        Transcriptome analysis reveals synergistic modulation of E-cadherin/N-cadherin in hMSC aggregates chondrogenesis

        Background N-cadherin-mediated cell adhesion is a vital inductor for mesenchymal condensation in chondrogenesis. Recent studies have revealed the involvement of E-cadherin in enhancing the multipotency of mesenchymal stem cells (MSCs) and limb development; however, the signaling crosstalk of E/N-cadherin remains unclear. Objective This study aimed to explore the synergistic modulation of E/N-cadherin in the chondrogenic differentiation of MSC aggregates. Methods Human E/N-cadherin-functionalized (hE/N-cad-Fc) poly (lactic-co-glycolic acid) (PLGA) microparticles (hE/N-cad-PLGA) were incorporated into the human MSC (hMSC) aggregates to upregulate the expression of the corresponding endogenous cadherin. The chondrogenic differentiation of the hMSC aggregates was initiated by hE/N-cad-PLGA, controlling the release of transforming growth factor-β (TGF-β). A transcriptome analysis was used to assess differentially expressed genes (DEGs) modulated by hE/N-cad-Fc in hMSC aggregate chondrogenesis. Gene functions and signaling pathways were assessed using Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. The associated biological pathways were assessed by a protein–protein interaction (PPI) network analysis, and the results were further confirmed by real-time quantitative PCR (qPCR) and western blotting. Results A total of 1083 DEGs, comprising 111 upregulated and 972 downregulated genes, were discovered to be related to the enhanced chondrogenic differentiation modulated by hE/N-cad-Fc. The GO and KEGG functional enrichment analyses revealed that hE/N-cad-Fc synergistically regulated the p53-related survival signaling pathway. PPI analysis revealed that mitogen-activated protein kinases (MAPK) caspase regulation is a core aspect of the chondrogenic differentiation process, confirmed by western blotting. Conclusion To the best of our knowledge, our study is the first to reveal that the synergistic modulation of E/N-cadherin enhances the chondrogenic differentiation of hMSCs via the ERK1/2-p53 signaling axis.

        Stenotrophomonas maltophilia outer membrane protein A induces epithelial cell apoptosis via mitochondrial pathways

        Stenotrophomonas maltophilia (S. maltophilia) is a common opportunistic pathogen in intensive care units and causes infections most often after surgeries in immune-compromised patients such as those undergoing chemotherapy. Outer membrane protein A (OmpA) is the most abundant of the outer membrane proteins in S. maltophilia. Previous studies on OmpA usually focus on its interaction with the host cells and its role in vaccine development. However, the impact of OmpA on the virulence of S. maltophilia to host cells and the effects on apoptosis remain unclear. In this study, we exposed purified recombinant S. maltophilia OmpA (rOmpA) to HEp-2 cells and investigated the effects of OmpA on epithelial cell apoptosis. Morphologic and flow cytometric analyses revealed that HEp-2 cells stimulated with rOmpA multiple apoptosis features, including nuclear roundness and pyknosis, chromatin aggregation, and phosphatidylserine eversion. We found that rOmpA regulated the protein levels of Bax and Bcl-xL in HEp-2 cells, leading to changes in mitochondria permeability and the release of cytochrome c and apoptosis-inducing factors into the cytoplasm. These subsequently activate the caspase-9/caspase-3 pathway that promote apoptosis. We also observed that rOmpA enhanced the generation of reactive oxygen species and increased intracellular Ca2+ levels in HEp-2 cells. Collectively, our data suggested that rOmpA induced epithelial cells apoptosis via mitochondrial pathways.

        Partially A-optimal blocked multi-factor designs

        In this paper, we propose a partially A-optimal criterion for block designs where multiple factors are arranged. The number of levels of each factor is assumed to be arbitrary and unequal block sizes are allowed. A sufficient condition is derived for a design to be partially A-optimal among all feasible designs. Then the properties of the selected design and its relation with orthogonal arrays are studied. Methods of constructing designs satisfying the sufficient condition are also given.

      • Cases and Inspirations of Pipeline Damages Due to Geohazard in China in Recent Years

        In past few years, a number of serious pipelines failures occurred in China due to the geological disasters. Five geological disasters induced pipeline events are presented, including the explosion accident of Sino-Myanmar gas pipeline in Qinglong county of Guizhou province in June 10, 2018. This paper briefly introduces the characteristics, forming conditions and inducing factors of geological hazards in each accident sites, analyzes the causes of pipeline damage, and summarizes the understanding and lessons, with a view to providing some enlightenment for pipeline geological hazard management in the future. The main understandings include that the human activities are the main factors leading to geological disasters, and four of the five cases related to landfills. Therefore, in the daily management of pipelines, it is necessary to pay much attention to and prevent large-scale engineering activities near the pipeline. The case that the landslide led to a pipeline failure in new Guangming district, Shenzhen, is a reminder that the pipeline in the path of landslide movement is also needed appropriate protection. The identification of geological disasters in the early stage is both important and feasible. In these cases, at least three disaster sites in Shenzhen, Pingxiang and Qinshui have obvious signs of early deformation. The conception of risk acceptability should be advocated in pipeline safety management. The risk of pipeline geological disasters cannot be completely eliminated, but the risk loss can be minimized by improving the emergency plans and other measures.

        Computational fluid dynamics simulation of hydrodynamics in an uncovered unbaffled tank agitated by pitched blade turbines

        Computational fluid dynamics (CFD) simulations were applied for evaluating the hydrodynamics characteristics in an uncovered unbaffled tank agitated by pitched blade turbines. A volume of fluid (VOF) method along with a Reynolds stress model (RSM) was used to capture the gas-liquid interface and the turbulence flow in the tank. The reliability and accuracy of the simulations are verified. The simulation results show that the vortex can be divided into central zone and peripheral zone, and flow field in the tank can be divided into forced vortex flow region and free vortex flow region. With the increase of impeller speed, the vortex becomes deeper, while the critical radius of the two zones keeps almost unchanged. The impeller clearance and the rotational direction have little effect on the vortex shape. The vortex becomes deeper with increasing of the impeller diameter or the blade angles at the same rotational speed. Power number is little influenced by the impeller speed, and decreases by about 30% when impeller diameter varies from 0.25T to 0.5T. When blade angle varies from 30o to 90o, power number increases by about 2.32-times. Power number in uncovered unbaffled tank is much smaller than that in baffled tank, but is very close to that in a covered unbaffled tank. The discrepancy of power number in uncovered unbaffled tank and that in covered unbaffled tank is less than 10%.

        Preparation and simulation performance of light carbon fiber paper-based electromagnetic shielding materials

        The arrival of the 5G era has made electromagnetic pollution a problem that needs to be addressed, and flexible carbon-based materials have become a good choice. In this study, wet continuous papermaking technology was used to prepare carbon fiber paper (CFP) with a three-dimensional conductive skeleton network; Molybdenum disulfide (MOS2)/ iron (Fe) @ carbon fiber paper-based shielding material was prepared by impregnating and blending molybdenum disulfide/iron (MOS2/Fe) phenolic resin MOS2/ Fe@ CFP. The morphology, structure, electrical conductivity, mechanical properties, hydrophobicity, and electromagnetic shielding properties of the composite were characterized. The results show that the three-dimensional network structure based on a short carbon fiber paper-based conductive skeleton and the synergistic effect of the MOS2 dielectric wave absorbing agent and Fe magnetic wave absorbing agent have good electromagnetic shielding performance. Conduct electromagnetic shielding simulation using HFSS software to provide options for the structural design of CFP. The electromagnetic shielding performance of CFP reaches 70 dB, and the tensile strength reaches 34.39 MPa. Based on the mechanical properties, the compactness of carbon fiber paper is ensured. The lightning damage model test using CST software expands the direction for the use of carbon fiber paper. In summary, MOS2/Fe @CFP with excellent shielding performance has great application prospects in thinner and lighter shielding materials, as well as high sensitivity, defense and military equipment.

        Structure evolution in carbon molecular sieve membranes derived from binaphthol-6FDA polyimide and their gas separation performance

        Here we reported a causal relationship between the molecular structure of binaphthol-based polyimideprecursor and the pore-size distribution of the derived carbon membrane. The binaphthol-basedpolyamide acid is synthesized from 2,20-diol-1,10-binaphthyl-6,60-diamine and 4,40-(hexafluoroisopro-pylidene)diphthalic anhydride (6FDA). Then, an azeotropic imidiaztion method was used to synthesizethe polyimide with naphthol groups (XS1). When the imidization is carried out by using acetic anhydride,the polyimide with acetyl groups (XS4) is achieved. The CMS membranes prepared by pyrolyzing XS1 andXS4 at 500, 550, and 600 ℃ are named using the temperature as the suffix, such as XS1-500. Their poreevolution has been investigated using TGA, FTIR, XRD, and Raman measurements. The trimodal pore-sizedistribution is in the carbon molecular sieve (CMS) membranes derived from XS4 and the CMS onesderived from XS1 exhibit a bimodal pore structure. Among them, XS4-500 exhibits the highest gaspermeabilities of 3332 barrer for CO2, 773 barrer for O2, and 119 barrer for N2. XS1-500 only affords theCO2, O2, and N2 permeabilities of 1086, 230, and 30.2 barrer. The esterification of naphthol not justdisturbs the hydrogen bonds between polyimide chains but also affects the pore generation of thederived CMS membranes. Our work provides an effective way to enhance the gas permeability of a CMSmembrane derived from the binaphthol-based polyimide.

        Evaluation of light-emitting diode colors and intensities on slaughter performance, meat quality and serum antioxidant capacity in caged broilers

        Objective: This study was to evaluate the interaction of three different light-emitting diode (LED) light colors (white, green, and blue) and three intensities (5, 10, and 15 lx) on slaughter performance, meat quality and serum antioxidant capacity of broilers raised in three-layer cages. Methods: A total of 648 (8-days-old) male broiler chicks (Cobb-500) were randomly assigned in 3×3 factorially arranged treatments: three light colors (specifically, white, blue, and green) and three light intensities (namely, 5, 10, and 15 lx) for 35 days. Each treatment consisted of 6 replicates of 12 chicks. The test lasted for 35 days. Results: The semi-eviscerated weight percentage (SEWP) in 5 lx white was higher than that in 15 lx (p<0.01). The eviscerated weight percentage (EWP) (p<0.05) and water-loss percentage (WLP) (p<0.01) decreased in 10 lx white light than those in green light. Under blue light, the content of hypoxanthine (Hx) in muscle was lower than that under white and green light (p<0.01). The content of malondialdehyde (MDA) in 15 lx blue light was higher than that in 10 lx green light (p<0.05). Light color had an extremely significant effect on thigh muscle percentage, WLP, Hx, and crude protein content (p<0.01). Light intensity had a significant effect on SEWP (p<0.05), EWP (p<0.05), lightness (L*) value (p<0.05), WLP (p<0.01), and the contents of superoxide dismutase (p<0.05), MDA (p< 0.01), glutathione peroxidase (p<0.01). Conclusion: Using white LED light with 10 lx light intensity can significantly improve the chicken quality of caged Cobb broilers, improve the content of inosine acid in chicken breast and enhance the antioxidant capacity of the body. We suggest that the broiler farm can use 10 lx white LED light source for lighting in 8 to 42 days.

        Influence of Solution Concentrations on Surface Morphology and Wettability of ZnO Thin Films

        ZnO thin films were grown on silicon substrates using a hydrothermal method. The XRD patterns show that all of the peaks can be attributed to the wurtzite structures of ZnO. The TC value of (002) plane and average crystal size increase first and then decrease with the increase of solution concentration. SEM and AFM results show that many dense hexagonal cylinder particles have been observed on the surface of the thin films, which grown at 0.08 and 0.10 mol/L. The surface roughness of the thin films deposited at 0.06,0.08, 0.10, and 0.12 mol/L are 24.5, 38.3, 32.0, and 39.4 nm, respectively. Surface wettability results show that the preferential orientation along c-axis and surface roughness contribute significantly to the hydrophobicity. The reversible switching between hydrophobicity and hydrophilicity is related to the synergy of the transition of wetting model, surface crystal structure, and surface roughness.
