      • An Immunohistochemical Study of Viral Antigen in Aborted Fetuses Naturally Infected by Bovine Viral Diarrhea Virus

        The tissue distribution and cellular localization of viral antigen in the brain of aborted fetus with bovine viral diarrhea virus(BVDV) infection was studied; BVDV antigens was detected in spleen, kidney, lung, eyelid as well as brain. In the brain, the virus was recognized in neurons and non-neuronal cells in the cerebellum and cerebrum. Many cells in the superficial layer and occasional Purkinje cells had BVDV antigens. As well, BVDV was also found in the perivascular cells, vascular endothelial cells and smooth muscle cells in the vessels and neuroglial cells in the white matter. This finding suggests that BVD virus favors infect progenitor cells in the brain, notably in the superficial layer of cerebellum, and damage normal development of cerebellum, which leads to cerebellar hypoplasia.

      • Hepatic Coccidiosis in a Native Korean Goat

        A case of chronic cholangiohepatitis associated with Eimeria spp. is reported in a 6-month-old, male, native Korean goat. The goat died after having a 1-week history of diarrhea, anorexia and weight loss. At necropsy, numerous multifocal to coalescing, pale and mottled red foci were present throughout the liver. Histologically, numerous coccidian parasites in both sexual and asexual stages were found in the intrahepatic biliary epithelia and bile duct lumens. Based on the light microscopic and ultrastrastructural features, the parasites present in the liver were compatible with the genus Eimeria ; however, the species was not speciated.

        Weekly subcutaneous injection of7 ,12-dimethylbenz[a]anthracene(DMBA) at a dose level of 0.25kg/mg body weight induced proliferative lesions in the spleen of syrian golden of syrian In addition, subcutaneous tumors at injection sites were observed. The splenic lesions included stromal hyperplasia and hacmangioma-like lesion.

      • Clinical, ophthalmological, and pathological findings of hypovitaminosis A in cattle

        From February to September in 1996, a lot of blind cattle were reported in Dangjin Area of Chungnam Province. Blindness were observed in the 143 calves of the 1,496 calves from 79 farms. Blind cattle were found only in the farms where green pasture was not fed. After administration of vitamin A as the type of feed additives and parenteral injection, the blindness cases were not occurred in those farms. Both pupils were totally dilated with the absence of pupillary light reflex in all blind cattle. In the ophthalmoscopic examination, the fundus revealed multi focal linear white mottling which was more severe in nontapetal fundus than tapetal fundus. Serum vitamin A concentration was as low as 4.1 ,$\mu\textrm{g}$/dl in the calves of the affected farms. Narrowing of optic foramen, retinal degeneration and optic disc atrophy were shown in pathological findings.

      • Fumonisin $B_1$ induced renal toxicity in Sprague-Dawley rats

        We investigated the effect of a single intravenous dose of Fumonisin $B_1(FB_1$) of rat kidney on the time sequence. Male Sprague-Dawley rats were intravenouslyin jected with FB$_1$at 1.25 mg/kg and were euthanized at 12 hrs, 1, 2, 4, and 6 days after the injection. In $FB_1$ treated rats, serum BUN and creatinine were elevated from 12 hrs. Microscopically, the initial target site was tubules of inner stripe, with mild degenerative and necrotic changes at 12 hrs, but the tubules recovered on day 4. In outer stripe, there were only a few scattered necrotic cells on day 1. These changes became more obvious over the time passed and most severe on day 4. On day 6, regeneration occurred, manifest as hypertrophic, basophilic tubular cells. The dying cells were proved to necrotic cells instead of apoptotic cells by TUNEL. Ultrastructural changes were cytoplasmic vacuole, dilated endoplasmic reticulum, swollen mitochondria, ballooned microvilli of the tubular cell in the outer stripe. These results showed that the renal tubules of outer medulla were the target to $FB_1$-induced nephrotoxicity in the rat. However, initial target was mner stnpe of medulla.

      • Cytokine expression and localization during the development of glomerulosclerosis in FGS mice

        To elucidate the mechanism of age-related development in FGS/NgaKIST mice with spontaneous glomerulosclerotic lesion, we examined expression and localization of various cytokine mRNA in the kidney in the progression of diseases. This mouse model is the first to develop spontanously occuring glomerosclerotic lesion in the kidney. In this study, we detected the up-regulation of local cytokine genes such as IL-1$\beta$, IL-2, IL-6, IL-10, TNF-$\alpha$, TGF-$\beta$, and IFN- $\gamma$ in the kidneys. In RT-PCR and Southern blot analysis, we detected gradual expressions of cytokine mRNA of IL-1$\beta$, IL-2, IL-6, IFN- $\gamma$, and TNF $\alpha$ mRNA during the course of disease. Other cytokines including IL -10 and TGF -$\beta$ were found to be appeared the slightly expressed level at 3 to 12 weeks before onset of inflammatory lesion but they are highly expressed at the end-stage of the disease accompaning high proteinurea and wasting. In situ RT-PCR, each cytokine mRNA were specifically localized in a variety of cells including mesangial, endothelial, parietal epithelial, tubular epithelial, arterial muscle cell, and infiltrated inflammatory cells. In addition, TNF - $\alpha$was detected moderately in the visceral and parietal epithelial cell, but weakly in endothelial and mesangial cells, whereas IL-1 $\beta$ and IL -6 were strong in mesangial regions. IL-6 and TNF- $\alpha$ was highly localized in the damaged proximal and collecting tubules. Especially, TGF -$\beta$ mRNA was highly found in mesangial cells within glomerulus and interstitium during the end-stage of this disease.. These results indicate that pro inflammatory cytokines such as IL-1 $\beta$, IL-2, IL-6, and TNF- $\alpha$ were gradually expressed from the early stage of this disease to the end-stage, and that IL-10 and TGF-$\beta$ may be important in the accumulation of extracellular matrix(ECM) within glomerulus and periglomerular fibrosis in the progression of this disease as well as tissue destruction in end-stage of this disease.

      • Enhanced Progression from Lung Adenoma to Adenocarcinoma in Progeny by Prozygotic Testicular X-ray Exposure of Mice

        Testicles of nine-week-old male CBA/J mice were X-ray irradiated (1 Gy or 2 Gy) and were mated one week later with untreated virgin 12-week-old females of the same strain. The 1-Gy offspring (88 males and 62 females), 2-Gy offspring (100 males and 93 females) and additional offs pring (83 males and 84 females) were treated once subcutaneously with 0.1 mg/g body weight of urethane at 6 weeks of age. These three groups of off offspring showed similar incidences of lung tumors in both sexes. Depending on the doses of paternal X-ray irradiation, increasing incidences of adenocarcinoma were observed in the male 1-Gy and 2-Gy offspring groups. An increased multiplicity of lung carcinomas was observed in the male 2-Gy progeny that was statistically significant when compared with the control group. The results indicate that prezygotic testicular X-ray exposure of patemal animals causes the shift of adenoma-carcinoma sequence towards malignancy in the progeny.

      • Pathological Changes in Rats Fed petasites japonicus Maxim II. Immunohistochemical Localization of Cytochrome P4502E1 and GST-P in Liver

        We investigated metabolism and carcinogenesis in livers of Sprague-Dawley rats fed juices and pelleted diets containing Korean native plants petasites japonicus Maxim by evaluating cell localization and expression of cytochrome P450s and GST-P. Anti-cytochrome P450s application in liver sections revealed three to four times increased expression of cytochrome P450E1 immunoreactivity in degenerative hepatocytes when compared to histologically normal hepatocytes. Anti-GST-P in showed positive pren plastic foci as well as in individual hepatocytes randomly scattered throughout all liver sections examined. Additionally GST-P was evident in proliferative endothelial cells and biliary epithelial cells in exposed rat livers. These results suggested that the increased level of cytochrome P4502E1 in affected hepatocytes was a direct consequence of Petasites japonicus toxicity. Further immunoreactivity to anti-GST-P in hepatocytes endothelial cells and biliary epithelial cells indicated a possible preneoplastic effects of Petasites japonicus in Sprague-Dawley rat.

      • Incidental Finding of Capillaria hepatica in a Raccoon Dog(Nyctereutes procyonoides)

        Capillaria hepatica a primary trichuroid nematode parasite in rodents, has been reported in wide range of secondary hosts including humans. We report a rare case of Capillariasis in a 4-year old raccoon dog(Nyctereutes procyonoides) submitted to the Veterinary Diagnostic Laboratory Chonbuk National University for necropsy on July 5, 1996. Grossly the thoracic cavity contained approximately 200 mls of blood-tinged exudate. Edematous lesion was present around aorta and vena cava. liver was slightly swollen and mottled and contained multifocal yellowish-white lesions ranging from 1 to 3 mm in diameter. Histologically liver lesions were localized around central veins and were characterized by cell swelling and necrosis with mixed inflammatory cells surrounding parasite eggs. These eggs were barrel-shaped with polar caps consistent with Capillaria hepatica.
