      • 한라산 자연샘물 및 야생노루의 분변에서 분리한 Yersinia enterocolitica의 특성 연구

        한라산 등산로 주변 자연샘물 6개소 Yersinia enterocolitica 분포와 자연샘물 및 야생노루의 분변에서 분리한 Y. enterocolitica에 대한 생화학적 성상, 생물형, 혈청형, 시험관내 병원성시험, 항생제 감수성 시험 결과는 다음과 같다. 자연샘물에서 Y. enterocolitica의 분포는 영실샘, 성판악샘, 어리목샘, 사재비샘, 노루샘, 오름샘의 순으로 각각 8-49, 8-33, >2-8, >2-4, >2-4, >2-2MPN/L를 보였으며, 등산로 중간(사재비샘, 노루샘, 오름샘)보다 고도가 낮은 등산로 입구(영실샘, 성판악샘, 어리목샘)에 위치한 샘물에서 높은 분포를 보였다. 분리된 Y. enterocolitica 29주에 대한 생물형의 분포는 1, 2, 3, 3A, 3B, 4, 5형의 7가지 생물형 중 1형(24/29, 82,8%) 및 2형(5.29, 17.2%)으로 나타났고, 지금까지 알려진 57개 혈청형 중 O:1·2혼합, O:3, O:5, O:8, O:9의 혈청형에 대한 시험결과는 O:1·2혼합형, O:5형 그리고 O:8형이 자연샘물에서 분리한 균주에서만 각각 2주, 4주, 2주의 분포를 나타냈으며 대다수인 72.4%(21/29)는 5개 혈청에 응집반응을 보이지 않았다. 시험관내 병원성 시험에서 2/29(6.9%)만이 자가응집 시험에 양성을 보였을 뿐 CRMOX 시험에서는 음성 반응을 보였다. 항생제 12종에 대한 감수성 시험 결과는 ampicillin, cephalothin, penicillin에서만 내성을 보였다. Yersinosis is known as an infectious disease caused by Yersinia pseudotuberculosis, or Yersinia. enterocolitica. Some strains of Yersinia enterocolitica are pathogenic to human. This study was conducted to examine the characteristics of Yersinia enterocolitica strains isolated from spring water at six points near the climbing routes of Mt. Halla as well as from the feces of roe deer. Data were obtained in several approaches such as biochemical characterization, biotypes, serotypes, virulence and antibiotic test for the isolates, and MPN (most probable number/L) techniques. Most probable numbers (MPN/L) of Y. enterocolitica in springwater were ranging from 8 to 49 in Yeongsil-spring, 8 to 33 in Seongpanak-spring, >2 to 8 in Eorimok-spring, >2 to 4 in Sajaebi- spring, Noru-spring, and >2 in Oreum-spring. This suggested that Y. enterocolitica strains might be distributed much more in spring waters in the lower altitude than those in the higher altitude. Twenty nine strains of Y. enterocolitica from spring water and feces of roe deer were selected for several tests. Twenty four strains (82.8%) were found to belong to biotype 1, and the rest (17.2%) were biotype 2. Autoglutination positive was observed in two strains among them (6.9%). In analysis of serotypes of O:l·2 mixed form, O:3 , O:5, O:8, and O:9 for the isolates, O:1·2 mixed type and O:8 type were 2 strains (6.9%) each, and O:5 type was 4 strains (13.8 %), while the other 21 strains (72.4%) did not show any agglutination of 5 serotypes. Antibiotic test for randomly selected strains of Y. enterocolitica showed that those were resistant to ampicillin, cephalothin, and penicillin.

        식품에서 분리한 Yersinia enterocolitica의 특성조사

        국내 식육중 Y. enterocolitica의 분포 조사 및 분리균의 특성을 조사하기 위하여 4월부터 11월사이에 상, 하반기로 나누어 소고기, 돼지고기, 닭고기 463건을 서울, 대전, 광주, 부산에서 구입하여 실험하였다. 전체 식육중 17.5% (81건)에서 Y. enterocolitica가 분리되었으며 닭고기(26.0%)가 가장 많이 오염되어 있었다. 분리율은 계절에 따라 차이를 보였는데 하반기에 수거한 돼지고기와 닭고기에서 상반기보다 2배이상 높은 균 분리율을 보였다. 항생제 감수성 실험결과 대부분의 분리균은 carbenicillin, ampicillin, erythromycin, penicillin, bacitracin에 대해 내성을 나타내는 반면 polymyxin B, kanamycin, trimethoprim/sulfamethoxazole, ciprofloxacin, tetracycline, nalidixic acid, gentamicin등에 감수성을 나타내었다. 분리된 Y. enterocolitica의 혈청형은 serotype O:5가 주종이었고 일본 및 유럽 등지에서 많이 분리되어지는 O:9는 분리되지 않았다. PCR 실험결과 81개의 분리균주중 8균주(10%)가 병원성을 갖고있는 것으로 밝혀졌는데 그 중 6균주가 돼지고기에서 발견되었다. 그리고 돼지고기 분리균주중 병원성이 없는 것으로 널리 알려져 있는 serotype O:5가 PCR 양성반응으로 병원성을 보여 이전 연구들과 상반된 결과를 나타낸 것에 대해서는 더 연구해야할 과제로 남아 있다. The incidence of Yersinia enterocolitica in raw meat was determined over 10 month period. Y. enterocolitica was isolated from 8.5% of beef, 17.0% of pork and 25.6% of chicken. Overall prevalence of Y. enterocolitica in raw meat was 17.5%. Seasonal difference was observed in isolation rate in which pork and chicken samples collected in the second half of the year twice was more than that of the first half of the year. Serotypes of Y. enterocolitica isolates were O:5 (17.3%), O:8 (8.6%), O:3 (6.2%), O:1,2 (1.2%), and others. The antibiotics susceptibility tests showed Y. enterocolitica isolates were resistant to carbenicillin, ampicillin, erythromycin, penicillin and bacitracin. In contrast it showed sensitivity to polymyxin B, kanamycin, ciprofloxacin, gentamicin, trimethoprim/sulfamethoxazole, nalidixic acid, and tetracycline. PCR with specific primers derived from ail gene of Y. enterocolitica was applied to detect and confirm pathogenic Y. enterocolitica. About 10% of the isolated Y. enterocolitica proved to be pathogenic and most of them were found in pork. However, proper cooking and meat process can kill and remove all Y. enterocolitica in meat, because the organism is very sensitive to heat.

        국내 사육돼지에서의 Yersinia enterocolitica 38 kDa outer membrane protein에 대한 항체가 분포

        Yersinia enterocolitica is an inhabitant in the lower intestinal tract of many domestic and wild animals as well as in the nature. Of the several forms of diseases caused by Y. enterocolitica, an acute enteritis, especially in young children, is the most common form. Infection of the bacteria usually occurs through fecal-oral route by contaminated foods or water, especially mountainspring water. Of the domestic animals, swine has been known as one of the most important carrier in the human infection. Based on the knowledge, prevalence of antibody against Y enterocolitica was investigated with swine sera collected from Korea for the survey of Y enterocolitica infection in swine. As the first step of this survey, we analyzed outer membrane protein (OMP) profiles of the representative strains of Y enterocolitica isolated from the feces of piglets and mountainspring water in Korea. Thirty-eight kDa OMP was identified as the common OMP regardless of origin, serotype, or biotype of Y enterocolitica isolates. Presence of antibody specific for 38 kDa OMP of Y enterocolitica in 1,076 swine sera collected from November 1999 to October 2000 was analysed with ELISA. Antibody titer in sows was significantly higher than that in piglets, growing pigs and finishing pigs (p<0.05). Also, there was seasonal difference in the prevalence of antibody against Y enterocolitica. These results would provide the basic knowledge for controlling the Y enterocolitica infection in human as well as swine.

      • Identification of <i>Yersinia enterocolitica</i> using a random genomic DNA microarray chip

        <P>Abstract</P><P>Aims: </P><P>To fabricate a DNA chip containing random fragments of genomic DNA of <I>Yersinia enterocolitica</I> and to verify its diagnostic ability.</P><P>Methods and Results: </P><P>A DNA microarray chip was fabricated using randomly fragmented DNA of <I>Y. enterocolitica</I>. Chips were hybridized with genomic DNA extracted from other <I>Y. enterocolitica</I> strains, other <I>Yersinia</I> spp. and bacteria in different genera. Genomic DNA extracted from <I>Y. enterocolitica</I> showed a significantly higher hybridization rate compared with DNA of other <I>Yersinia</I> spp. or bacterial genera, thereby distinguishing it from other bacteria.</P><P>Conclusions: </P><P>A DNA chip containing randomly fragmented genomic DNA from <I>Y. enterocolitica</I> can detect <I>Y. enterocolitica</I> and clearly distinguish it from other <I>Yersinia</I> spp. and bacteria in different genera.</P><P>Significance and Impact of the Study: </P><P>A microarray chip containing randomly fragmented genomic DNA of <I>Y. enterocolitica</I> was fabricated without sequence information, and its diagnostic ability to identify <I>Y. enterocolitica</I> was verified.</P>

        약수터수로부터 분리한 Yersinia enterocolitica의 성장특성 및 Plasmid 유형

        The studies were conducted to explore the dffects of growth or survival against various factors and plasmid profiles of 49 Y. enterocolitica isolated from springs water. In the presence of calcium hypochlorite, y. enterocolitica was entirely extinguished by exposure for 33 hours at 0.8 ppm concentration, and was grown up to 7% NaCl, but not at 95 NaCI. Y. enterocolitica was presented optimal growth at pH 7.0 anad 9.0, and not allowth the growth at pH3.0, 5.0 and 11.0. The optimal temperature for growth of Y. enterocolitica was 25$\circ$C and 35$\circ$C, and allowed the growth at refrigerant temperature, 5$\circ$C. Y. enterocolitica was remarkably decreased by exposure for 30 seconds under UV light, and entirely extinguished by exposure for 90 seconds. Therefore, UV light was effective for sterilization of Y. enterocolitica. Fourty-nine strains of Y. enterocolitica were harbor plasmid DNA of approximately 46 Kb molecular weight.

        부산지역 약수터수로부터 Yersinia enterocolitica의 분리에 관한 연구

        On the purpose pf epidemiological study related to yersiniosis, a total of 720 aprings water collected from 60 points in the Pusan area were examined for the presence of Y. enterocolitica and also the isolation rates, biotype, serotype, biochemical properties and antibiotic susceptibility. Fifty-eight(8.0%) strains of Yersinia species were isolated from 720springs water. Isolation rate for each species was 49 (6.8%)strains of Y. enterocolitica, 3 (0.4%) strains of Y. pseudotuberculosis and 6 (0.8%) strains of y. frederiksenii. Seasonal distribution of isolated Yersinia sp. were shown considerably from November to April, and Y. enterocolitica was especoally isolated in order of January (20.4%), December (16.3%), March(14.3%) and April(8.2%). Isolated T. enterocolitica was divided into 4 kinds of biotype such as 1, 2, 3, and 3B. Distribution of each biotype was shown in order of biotype 1 (51.0%), biotype 2 (30.6%), biotype 3B (16.3%) and biotype 3 (2.1%). The serotypes pf 49 Y. enterocoliticawere typed 7 kinds of werotypes (0 ; 3, 0 : 5, 0 : 9, 0 : 13, 0 : 18 and 0 : 21), and serotype 0 : 8(34.7%). 0 : 9(30.6%) and 0 : 3(10.2%) were encountered most frequently.

        GC/MS을 이용한 Yersinia enterocolitica 균체 및 배지의 대사체 분석

        배양시간(6, 12, 24, 48시간)에 따른 Yersinia enterocolitica 균체 및 배지 대사체들의 변화를 이해하기 위하여 GC/MS와 다변량통계분석을 이용하여 분석한 결과 Partial Least Squares-Discriminant Analysis(PLS-DA) scores plots 상에서 배양시간에 따라 분석 시료들이 뚜렷하게 분리되었다. 시료들 사이의 차이에 관여하는 대사물질들을 동정한 결과 균체에서는 ethylene glycol, valine, ethanolamine, succinic acid, adenine, stearic acid, 배지에서는 glycine, fumaric acid, threonine, aminomalonic acid, malic acid, glutamic acid, citric acid가 배양시간에 따른 차이가 나는 것으로 확인되었다. 이들 대사물질들을 이용하여 관련 대사경로를 도출한 결과 배양시기에 따라 에너지 생성에 관여하는 대사경로가 주로 변하는 것으로 관찰되었다. Y. enterocolitica의 경우 대수기 초기에는 TCA cycle을 통해 에너지를 공급하다가 정지기에 들어서면서 정상적인 TCA cycle를 수행하지 못하고 ethanolamine 및 ethylene glycol 등 다른 탄소원 또는 질소원을 공급하는 것으로 확인되었다. 비록 관련된 많은 연구가 필요함에도 불구하고 대사체분석 기술을 활용한 Y. enterocolitica 균체 및 배지 분석 연구는 배양시간에 따른 대사과정의 변화를 관찰할 수 있을 뿐만 아니라 향후 균체 연구의 다양한 분야에 접목될 수 있을 것으로 사료된다. To better understand microbial metabolism, metabolomic profiles of Yersinia enterocolitica and its growth media at the different growth stages(6, 12, 24, and 48h) were analyzed using gas chromatography-mass spectrometry(GC/MS). The collected data were statistically analyzed and visualized by Partial Least Squares-Discriminant Analysis(PLS-DA). Significant differences among sample groups were observed on the PLS-DA scores plot. The differences of thirteen metabolites including amino acids, organic acids related to the TCA cycle, ethylene glycol, and ethanolamine were observed over the growth stage. Based on these metabolites, the metabolic pathway associated with the growth stage of Y. enterocolitica has been proposed. Energy sources of Y. enterocolitica might be changed according to the growth stage. This metabolomic study suggested that energy at the log phase might be produced via the TCA cycle but after the stationary phase, and thus the TCA cycle was dysregulated and other carbon and/or nitrogen sources such as ethylene glycol and etanolamine might be required.

      • Terminal Ileitis Caused by Yersinia enterocolitica and Presents with Intussusception Mimicking Crohn’s Disease

        The pathological importance of the infection by Yersinia enterocolitica has been increasing. Even though Yersinia has been implicated in Crohn’s disease (CD), its role in CD is controversial. We present a case of a child with terminal ileitis which is due to Yersinia enterocolitica infection and mimics CD. This case demonstrates that clinical manifestations, sonographic, radiologic, endoscopic, and histologic findings of Yersinia enterocolitica enteritis presenting with intussusception. We compared the endoscopic findings and fecal calprotectin between the acute infection phase and the recovery phase after conservative treatment. We also compared previous cases of children with Yersinia enterocolitica infection mimicking CD. Yersinia enterocolitica infection should be included in the differential diagnosis of patients suspicious of CD.

        Yersinia enterocolitica infection in adults in Korea is reported rarely. Therefore, the possibility of Yersinia infection as a cause of abdominal pain may be overlooked. Because its clinical features are similar to those of acute appendicitis or other diseases that require operations, Yersinia enterocolitis should be diagnosed before surgery. We recently experienced a case of Y. enterocolitica enterocolitis accompanied by postoperative colonic adhesion. A 39-year-old female patient with a 5-year history of asymptomatic uterine myoma underwent transabdominal hysterectomy due to abdominal pain. However, the pain worsened. On CT, diffuse swelling of the right colon and adhesion of the sigmoid colon with luminal narrowing were found. Colonoscopy showed multiple aphthous ulcers with mucosal hyperemia from the cecum to the sigmoid colon. Multiplex PCR with stool specimens, a hemagglutination test for Y. enterocolitica O:3, and tissue culture were positive for Y. enterocolitica. After fluids and antibiotic therapy, her symptoms were relieved. (Korean J Med 2012;83:757-763)

        돼지에서 분리한 Yersinia enterocolitica의 생물형, 혈청형 및 항균제 감수성

        A study on the isolation of Yersinia from the feces of healthy pigs and the biotype and serotype and susceptibility to 16 antimicrobials was carried. Out of 853 pigs, Yersiniae were isolated from 349 pigs(40.9%). Of 349 isolates, 289 isolates(82.8%) were Yersinia enterocolitica and 54(15.5%) were Y kristensenii, 3(0.9%) were Y pesudotuberculosis and the rest 3(0.9%) were Y prederiksenii. Out of 289 isolates of Y enterocolitica, the predominants biotype was 3B comprising of 165 isolates(57.1%) and followed by biotype 2, comprising of 49 isolates(17.0%), bioptype 3A, comprising of 41 isolates(14.2%) and biotype 4 comprising of 23 isolates(8.0%). And the predominant serotype was 0 : 3 comprising of 231 isolates(79.9%) and followed by serotype 0 : 9 comprising of 42 isolates(14.5%) and 0 : 21 comprising of 10 isolates(3.5%). Y. enterocolitica were resistant to cephalothin(99%), novobiocin(99%), erythromycine(83%), ampicillin(83%) and carbenicillin(81%) and susceptible to amikacin(100%), colistin(100%), gentamicin(100%), kanamycin(100%), polymyxin B(100%), tobramycin(100%), chloramphenicol(99%), nalidixic acid(99%), neomycin(99%), streptomycin(99%) and tetracycline(99%). Most strains of biotype 2/serotype 0 : 9 were susceptible to carbenicillin(100%) and ampicillin(61%) but the other biotype/serotypes were resistant to these antibiotic.
