      • Highly ordered and disposable nanopillar array for detection of beta-amyloid

        The sensitive evaluation of Alzheimer's disease (AD) is important for monitoring of progression of disease. This study established the highly ordered and flexible nano-pillar electrode (NPE) for detecting the beta-amyloid. The NPE was manipulated by soft/photolithography principle. The polyurethane (PU) was firstly fabricated based on nanohole-patterned mask. Then, the NPE surface was modified with Ti/Au with 20/200 nm. The nanopillar has the 1500 nm height and 500 nm diameter with 1:3 aspect ratio. Based on the NPE, the self-assembled monolayer (SAM) was modified to densely immobilize the beta-amyloid antibodies. Sequentially, the immunoassay was implemented with the 0 - 1.0 ng/mL of beta-amyloid. The horseradish peroxidase (HRP)-based electrochemical analysis was applied, and the beta-amyloid was quantified by using the square wave voltammetry (SWV). The exhibited test result of limit of detection was 0.14 ng/mL with 10% reproducibility for beta-amyloid in artificial tear sample in whole detection range. Based on the reliable test result, the developed biosensing principle with NPE sensor shows the sensitive electrochemical behavior for real sample-based beta-amyloid, proving possibilities for sensitive diagnosis in medical and commercial field as a point-of-care testing (POCT) technologies.
