        TEPC (Tissue Equivalent Proportional Counter) was usually used for high LET radiation dosimetry. We developed a prototype TEPC for micro-dosimetry in the range of 0.2~300 keV/μm. And, the simulated site diameter of the TEPC is 2 μm, of similar size to a cell nucleus. For purposes of characterization the response for high LET radiation of the TEPC has been investigated under 135MeV/u Carbon ions in HIMAC (Heavy Ion Medical Accelerator). We determined the gas multiplication factor and measured the lineal energy spectrum [yd(y)] of 135 MeV/u Carbon ions. The value of the gas multiplication factor was 315 at 700 V bias voltage. As a result of the experiment, we could more understand the performance of the TEPC for high LET (Linear Energy Transfer) radiation. And the procedure of high LET radiation dosimetry using TEPC is established.

        Anti-Apoptotic Pathway by Specific Wavelength of Irradiation in SNP-Induced SH-SY5Y Neuroblastoma Cell

        The nitric oxide(NO) is a major factor contri buting to t he loss of neurons in ischemic st roke. demyelina t ing diseases, and other neurodegenerative di sorders . But it is known that NO is not function ing as a direct neurotoxin. NO combined with superoxide(02-) by the diffusion-contl'O ll ed reaction, formed a peroxin it ri te anion (ONOO-)‘ which this s pecies has been shown to contribute to oxidative s igna ling and damage. ONOO stimuJates apoptosis in many cell types. whether ONOO acts direct ly as an ox idant 0 1' the induction of apoptosis is because of the radicals derived from ONOO- decompositi on . But. the mecha ni sm by which ONOO- induces apoptosis is un clear although subsequent forrnation 0 1' reactive oxygen s pecies(ROS) has been suggested in a few reports The aim of this study is to investigate the a nti -apoptotic pathway by inhibi tion 0 1' ONOO synthesis t hrough scavenging of ROS us ing s pecific wavelength 0 1' light irradiation . The present study investi gated the a nti -apop totic effect of the specific wavelength 0 1' irradi ation in Sodium Nitroprusside(SNP) t reated SJ-I-SY5Y ceJls, by MTT, DNA fragmentation, and flow cytometric assay and th rough western blot and caspase-3, -9 activity assay for confirmation of caspase pathway. Also. NO reJease and ROS leveJ was measured in order to observe the changes of NO involved in radical by Griess reaction analysis and DCF'-DH. Results showed that the cell viability were r educed by about 50% of control group by SNP treatment, but re covered to about 80% by 590nm irradiation . The apoptotic cells were observed by flow cytomet ry and DNA fra g mentation assay in SNP-treated group‘ but 590nm irradiation led apoptotic feature to be reduced . Released NO a nd ROS level were increased after SNP treatment but ROS level was dec reased in 590nm irradi at ion - treated group, in spite of high NO concentration fo llowing SNP treatment Also. SNP t reatment led cytochrom C release but 590nm irraidiation inhibit it, hence the expression 0 1' caspase-3 and -9 was dec reased sign ificantly‘ These results showed that 590nm irradiation protect neuronal death thl'Ough bl ocking of NO-induced mi tochondri al apoptotic pathway. Also, it suggests that specific wavelength of irradi ation was used for prevent ion from neurodegenerative disordcr progression

      • Interrelationship Between Clonidine-Induced Hypotension and Opiate Receptors in Rabbits

        Clonidine은 특히 중추에서 adrenergic alpha₂-receptor의 효능약으로 카테콜아민의 유리를 억제하므로써 혈압강하제로 상용되는 약물이다. Urethane 마취 가토를 이용하여 clonidine 의 혈압하강 반응 및 심박감소 반응에 대한 opiate 수용체와의 상관성을 관찰하기 위하여 본 연구를 시행하여 얻어진 결과는 다음과 같다. Clonidine(5-20ug/㎏)을 가토의 대퇴정맥내로 투여시 용량의존성의 현저한 혈압하강 및 심박감소 반응을 나타내었다. 또한 Clonidine(2.5-10ug)을 측뇌실내로 투여시에도 용량의존성의 유의성인 혈압 하강 및 심박감소 작용을 나타내었다. Opiate 수용체 길항약인 naloxone(30ug /㎏/30min)을 정 내로 전처치한 다음에는 clonidine의 정맥 및 측뇌실내 투여에 의한 혈압강하 및 심박감소 반응이 뚜렷이 억제 되었다. 측뇌실내로 naloxone(8ug)을 투여한 후에도 정맥 및 측뇌실내 clonidine으로 나타나는 혈압하강 및 심박감소 작용이 현저하게 감약되었다. 본 연구에서 사용된 용량에서 naloxone은 측뇌실내 및 정맥내에 투여시 혈압 및 심박에 별다른 영향을 미치지 못했었다. 이상과 같은 연구결과로 보아 clonidine의 혈압하강 및 심박감소 작용은 적어도 opiate 수용체의 활성화를 통해서 나타나는 것으로 사료된다. The present investigation was designed to examine the interrelationship between clonidine-induced hypotension and opiate receptors in the urethane-anesthetized rabbits. Clonidine (5-20ug / kg) given into a femoral vein of the rabbit produced a dose-related, marked fall in arterial blood pressure and a reduction in heart rate. Intraventricular clonidine (2.5-10ug) caused dose-dependent and significant hypotensive and bradycardic responses. Intravenous naloxone (30ug / kg / 30min), an opiate receptor antagonist, when given prior to c1onidine, prevented significantly hypotensive and bradycardic responses induced by introvenous of intraventricular clonidine. Naloxone injected into a lateral ventricle before clonidine administration inhibited greatly intravenous and intraventricular clonidine-evoked depressor and bradycardia. Both intravenous and intraventricular naloxone at doses used in the present study did not affect arterial blood pressure and heart rate, suggesting that naloxone increases sympathetic nerve activity peripherally, and may abolish clonidine-induced depressor and bradycardia by also increasing sympathetic activity in the central nervous system through blockade of opiate receptors. From the present experimental data, it. is considered that the hypotensive and bradycardic effect of clonidine may be mediated at least partly through interactions with endogenous opiate peptides.

      • Experimental Studies On extension of survival time for tumor bearing and immunomodulatory effects of Yukgunjatang,Soshihotang,Houttunynia Herba

        In order to investigate the effects of Yukunjatang. Soshihotang. Houttuynia Herba. on extension of survival title for tumor bearing effect after developing of tumors followed by being grafted with the Sarcoma-180 cells and immunomedulatory effects in mice. the extracts of its herbal medicines were orally administered to mice for 21 days. The experimental studies were performed for observation of extention of survival time for tumor bearing effect, quantitation of T cells and B cells, Interleukin-2 productivity, delayed type hypersensitivity (DTH), lymphocyte transformation for cell mediated immune response, hemagglutinin titer and hemolysin titer far humoral immune response and splenic natural killer cell activity (NKCA) in ICR or C5 7 BI/6mice. The results were summerized as follows: 1.Yukgunjatang and Soshihotang were significantly recognized to extend the survival time of tumor bearing mice (p < 0.05 respectively). 2. For the effect of T cell and B cell quantitation, the only Soshihotang-medicated group showed a significant increase on Interleukin-2 productivity (P < 0.05). 3.Yukgunjatang-medicated group showed a significant increase on Inlerleukin-2 productivity (p < 0.05). 4.Delayed type hypersensivity was not significantly increased in all sample groups as compared with control group. But Houttuynia Herba-medicated group showed a little higher than that of the control group. 5.Natural killer cell activity was not significantly increased in all sample groups as compared with control group. But Houttuynia Herba-medicated group showed a little higher than that of the control group in E/T ratio 50:1 6.Lymphocyte transformation was significantly increased in Soshihotang, Houttuynia Herba, and Soshihotang plus Houttuynia Herba-medicated groups (p < 0.01, p < 0.02. p < 0.02 respectively). 7.Serum antibody titer was not significantly increased in all sample groups as compared with control group. Acoording to tole above results. Yukgunjatang, Soshihotang, and Hottuynia Herba are considered to suggest useful herbal medicines on extension of survival time for tumor beraring and immunomodulatory effect. And then it is expected that medicines are more helpful in treatment of cancer patients.

        Background: Concerns have emerged about the adverse effect of phthalates on human health. Objectives: The aim of this study was to investigate the association between di(2-ethylhexyl) phthalate (DEHP) exposure and atopic dermatitis (AD) in Korean children, focusing on the potential dose.response relationship. Methods: A matched case.control study was conducted from May to October 2012. Subjects from 3 to 6 years of age were recruited from kindergartens and daycare centres in Seoul, Korea. The clinical diagnosis of AD was made by dermatologists. A total of 224 cases and 224 age- and sex-matched controls were included. The levels of two phthalate metabolites [mono(2-ethyl-5-hydrohexyl) phthalate (MEHHP) and mono(2-ethyl-5-oxohexyl) phthalate (MEOHP)] of DEHP in urine samples were measured. Results: The effects of DEHP varied by age, and an increased risk for AD was associated with DEHP at age 3 years (odds radio 2?51, 95% confidence interval 1?02.6?20). The association was in the opposite direction in the other age groups although there was no statistical significance. The effects of DEHP on AD was observed differently by the level of the body burden. The predicted risk for AD based on the results of multiple logistic regression analysis showed a nonmonotonous association (U-shaped) between the level of DEHP and the risk of AD. Conclusions: This finding might suggest that the effects of DEHP on AD may be different according to the exposure level or age of the subject. Further longitudinal investigations with a suitable design to investigate the nonmonotonous association should be encouraged.

      • Survey of scrub typhus vectors at epidemic regions in Jeollabuk-Do, Korea 2016

        A surveillance of chigger mites was performed to monitor the incidence of scrub typhus vectors at 5 environmental collection points of 3 locations (Muju, Namwon and Gimge) from April 2016 in Jeollabuk-Do, Korea. During the surveillance period, 3,292 chigger mites were collected and the predominant species were Leptotrombidium pallidum (42.9%). The high environmental collecting rates were recorded at reservoir bank (31.6%) and grassfield (29.7%). We detected Orientia tsutsugamushi from the samples collected in Jeollabuk-Do using nested polymerase chain reaction. The specific DNA of O. tsutsugamushi were detected in 11 pools among total 119 pools and its minimum positive rate was 0.35%. In order to clarify the relationship between chigger mites and scrub typhus cases, further survey will be needed.

        Simultaneous Quantification of Oleins (triolein, diolein and monoolein) in Mouse Feces using Liquid Chromatography-Electrospray Ionization/Mass Spectrometry

        Fat malabsorption is an important cause of poor growth in infancy and childhood. Steatorrhea tests have been developed using various methods. Traditional measurements of stool fat, however, require large samples and it often takes as a week to complete the analysis. In this paper, a liquid chromatography-electrospray ionization/mass spectrometry (LC-ESI/MS) method was developed for simultaneous quantitative analysis of triacylglycerols, triolein, diolein and monoolein, in mouse feces. Moreover, the procedure was rapid, simple as well as compatible with LC-ESI/MS. Chloroform-isopropyl alcohol solution was used for fat-soluble sample extraction. After centrifugation and filtration, an analytical solution was prepared. Triolein, diolein and monoolein were separated using non-aqueous reversed-phase column with the mobile phase consisting of A (methanol) and B (acetone-isopropyl alcohol). The precision (% CV) and accuracy (% bias) of the assay were 3.8-14.7% and 85.2-114.9%, respectively. This method has been successfully applied to simultaneous determination of triolein, diolein and monoolein in feces from 30 mice. This method can therefore be applied to measure triacylglycerols in mouse feces accurately and precisely by LC-ESI/MS, thereby helping to predictive biomarker in fat malabsorption and diagnostic research.
