        표준항혈청 생산 및 한우 ( Bos taurus coreanae ) 의 혈액형에 관한 연구

        These studies were carried out to produce standard antisera for the serological blood typing of cattle and to clarify the genetic characteristics of Korean Native Cattle. For the serological blood typing, thirty-three standard antisera(reagents) were produced by iso-immunizations and absorptions with two hundreds animals. Seven hundreds and forty-two Korean Native Cattle were bloodtyped by those antisera. Also the phenogroups (alleles) in each blood group system were investigated. Two hundreds animals were bloodtyped with standard antisera which introduced from bovine bloodtyping laboratory, Saskatoon, Canada and used far isoimmuniz anon to produce immunesera. Thirty-two reagents(A1, A2 and Z` of A system; B, C, I1, Y2, A`1, A`2, B`, D`, E`2, E`3, I` and J` of B system; C1, C2, X1, X2, R1, R2, W and L` of C system; F and V of I` system; J of J system; S, H`, U1, U`1, U` and U$quot; of S system: Z of Z system) were produced by iso-immnunizations and absorptions. And J reagent was seperated from natural serum. In ISAG international comparison test in 1991, the specificities of these thirty-three antisera were compared with those of other laboratory. In A blood system, four kinds of phenogroups were appeared. The gene frequency of A1Z` phenogroup, which has been characteristically reported in Indian cattle breed, was 0.206. In B blood system, thirty-five kinds of phenogroups were appeared and showed similar gene frequencies in each other. In C blood system, sixteen kinds of pheuogroups were appeared. The gene frequency of X2 phenogroup was the highest(0.300). In F blood system, three kinds of alleles were appeared. The gene frequency of $quot;-$quot; allele which has been characteristically reported in Indian cattle breed, was 0.213. In S blood system, ten kinds of phenogroups were appeared. The gene frequency of $quot;-$quot; and G$quot; phenogroups were relatively high. In J and Z blood system, the gene frequency of J and Z alleles were 0.432 and 0.600.

        Gsa 유전자의 돌연변이가 쥐의 소마토스타틴 수용체 유전자의 발현에 미치는 효과

        Background: The growth hormone (GH)-secreting pituitary adenoma with mutation of Gsa gene (gsp oncogene) is known to show a higher response to somatostatin. The mechanism for the higher response is unclear. We previously demonstrated that transcription of the rat somatostatin receptor (SSTR) gene was increased by cAMP. Therefore, we investigated whether the mutation of Gsa gene can increase SSTR gene expression. Methods: The mutant Gsa expressing plasmid of which arginine of codon 201was replaced with histidine was transfected transiently and permanently into GH3 cells. cAMP and rat GH were measured by radioimmunoassay. mRNA expression of the rat SSTR2 was determined by a semiquantitative RT-PCR. Results: The intracellular cAMP production was increased by 1.8 folds in the transiently transfected cells and by 1.5 folds in permanently transfected cells compared to those in the cells transfected with the vehicle plasmid, plasmid expressing the wild type or the plasmid expressing the silent mutant of codon 226. The transcriptional activation by cAMP response element of somatostatin.gene was also increased by 2 folds 24 hours after transient transfection and by 3.5 folds in the permanently transfected cells. The transcriptional activation was not enhanced by forskolin in the transiently transfected cells, whereas it was more remarkably inhibited by somatostatin compared to that in the other transfected cells. The expression of SSTR2 was increased by 1.8 folds 24 hours after transfection in the transiently transfected cells and by 3 folds in the permanently transfected cells, and it was not enhanced by forskolin and isobutylmethylxanthine. The secretion of GH from the transiently transfected cells was not significantly higher than that from the wild type-expressing cells, but it was higher in the permanently transfected cells. The suppression of GH by somatostatin was more prominent in the permanently transfected cells compared to the other transfected cells. Conclusion: This study suggests that a higher expression of SSTR induced by the mutant Gsa may be a mechanism by which gsp-positive adenomas showed a higher response to somatostatin (J Kor Soc Endocrinol 14:657-666, 1999).

        한우 , 제주재래흑우 , 흑모화우와 갈모화우에서의 MSH Receptor(MC1R) 유전자의 유전자형 및 빈도 비교

        PCR-RFLP analysis of MSH receptor (melanocyte-stimulating hormone receptor; MCIR) gene was carried out to confirm the genotypes and frequencies in Korean Cattle, Cheju Native Black Cattle, Japnaese Black, Japanese Brown, Holstein and Angus. Genetic relationship of these breeds and coat colors to alleles at the Extension locus, that encodes MCIR were also determined. Allele E^D E^+ and a were detected by digestion with BsrF I (or Msp I ) and Aci I . A Dominant allele E^D, which is considered to be typical allele for dominant black coat color in cattle breeds, such as Holstein and Angus, was not detected in Korean Cattle and Japnases Brown with yellowish-brown coat color. Among sixty-two Cheju Native Black Cattle and fifteen Japanese Black, only ten and eleven heads had the dominant allele E? respectively. The remaining animals possessed the genotype E^+/E^+ or E^+/e, which probably reflect alleles at the A-locus which are supposed to give rise to the recessive black color. Several animals of Cheju Native Black Cattle and Japanese Black possessed the genotype E^D/-, probably reflecting the genetic introgression of E^D allele into these breeds during the crossbreeding period for improving. These results show that the difference of MCIR genotype and frequency among these breeds may be useful for conservation of endangered Cheju native Black Cattle and also genetically discreminating between meat of Korean Cattle and that of Holstein

        Pore size-controlled carbon aerogels for EDLC electrodes in organic electrolytes

        <P>Pore size-controlled carbon aerogels (CA_X) are prepared from two starting materials: resorcinol and formaldehyde. Carbon aerogels with different pore sizes are successfully obtained by simply varying the polymerization time (X) in the polycondensation of resorcinol and formaldehyde. Pore sizes of the carbon aerogels monotonically increase with increasing polymerization time. The pore size of the carbon aerogel plays an important role in determining its electrochemical behavior (i.e., specific capacitance in EDLC) especially at high current density. A sufficiently large pore size in a carbon aerogel is required for easy transport of electrolyte ions, and these types of carbon aerogels provide excellent electrochemical performance as an EDLC electrode. CA_20, which was polymerized for the longest time, have the largest pore size and the largest pore volume. CA_20 exhibit the highest specific capacitance due to facile electrolyte ion transport. As a result, design of carbon aerogels with a sufficiently large pore size while maintaining a high specific surface area is very important for developing carbon aerogels for use in EDLC electrodes. In addition, we propose a schematic reaction pathway for the preparation of carbon aerogels in ambient conditions from resorcinol and formaldehyde using a base catalyst based on the characterization results.       • Smartphone Diagnostics Unit (SDU) for the assessment of human stress and inflammation level assisted by biomarker ink, fountain pen, and origami holder for strip biosensor

        <P>There is a growing interest in the area of affective disorder (i.e. psychological stress and depression) among advanced countries. Since the real-time diagnosis of human stress has been of great importance, we have recently reported the Smartphone Linked Stress Measurement (SLSM); the smartphone holder yet has two negative points, which are too excessive weight to carry and expensive cost to manufacture. In this paper, we have further developed the SLSM for the psychological stress in order to overcome the drawbacks mentioned above. We call the newly developed system as the Smartphone Diagnostics Unit (SDU). The SDU is composed of an origami smartphone holder, three cotton swabs, three strip biosensors, and three bottles of buffer solution. We demonstrated a light-weighted and economical origami smartphone holder to fulfill an easy-to-carry unit and low cost of fabrication, respectively. To validate the SDU, two kinds of stress-related biomarkers in human saliva, such as cortisol and C-reactive protein (CRP), were measured. The successful measurement of the concentration of salivary biomarkers enables the SDU to be useful for the real-time detection of human psychological stress in the future. (C) 2016 Elsevier B.V.         To see whether there is any differencies in RNA dependent DNA polymerase activities between monoclonal antibody-producing hydridoma cells and non-producing hybridoma cells, Balb/c female mice were immunized with the purified viral surface protein of D-variant of encephalomyocarditis virus and then fused with myeloma cells (NR-1). After cloring, monoclonal antibody-producing hybridoma cell lines were separated from non-producing hydridoma cell lines. RNA-dependent DNA polymerase activities were measured in the supernatant of monoclonal antibody-producing hybridoma clones and non-producing hybr idoma clones, and myeloma cells as control, Monoclonal antibody-producing hybridoma cells showed statistically significant higher activity as compar compare to that of nonproducing hybridoma cells. To find whether RNA-dependent DNA polymerase releasing cells aware secreting or budding C-type virus particles, those cells were examined with electron microscope. The hybridoma cell which secrete large amount of RNA-dependent DNA polymerase shows significant number of extracellular C-type virus particles. In constrast, non-producing hydridoma cells contains a lot of intracellular C-type virus particles. It is concluded that monoclonal antibody-producing hydridoma cells released particles. It is concluded that monoclonal antibody-producing hydridoma cells released significant amount of RNA-dependent DNA polymerase land extracellular C-type virus particles, while non-producing hydridoma cells showed less release of RNA-dependent DNA polymerase and contains intracellular C-type virus particles.

        분만년도, 계절 및 산차가 한우의 비유곡선에 미치는 영향

        축산기술연구소 대관령지소에서 사육된 한우 암소 118두에 대해 산유량 및 비유곡선을 추정하고자 하였다. 분만 후 1, 2, 3 및 4개월의 일일 평균 산유량은 3.74±0.96㎏, 3.64±0.97㎏, 3.26±1.15㎏ 및 2.99±1.08㎏ 이었으며 4개월간의 일일 평균 산유량은 3.52±0.84㎏이었다. 일반적인 비유곡선의 상수 추정치는 A(분만 직후 첫유량)가 2.4845, b(상향경사도)가 0.1734 및 c(하향경사도)가 0.0060이었다. 최고 추정유량은 3.75㎏이었고 최고 유량 도달시기는 29.03일이었는데 초산우 보다는 경산우가 최고 추정 유량이 높고 최고 유량 도달시기가 늦어지는 것으로 나타났으며 봄철 분만우가 가을철 분만우에 비해 최고 추정 유량이 높고 최고 유량 도달시기가 늦어지는 것으로 나타났으며 봄철 분만우가 가을철 분만우에 비해 최고 추정 유량이 높고 최고 유량 도달시기도 빨라지는 경향을 보였다. This study was conducted to investigate the effect of environmental factor on milk yield and to estimate lactation curve in Korean cattle. The data for milk yields were collected from 118 cows from 1997 to 2000 at National Livestock Research Institute in Daekwanryoung, Kangwon-do. Average daily milk yields for 1st, 2nd, 3rd and 4th month after calving were 3.74㎏, 3.64㎏, 3.26㎏ and 2.99㎏. Average daily milk yield for the four months was 3.52㎏. The milk yields for cows calved in spring were larger than those calved in fall. Lactation curve of Korean cattle was y_n=2.4845n^0.1734e^-0.0060n. Peak milk yield was 3.75㎏ on 29.03 day after calving. The peak milk yields for multi-parous cows were larger than those of primiparous cows. The peak milk yields for multi-parous cows reached later than those for primiparous cows. The cows calved in spring had higher and earlier peak milk yields than those calved in fall had.

      • 일개 농촌지역주민 이병실태에 관한 조사 : 경기도 남양주군 수동면 Su-Dong Myun, Nam Yang-Ju Gun, Kyung-Gi Do

        A study on the morbidity status in a Korean rural community, Su-Dong Myun, Nam Yang-Ju Gun, Kyung-Gi Do was carried out, through analyzing the medical records of Su-Dong Myun Health Subcenter during the period of January to December, 1983 and the following results were obtained. 1) Population of Su-Dong Myun was 5,040 composed of 2,561 male and 2,479 female and the sex ratio was 103.3. 2) Population of Su-Dong Myun by age was composed of 31.5% age group of 0-14, 47.1% age group of 15-49 and 21.4% age group of 50 and over. and dependency rate of Su-Dong Myun Population was 77.1% in 1983. 3) The number of new patients visited to Su-Dong Myun Health Subcenter was 2,811 composed of 51.9% male and 48.1% female during the year of 1983. 4) Annual utilization rate in total was 557.7 per 1,000 population and 570.1 for male and 545.0 for female, and the highest age specific utilization rate was 1,807.6 per 1,000 in the age group of 0-4 and lowest was 242.4 per 1,000 in the age group of 20-24. 5) The order of five major diseases was Diseases of the Respiratory System (36.8%), Diseases of Digestive System (21.1%), Diseases of the Skin and Subcutaneous (14.3%), Accidents, Poisonings, and Violence (11.4%) and Diseases of the Nervous System and Sense Organs (8.4%). 6) The average visiting time of out-patients in the Health Subcenter was 1.60 per case. The range of average visiting times of diseases was 1.00 to 5.02. 7) The monthly distribution of out-patients visited to Su-Dong Myun Health Subcenter was shown the highest rate in August (71.0) and the lowest in February (37.1) per 1,000 poulation. 8) The annual inhabitant utilization rate by Rie was the highest in Ribsouk 3 Rie (842.4), and the lowest in Gidoon Rie(249.4) per 1,000. 9) The proportion of medical fee of patients visited to Health Subcenter by sources were 66.4% of patients paid by Su-Dong Myun Community Health Development Association and patient self, 17.0% by Medical Insurance Program and 16.6% by Government Medical Aid Program. 10) Average medical fee per first visiting was 2,763±863(₩) and average medical fee per case was 4,002±3,701(₩).

        선진복합재료 적층판의 동적 층간 파괴 인성평가

        In this paper, an investigation was performed on the dynamic interlaminar fracture toughness of CFRP(carbon fiber reinforcement plastics) composite laminates. Composite laminates used in this experimentation are CF/EPOXY and CF/PEEK laminated plates. In the experiments, Split Hopkinson's Bar(SHPE) test was applied to dynamic and notched flexure test. The mode II fracture toughness of each unidirectional CFRP was estimated by the analyzed deflection of the specimen and J-integral with the measured impulsive load and reactions at the supported points. As an experimental result, the vibration amplitude of CF/PEEK laminates appear more than that of CF/EPOXY laminates for the J-integral and displacement velocity at a measuring point. Also, it is thought that the dynamic. fracture toughness of two kind specimens(CFRP/EPOXY and CF/PEEK) with the in crease of displacement velocity becomes a little greater at a measuring point within the range of measurement.

      • Facilitated intracellular delivery of peptide-guided nanoparticles in tumor tissues

        Macromolecular nanoparticles can extravasate and accumulate within tumor tissues via the passive targeting system, reflecting enhanced permeability and the retention effect. However, the unsatisfactory tumor therapeutic efficacy of the passive-targeting system, attributable to the retention of extravasated nanoparticles in the vicinity of tumor vessels, argues that a new system that facilitates intracellular delivery of nanoparticles within tumors is needed. Here, we developed hydrophobically modified glycol chitosan (HGC) nanoparticles conjugated with interleukin-4 receptor (IL-4R) binding peptides, termed I4R, and tested them in mice bearing IL-4R-positive tumors. These HGC-I4R nanoparticles exhibited enhanced IL-4R-dependent cellular uptake in tumors compared to nonconjugated nanoparticles, leading to better therapeutic and imaging efficacy. We conclude that I4R facilitates and enhances cellular uptake of nanoparticles in tumor tissues. This study suggests that the intracelluar uptake of nanoparticles in tumors is an essential factor to consider in designing nanoparticles for tumor-targeted drug delivery and imaging.
