Helicobacter pylori와 대장균의 Shuttle Vector 개발

In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

최선의 복지정책은 무엇인가? 또는 정부가 가난한 사람들을 도와줄 수 있는 최선의 방법은 무엇인가? 자발적 자선이야말로 빈곤을 해결하는 데 있어서 문제가 전혀 없는 방법이다. 그리고 자발적 자선으로 빈곤을 해결하는 데 큰 문제가 없다. 그러나 그런 자발적 자선보다도 더 좋은 빈곤 해결책 또는 빈곤 해결을 위한 최선의 해결책은 각자가 자신의 이익을 위하여 노력하도록 하는 것이다. 다시 말하면, 이타심이 아닌 이기심이야말로 빈곤을 해결할 궁극적인 해결책이다. 그 점을 인식하는 것이 빈곤을 궁극적으로 해결하기 위한 첫걸음이다.

고온 초전도체 Y₁-x Gdx Ba₂Cu₃O7-y의 임계전류밀도에 관한 연구

고온 초전도체 Y₁-??Nd??Ba₂Cu₃O??-??(x=00.1, 0.2, 0.3,...,1.0)를 고상 반응법을 이용하여 제작하였다. 온도에 따른 전기적 비저항의 측정과 자기적 감수율의 측정을 통하여 구한 초전도 전이온도는 92.1∼93.5K 사이에 존재하였다. 77K에서 시료의 자기적 감수율은 -5.06∼-6.83Χ10??emu/㎤ㆍOe 사이에 존재하였으며, 상온에서의 비저항은 x=0.6인 시료를 제외하고 x가 증가함에 따라 감소하였다. 수송 임계전류밀도는 자기장이 0일때 51.6∼80.6 A/㎠사이에서 측정되었으며 x가 증가함에 따라 대체로 증가하였다. 자기장을 가해준 상태에서 측정한 수송 임계전류밀도 는 자기장이 증가함에 따라 급격히 감소하였다. 자기 이력곡선으로부터 Bean의 공식을 이용하여 구한 임계전류밀도는 수송 임계전류밀도의 0.8∼2.7Χ10??배로 크게 나타났다. High-Tc superconductors Y₁-??Nd??Ba₂Cu₃O??-??(x=0.1, 0.2, 0.3,…,1.0) have been prepared by solid state reaction method. The superconduction transition temperatures determined by the temperature dependence of electric resistivity and magnetic susceptibility are in the range between 92.1 and -6.83Χ10??emu/㎤·Oe and the resistivities at room temperature are decreased as increasing Gd contents except for the value of x=0.6 sample. Transport critical current densities have the value of 51.6∼80.6 A/㎠ and are generally increased with increasing the magnetic field. Magnetic critical current densities obtained from the magnetization curve using the Bean's law are about 0.8∼2.7Χ10??times of the transport J??.

소아에서 간질발작의 임상양상과 뇌파검사의 관계

목적:발작의 분류는 발작의 원인, 예후를 추정하거나 항경련제를 선택하는 데 있어 매우 중요하며 뇌파검사는 발작의 종류를 구분하고 적절한 치료를 하기 위해 시행하는 중요한 검사이다. 저자들은 소아에서 임상적 발작이 확실한 경우 간질발작의 임상양상과 뇌파검사 간의 일치 정도를 알아보고자 한다. 방법:2000년 1월 1일부터 2008년 9월 30일까지 1회 이상의 간질발작을 주소로 가천의대 길병원 소아청소년과에 방문한 환자 중 본원에서 처음으로 뇌파검사를 시행한 환자를 대상으로 하였다. 발작의 임상양상은 목격자와 본인의 설명을 기록한 의무기록지를 기준으로 하여 부분발작과 전신발작으로 분류하였고 뇌파검사는 발작간간질양방전을 기준으로 정상과 비정상으로 분류하였으며 각성과 수면상태 모두에서 시행하였다. 결과:총 461명의 환자의 뇌파검사 시의 평균 연령은 6.7세였고 남자는 247명, 여자는 214명이었다. 전체 환자에서 발작의 임상양상은 부분발작이 310명, 전신발작이 187명이었다. 각성뇌파검사에서 비정상은 158명으로 부분발작파가 118명, 전신발작파가 59명, 두 가지 발작파가 한 환자에서 나온 경우가 19명이었고, 수면뇌파검사에서는 비정상이 239명으로 부분발작파가 196명, 전신발작파가 77명, 두 가지 발작파가 한 환자에서 나온 경우가 34명이었으며 전체 환자에서는 비정상이 273명으로 부분발작파가 216명, 전신발작파가 97명, 두 가지 발작파가 한 환자에서 나온 경우가 40명이었다. 간질증후군은 전체 환자 중 90명에서 관찰되었다. 146명(41.5%)의 부분발작과 44명(23.4%)의 전신발작 및 79명(87.8 %)의 간질증후군이 뇌파소견과 일치하였다. 결론:영유아 및 소아에 있어 발작을 진단할 때에는 발작양상 분 아니라 정확한 뇌파소견을 세심하게 고려하여야 한다. Purpose:Electroencephalography(EEG) is an essential method carried out for classifying seizures and taking appropriate treatment. The aim of this study is to investigate the concordance between clinical findings of epileptic seizures and EEG in children. Methods:We enrolled 461 patients from those who visited Gil hospital from January 1, 2000 to September 30, 2008 with the chief complaint of epileptic seizure more than once and checked their first EEG at the same hospital. The clinical findings of seizure were based on the charts and interictal waking and sleep EEGs were done. Results:The mean age of the patients was 6.7 years old. 497 epileptic seizures occurred and its clinical finding included 310 of partial seizures and 187 of generalized seizures. In 315 waking EEG 158 were abnormal including 118 of partial seizures, 59 of generalized seizures and 19 of both seizures, in 431 sleep EEG 239 were abnormal including 196 of partial seizures, 77 of generalized seizures and 34 of both seizures, and in waking and/or sleep EEG 273 were abnormal including 216 of partial seizures, 97 of generalized seizures and 40 of both seizures. Epileptic syndromes were observed in 90 patients. 146(41.5%) of partial seizures, 44(23.4%) of generalized seizures, and 79(87.8%) of epileptic syndromes accorded with EEG. Conclusion:When diagnosing seizures in children, we must consider not only clinical findings but also accurate EEG findings.

소상공인 보증지원과 경영성과: 서울지역을 중심으로

본 연구의 목적은 서울지역의 소상공인 보증지원과 경영성과를 분석함에 있다. 2016년부터 2019년까지 4개년 사이 서울신용보증재단으로부터 창업자금, 창업교육, 창업컨설팅, 사후관리 등 보증지원을 받은 소상공인 사업자 1,375개의 자료를 분석하였다. 회귀분석한 결과는 다음과 같다. 첫째, 남성 사업자, 사업자의 높은 연령, 음식점 업종이 매출액에 긍정적 영향을 주었으며, 총 자산이 높을수록, 임차료가 높을수록 매출액이 증가하였다. 둘째, 남성 사업자, 사업장 소재지가 서울동부 지역인 경우 순이익에 긍정적 영향을 미쳤으며, 총자산이 높을수록, 임차료가 높을수록 순이익에 긍정적이었다. 셋째, 남성 사업자, 사업장 소재지가 서울동부 지역인 경우 투자수익률에 긍정적 영향을 주었으며, 임차료가 높을수록 투자수익률에 긍정적이었다. 본 연구는 소상공인 보증지원으로 경영성과를 높이기 위한 시사점을 다음과 같이 들 수 있다. 첫째, 여성사업자의 역량강화를 위해 여성사업자 중심의 경영지원 서비스 정책을 수립하고 집중적인 교육이 이루어지도록 실행방안을 강화해야 한다. 둘째, 소상공인의 비즈니스가 지속성장할수 있도록 지원대상 소상공인을 발굴하고 자금과 컨설팅 지원을 확충해야 한다. 셋째, 상권분석, 사업계획서 작성, 자금조달, 마케팅전략 등 경영자의 역량을 강화하는 교육과 컨설팅 공급이 업종별로 지역별로 충분히 이루어져야 한다. The purpose of this study is to analyze the effect of guarantee support for small business owner in Seoul on business performance. This study analyzed 1,375 data that received guarantee support of start-up fund, start-up education, start-up consulting and follow-up management from the Seoul Credit Guarantee Foundation from 2016 to 2019. The results of the regression analysis are as follows. First, the male founder, the higher the founder's age, the restaurant industry had a positive effect on sales amount. And the higher the total assets, the higher the rent, the more positive the sales amount. Second, the male founder, business location in eastern Seoul had a positive effect on net income amount. Also the higher the total asset, the higher the rent, the more positive the net income amount. Third, the male founder, business location in eastern Seoul had a positive effect on the return on investment. Also the higher the rent, the more positive the return on investment. This study may include the following implications for enhancing business performance with small business owner‘s guarantee support. First, in order to strengthen the capabilities of female entrepreneurs, policies on management support services centered on female businesses should be established and implemented to provide intensive education. Second, it is necessary to find small business owners eligible for support and expand funding and consulting support so that small business owners can continue to grow. Third, education and consulting supply should be sufficient for each industry, each area to strengthen management's capabilities, such as commercial area analysis services, business plan preparation, financing, and marketing strategies.
