        Tunicamycin enhances TRAIL-induced apoptosis by inhibition of cyclin D1 and the subsequent downregulation of survivin

        TNF-related apoptosis-inducing ligand (TRAIL) has been proposed as a promising cancer therapy that preferentially induces apoptosis in cancer cells, but not most normal tissues. However, many cancers are resistant to TRAIL by mechanisms that are poorly understood. In this study, we showed that tunicamycin, a naturally occurring antibiotic, was a potent enhancer of TRAIL-induced apoptosis through downregulation of survivin. The tunicamycin-mediated sensitization to TRAIL was efficiently reduced by forced expression of survivin, suggesting that the sensitization was mediated at least in part through inhibition of survivin expression. Tunicamycin also repressed expression of cyclin D1, a cell cycle regulator commonly overexpressed in thyroid carcinoma. Furthermore, silencing cyclin D1 by RNA interference reduced survivin expression and sensitized thyroid cancer cells to TRAIL; in contrast, forced expression of cyclin D1 attenuated tunicamycin-potentiated TRAIL-induced apoptosis via over-riding downregulation of survivin. Collectively, our results demonstrated that tunicamycin promoted TRAIL-induced apoptosis, at least in part, by inhibiting the expression of cyclin D1 and subsequent survivin. Of note, tunicamycin did not sensitize the differentiated thyroid epithelial cells to TRAIL-induced apoptosis. Thus, combined treatment with tunicamycin and TRAIL may offer an attractive strategy for safely treating resistant thyroid cancers.       • Effects of Systemic and Focal Hypoxia on the Activities of Rostral Ventrolateral Medullary Neurons in Cats

        Rendering the brain ischemic would evoke the cerebral ischemic reflex which is characterized by an arterial pressor response, apnea and bradycardia. Since the rostral ventrolateral medulla (RVLM) is known to play a key role in the maintenance of normal cardiopulmonary activity, during the cerebral ischemic reflex some cardiac related cells should be excited and respiration related cells inhibited. In this context, the responses of RVLM neurons to systemie and focal hypoxia were analyzed in the present study. Twenty-five adult cats of either sex were anesthetized with ${\alpha}-chloralose$ and the single neuronal activities were identified from RVLM area. For the induction of focal hypoxia in the recording site, sodium cyanide was applied iontophoretically and for systemic hypoxia the animal was ventilated with nitrogen gas for a twenty-second period. Cellular activities were analyzed in terms of their discharge pattern and responses to the hypoxia by using post-stimulus time and single-pass time histograms. Of eighteen cardiac related cells recorded from the RVLM area, twelve cells were excited by iontophoresed sodium cyanide and of twenty-five respiration related cells, fourteen cells were excited by iontophoresed sodium cyanide. Remaining cells were either inhibited or unaffected. Eight of fifteen cells tested with iontophoresed sodium lactate were excited and remaining seven cells were inhibited. Systemic hypoxia induced by nitrogen gas inhalation elevated the arterial blood pressure, but excited, inhibited or unaffected the single neuronal activities. Some cells showed initial excitation followed by inhibition during the systemic hypoxia. Bilateral vagotomy resulted in a decrease of arterial pressor response to the systemic hypoxia, and a slight decrease in the rhythmicity related to cardiac and/or respiratory rhythms. The single neuronal responses to either systemic or focal hypoxia were not affected qualitatively by vagotomy. From the above results, it was concluded that the majority of the cardiac- and respiration- related neurons in the rostral ventrolateral medulla be excited by hypoxia, not through the mediation of peripheral chemoreceptors, and along with the remaining inhibited cells, all these cells be involved in the mediation of cerebral ischemic reflex.

        First Report of an Unrecorded Nematode-Trapping Fungus Species Monacrosporium phymatopagum in Korea

        A fungus that can capture nematodes by means of sessile adhesive knobs was isolated from rotten wood in Uiseong, Korea. It was found to produce single, spindleshaped,3-4 septate (commonly 4-septate) conidia, 44.8μm (range, 41.6-50.1 μm) long and 13.3 μm (range, 10.7-15.4 μm) wide. Conidiophores were found to be hyaline,erect, straight, and 202.7-245.7 μm high. On the basis of these morphological features, the fungus was identified as Monacrosporium phymatopagum. This is the first report of M. phymatopagum in Korea which can be a potential biological control resource of plant parasitic nematode.

        Korean Red Ginseng Attenuates Hepatic Lipid Accumulation via AMPK Activation in Human Hepatoma Cells

        In this study, we examined Korean red ginseng (KRG) extract affects on the lipid metabolism in HepG2cells. Increase in AMP-activated protein kinase (AMPK)phosphorylation was observed when the cells were treated with KRG. Activation of AMPK was also demonstrated by measuring the phosphorylation of acetyl-CoA caboxylase (ACC), a substrate of AMPK. KRG down-regulated gene expressions of sterol regulatory element binding protein 1c (SREBP1c) and its target proteins, such as fatty acid synthase (FAS) and stearoyl-CoA desaturase (SCD1) in time- and dose-dependent fashions. In contrast, gene expressions of peroxisome proliferator-activated receptor α(PPARα) and CD36 were increased. These effects were reversed in the presence of compound C, an AMPK inhibitor. However, there were no differences in gene expressions of SREBP2, 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase, and low-density-lipoprotein receptor (LDLR). Taken together, KRG induced supression of SREBP1c and activation of PPARα via AMPK and these effects seem to be one of anti-hyperlipidemic mechanism of KRG in HepG2 cells.

        Phellinus linteus Extract Exerts Anti-asthmatic Effects by Suppressing NF-${\kappa}B$ and p38 MAPK Activity in an OVA-induced Mouse Model of Asthma

        Phellinus linteus has been used as a traditional herbal medicine in Asian countries and is known to have anti-tumor, immunomodulatory, anti-inflammatory, and anti-allergic activities. However, the protective effects of P. linteus against experimental asthma have not been fully investigated. The objective of this study was to determine whether P. linteus ethanol extract (PLE) suppresses inflammatory response in an OVA-induced asthma model. As expected, the oral administration of PLE significantly inhibited eosinophilic airway inflammation and airway hyperresponsiveness in OVA-challenged BALB/c mice. Supporting these data, the augmentation of Th2 cytokines (IL-4, IL-5, and IL-13), eotaxin, and adhesion molecules in lung tissues and bronchoalveolar lavage fluid after OVA inhalation was markedly attenuated by PLE. Furthermore, PLE reduced OVA-induced activation of NF-${\kappa}B$ and p38 MAPK in lung tissues. Therefore, our results suggest the potential of P. linteus as a therapeutic agent for asthma.

        Adsorption of Bovine Serum Albumin onto Tannic Acid-Immobilized Chitosan Resin

        A novel tannic acid-immobilized chitosan resin (TICR) was prepared by Mannich reaction for the adsorption ofproteins. The physical properties of TICR were characterized and the effects of contact time, pH, and initial concentration of(bovine serum albumin) BSA on its adsorption by TICR were investigated. The Langmuir isotherm model was applied todescribe the adsorption isotherm. The equilibrium data are fitted to the Langmuir isotherm model. The maximum monolayerBSA adsorption capacities of TICR were found to be 1.094, 1.487, and 1.694 mg/g at 298, 308, and 318 K, respectively. Furthermore, the data were analyzed on the basis of pseudo-first order, pseudo-second order, and intraparticle diffusionmodels. The correlation results suggested that the pseudo-second order model fitted the experimental data very well. Thethermodynamic study indicated that the adsorption process of BSA onto TICR was endothermic, and the Gibbs free energy(ΔGo), enthalpy (ΔHo), and entropy (ΔSo) of the adsorption process were calculated according adsorption isotherm data. TICRcould be reused for 10 times with only 19 % loss of adsorption capacity for BSA.

        Graphene nanosheet/silicone composite with enhanced thermal conductivity and its application in heat dissipation of high-power light-emitting diodes

        Heat dissipation from light-emitting diodes (LEDs) has become a serious problem because of the LEDs' high luminosity and power. This problem could be solved by improving the dissipation efficiency of each LED system component. A microwave-reduced graphene nanosheet/silicone (GN/silicone) composite with a high thermal conductivity and stability was prepared by mechanical blending. The thermal conductivity of the composite reaches 2.7 W/(m K) with only 1.5 wt% loading, and is 12 times higher than the pure silicone matrix. When used as a thermal interface material between high-power LED chip module substrates and heat sinks, the thermal conductive GN/silicone composite could decrease the temperature difference between the substrate and shell. It could also improve the system heat transfer efficiency. The temperature gap between the heat slug and the heat sink was less than 2 C with a 1.5 wt% loading of GNs.

        Evaluation of reference genes for miRNA expression analysis in Galeruca daurica (Coleoptera: Chrysomelidae) using qRT‐PCR

        The selection of suitable reference genes (RGs) for data normalization is a crucial step for obtaining reliable results from gene expression analysis using the quantitative real-time polymerase chain reaction (qRT-PCR). Expression stability of eleven putative RGs (miR-100-5p, miR-92a-3p, miR-998-3p, miR-279d-3p, miR-305-5p, miR-276a-3p, miR-275-3p, miR-9a-5p, miR-2a-3p, U6 snRNA and 5S rRNA) was assessed by using a web-based tool: RefFinder, which integrates four algorithms (Delta Ct, geNorm, NormFinder, and BestKeeper), and the best combination number of RGs was calculated by geNorm. Validation of selected RGs was performed by an expression analysis of let-7-5p under different experimental conditions. The best combinations of RGs were shown as follows: miR-100-5p + miR-275-3p for temperature treatment, miR-100-5p + miR-305-5p for adult tissues, miR- 276a-3p + miR-9a-5p for genders, miR-2a-3p + miR-276a-3p for days after eclosion, U6 snRNA + miR-9a-5p + miR-2a-3p for developmental stages, and U6 snRNA + miR-279d-3p for all above conditions, and there was no one RG suitable for all experimental conditions. Incorrect RG selection affected the evaluation of let-7-5p abundance. Our findings demonstrate that an integrated approach of multiple algorithms is necessary to identify reliable, stable RGs for miRNA evaluation, and provide a basis for further studies on expression and function analysis of miRNAs and their target genes in Galeruca daurica, a new pest with great outbreaks in Inner Mongolia since 2009.
