Importance-Performance Analysis와 Borich 요구도 분석 방법을 활용한 임상간호실무 교육 요구도: 일개 상급종합병원 일반병동 간호사 대상

Purpose: This study’s aim was to confirm the importance and performance of the clinical practice of nurses working in the general wards of a tertiary hospital and to analyze their educational needs. Methods: The study employed a descriptive research design, and a self-reported questionnaire was developed and used for nurses in a tertiary hospital. Data were collected from July 22 to July 29, 2022, analyzed by an independent t-test, paired t-test, and one-way ANOVA for the importance and performance of each clinical nursing practice according to general characteristics, and then a post hoc verification was performed by Scheffé’s test. An Importance-Performance Analysis and Borich needs assessment model were used to analyze clinical nursing education needs. Results: Clinical nursing practice performance showed a significant difference according to length of total clinical career, time working in current department, and preceptor experience. According to the results of the Importance-Performance Analysis, neurological evaluation, and nursing intervention, artificial respirator and high-flow oxygen inhalation nursing, chemotherapy, emergency nursing, and cardiopulmonary resuscitation were identified as belonging to the “concentrate here” quadrant. All these items ranked in the top 10 in the Borich needs assessment model. Conclusion: Based on these results, the current education system should be reviewed, and short and long term education strategies based on educational needs should be established to strengthen the competence of nurses.
MicroRNA super-resolution imaging in blood for Alzheimer’s disease

We propose a novel blood biomarker detection method thatuses miRNA super-resolution imaging to enable the early diagnosisof Alzheimer’s disease (AD). Here, we report a singlemoleculedetection method for visualizing disease-specificmiRNA in tissue from an AD mice model, and peripheral bloodmononuclear cells (PBMCs) from AD patients. Using optimizedMagnified Analysis of Proteome (MAPs), we confirmed that fivemiRNAs contribute to neurodegenerative disease in the brainhippocampi of 5XFAD and wild-type mice. We also assessedPBMCs isolated from the whole blood of AD patients and ahealthy control group, and subsequently analyzed those samplesusing miRNA super-resolution imaging. We detected moremiR-200a-3p expression in the cornu ammonis 1 and dentategyrus regions of 3 month-old 5XFAD mice than in wild-typemice. Additionally, miRNA super-resolution imaging of bloodprovides AD diagnosis platform for studying miRNA regulationinside cells at the single molecule level. Our results present apotential liquid biopsy method that could improve the diagnosisof early stage AD and other diseases.

<P>Tristetraprolin (TTP), a well-characterized AU-rich element (ARE) binding protein, functions as a tumor suppressor gene. The purpose of this study was to investigate whether a bioactive substance derived from a natural medicinal plant affects the induction of TTP and to elucidate its mechanism. We examined the effects of natural bioactive materials including Resveratrol (RSV), thymoquinone (TQ) and curcumin on the expression of TTP in cancer cell. TQ derived from a natural plant Nigella sativa increased the expression levels of TTP mRNA and proteins in a dose-dependent manner in gastric and breast cancer cells. TQ-induced TTP increased the instability of MUC4 mRNA by direct binding of TTP to ARE in the 3′UTR of MUC4 mRNA. The induction of TTP by TQ also reduced the proliferation, migration and invasion of cancer cells. The expression of the epithelial-mesenchymal (EMT)-related genes, which were target genes of TTP, was also decreased by the TQ treatment. In the in vivo experiments using mouse melanoma cells, TQ-induced TTP inhibited metastasis of tumor cells. We have found that TQ-induced TTP might inhibit metastasis by reducing tumor cell migration and invasion through destabilization of MUC4 mRNA, which suggest the MUC4 as a novel target to TTP.</P>
Expression analysis of transcripts responsive to osmotic stress in Deschampsia antarctica Desv.

Deschampsia antarctica Desv., the only monocot species thriving in the tough conditions of the Antarctic region, is surrounded by regions with snow and ice where available amount of liquid water is very limited. Considering the species as an invaluable resource for the identification of genes associated with tolerance to osmotic stress, we have characterized the physiological response and the changes in gene expression induced by PEG treatment using a cDNA macroarray. Since relative chlorophyll contents of leaves decreased to 69 %, proline levels were changed significantly, and expression levels of dehydrin and late embryogenesis abundant protein were significantly up-regulated at 1 day after PEG treatment, we used corresponding plant samples as the material for gene expression analysis. As a result of hybridization and analysis of cDNA macroarray, we identified 44 and 20 transcripts that were up-regulated and down-regulated by PEG treatment respectively, and they were divided into several functional categories. Most of the up-regulated transcripts are involved in 'Response to stimulus', 'Amino acid metabolism', and 'Respiration'. On the other hand, most of the down-regulated transcripts are involved in 'Photosynthesis' and 'Translation'. Among the up-regulated transcripts, a dagger-1-pyrroline-5-carboxylate synthase 2 gene has shown the highest expression level at 1 day after PEG treatment suggesting its possible role as a main player for proline biosynthesis in D. antarctica under osmotic stress. Collectively, these data suggest that the transcripts differentially expressed under osmotic stress may represent candidate genes for dissecting molecular mechanism of adaptation to extreme environment for plants.

<P>We demonstrate the fabrication of highly transparent, electro-conductive, durable, and eco-friendly nanofiber (NF) textiles based on keratin using highly conductive silver nanowire (Ag NW) networks on transparent nano-fibrous substrates for multifunctional, high-performance flexible heaters. These NF-based textiles, including keratin, have a high transparency of more than 89% in the visible area, making them very useful for manufacturing transparent wearable electronics. The Ag NW networks on the nanofibers provide a low sheet resistance of 25 Ω sq<SUP>−1</SUP>, with a maximum optical transparency of 85% and superior flexibility with mechanical durability up to 1000 cycles of bending and release. Flexible and transparent wearable electronic devices, such as heaters, with an exceptionally high mechanical stability and foldability with negligible changes in electrical conductivity have been successfully fabricated by using this simple method. Also, transparent conductive NFs based on keratin are a promising wearable electronic material for a wide range of high performance, low cost, biocompatible and wearable electronics applications. This study successfully demonstrates a flexible heater based on transparent conductive NFs with a wide operating temperature up to 65.75 °C, a good recovery time of about 10 s and low power consumption (0.38 W) for wearable electronics applications.</P>
