바스타를 살포하고 3주 후에 관찰한 결과 형질전환 식물체는 외형적으로 아무런 피해를 받지 않고 정상적으로 생장하였으나, wild type의 식물체와 잡초는 모두 고사하였다. 이상의 결과를 종합해보면 hygromycin과 bialaphos 저항성 유전자는 Agrobacterium 감염법을 이용하여 단자엽 식물인 벼에 도입할 수 있다는 것을 보여준 것이며, 또한 bialaphos 저항성 유전자가 식물에 도입됨으로써 비선택성 제초제에 대한 저항성 식물을 개발 할 수 있다는 것을 보여 주었다. The bialaphos is a potent inhibitor of glutamine synthease in higher plants and is used as a non-selective herbicide. We have used the bialaphos resistant gene (Bar) encoding for an acetyltransferase isloated from Streptomyces hygroscopicus SF1293. Callus derived from mature seeds of rice (Oryza sativa L. cv. Dong Jin) were co-cultivated with Agrobacterium tumefacieus EHA101 carring a plasmid pGPTV-HB containing genes for hygromycin resistance (HygR) and Bar. Transgenic plants showing in vitro resistance to 50 mg/L hygromycin and 10 mg/L bialaphos were obtained by using a two-step selection/regeneration procedure. Transformation efficiency of rice was about 30% which was as high as reported in other dicotyledons. Progenies (T_1 generation) derived from primary transformant of 17 lines were segreagated with a 3 resistant:1 sensitive ratio in medium containing hygromycin and bialaphos. Stable integration of Bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from T_2 progenies. Transgenic plants (T_3) grown in the field were resistant to bialaphos (Basta) at a dosage lethal to wild type plants.

In recent years the amount of sequencing has increased tremendously, especially due to the introduction of automated DNA sequencing. PCR is principal method for the synthesis of templates for DNA sequencing. Consequently, the need for PCR cleanup for template preparation has increased. Already, many PCR purification kits are marketed. This DNA purification kits uses most glass membrane or silica gel, So, can do rapidly efficient purification by simple miniprep. But, efficiency of purification kits is different according to manufacturer and is very expensive. Therefore, we compare the homemade Diatomaceous earth column with major manufacturer's spin column methods for DNA purification. As a result, we found that the homemade Diatomaceous earth column method resulted in equal or better yields relative to the commercial purification kits.

Validation of the oxford classification of iga nephropathy: A Single-Center Study in Korean Adults

Background/Aims: The recently published Oxford classification of IgA nephropathy (IgAN) proposed a split system for histological grading, based on prognostic pathological features. This new classification system must be validated in a variety of cohorts. We investigated whether these pathological features were applicable to an adult Korean population. Methods: In total, 69 adult Korean patients with IgAN were analyzed using the Oxford classification system at Soonchunhyang University Hospital, Seoul, Korea. All cases were categorized according to Lee`s classification. Renal biopsies from all patients were scored by a pathologist who was blinded to the clinical data for pathological variables. Inclusion criteria were age greater than 18 years and at least 36 months of follow-up. We excluded cases with secondary IgAN, diabetic nephropathy combined other glomerulopathies, less than 36 months of follow-up, and those that progressed rapidly. Results: The median age of the patients was 34 years (range, 27 to 45). Mean arterial blood pressure was 97 ± 10 mmHg at the time of biopsy. The median follow-up period was 85 months (range, 60 to 114). Kaplan-Meier analysis showed significant prognostic predictions for M, E, and T lesions. A Cox proportional hazard regression analysis also revealed prognostic predictions for E and T lesions. Conclusions: Using the Oxford classification in IgAN, E, and T lesions predicted renal outcome in Korean adults after taking clinical variables into account.

기종성 신우신염은 주로 당뇨병환자나 폐쇄성 요로질환을 가진 환자에 발생하며 특징적으로 신실질 및 그 주위조직에 가스를 형성하면서 심한 조직괴사를 일으키는 매우 드문 급성 화농성 신감염으로 사망률이 높은 질환이다. 이 질환은 1989년 Kelly와 MacCallum에 의해 처음 보고된 이래 외국문헌에 약 90례 정도가 보고되어 있으며 국내에는 28례가 보고되어 있다. 수액 및 전해질 보충, 인슐린 투여를 통한 적절한 혈당조절 및 항생제 투여등의 내과적 치료와 절개배농 및 신적출술등의 수술적 치료 방법이 있다. 내과적 치료로 가스의 감소가 없는 경우에는 즉각적인 수술적 치료를 시행하여야하므로 이 가스변화에 대한 추적관찰이 중요하다 하겠다. 본 저자들은 당뇨병환자에서 발생한 기종성 신우신염 1례를 내과적 요법을 치험하였기에 문헌고찰과 함께 보고하고자 한다. Emphysematous pyelonephritis is rare, life-threatening infection of the renal parenchyma and perirenal tisseue. This disease is characterized by the production of intrarenal and perirenal gas and is frequently encountered in patients with diabetes mellitus or urinary obstruction. We experienced a case of emphysematouse pyelonephritis in a 62 years old women with poorly controlled diabetes mellitus who had been managed with medical theraphy. So we report this case with a review of the referenced literatures.

Shelf Life Determination of Precooked Frozen Meat Dumplings

Quality of frozen foods is significantly affected by the storage temperature and tie, and the consumers' demand for safe quality frozen foods is increasing. The objective of this research was to estimate the shelf life of frozen cooked meat dumplings, based on the changes in the selected physico-chemical and sensory quality attributes. Precooked frozen meat dumplings sampled from the same lot produced at a frozen food company in Seoul were stored at -10, -15, and -20℃. The meat dumplings sampled at 3- to 4-week intervals were subjected to the determination of volatile basic nitrogen (VBN), thiobarbituric acid (TBA), peroxide value (POV), surface color and shear value after thawing at 5℃ for 15 h. High quality life (HQL) was determined through the triangle test and practical quality life (PQL) through the scoring test. VBN, TBA and POV increased as the storage period was increased. HQL of the frozen meat dumplings were 189 and 252 days at -10 and -15℃, respectively. The estimated PQL were 349, 475, and 491 days at -10, -15, and -20 ℃, respectively. Statistical analysis indicated that there were significant relationships between sensory scores and VBN and POV of the stored meat dumplings. VBN was selected as the most reliable objective quality index for the shelf life determination (PQL) of the precooked frozen meat dumplings.
