Catabolic Plasmid-Mediated Heavy Metal Resistance in Herbicide Diuron-Degrading Pseudomonas species

Three Pseudomonas strains (Bk8, Bk9, Bk10) selected from soil for their ability to degrade herbicide diuron were tested for their heavy metal resistance. The growth of these catabolic strains on a minimal medium with various concentrations of Cd^2+, Zn^2+, Ni^2+', and Hg^2+ revealed a minimal effect on the carbon source for the inhibitory effect of the metals. One of these strains, namely, Bk8, exhibited a high resistance to the heavy metals as compared to the two other strains. This strain harbors plasmid pBk8 (110kb) and contains at least four determinants encoding heavy metal resistance. Nickel and zinc resistance are encoded by genes located on the chromosome, while cadmium and mercury resistance are on plasmid pBk8. Accordingly, the characteristics of strain Bk8 suggest that it would be useful in the bioremediation of aromatic compounds in the presence of toxic heavy metals as co-contaminants.
Pascal Distribution Series Connected with Certain Subclasses of Univalent Functions

The aim of this article is to make a connection between the Pascal distribution series and some subclasses of normalized analytic functions whose coefficients are probabilities of the Pascal distribution. For these functions, for linear combinations of these functions and their derivatives, for operators defined by convolution products, and for the Alexander-type integral operator, we find simple sufficient conditions such that these mapping belong to a general class of functions defined and studied by Goodman, Rønning, and Bharati et al.
Plasmid-Mediated Aniline Assimilation by Pseudomonas sp. B10

An aniline-utilizing microorganism identified as a species of Pseudomonas was isolated from soil contaminated highly with aniline and urea-herbicide. This strain was able to utilize aniline as the sole source of carbon and energy, and was shown to harbor a single large plasmid mediating the aniline assimilation. Subsequent plasmid-curing of this bacterium resulted in the abolishment of the aniline utilizing phenotype and the loss of catechol-C2, 3O-oxygenase. The reestablishment of the plasmid, denoted pB10, in cured Pseudomonas sp. via filter surface mating, resulted in restoration of the aniline assimilation abilities and enzyme activity.

A group of new N,N-bis(5-acetylpyridin-2-yl)phenylamine derivatives was synthesized in good yield applying an optimized Buchwald-Hartwig amination protocol. The synthesized compounds showed UV absorption maxima in the range of 320-360 nm, and showed good luminescence at dilute concentrations in the blue region of the spectra (in the range of 480-497 nm). They showed also a bathochromic shift associating the increase in solvent polarity. The synthesized compounds could be investigated for use in OLEDs or as potential monomers for PLEDs.

Phenylaminopyrimidines represent a large group of new selective anticancer agents, the majority of which exert their action through the inhibition of specific kinases. In this study, a new series of N-substituted-2-aminopyrimidines has been designed and synthesized. A selected group of the synthesized derivatives was screened at a single dose concentration of 10 ${\mu}M$ over a panel of 60 cancer cell-lines. Compound 12e has showed great inhibitory and strong lethal effect over almost all of the 60 cell-lines and accordingly was further tested in a 5-dose testing mode to determine its $IC_{50}$ values, where it showed great efficacies with intermediate potencies over the tested cell-lines. The compound was also tested over a panel of 52 kinases to explore its kinase inhibitory profile, and was found to be a selective but moderate inhibitor over FLT3 kinase.
A Highly Selective Staurosporine Derivative Designed by a New Selectivity Filter

KIST301135 was semi-synthetically prepared by the reaction of Staurosporine with triphosgene in anhydrous dichloromethane. The structure of KIST301135 was confirmed by $^1H$ NMR, $^{13}C$ NMR, and high resolution mass spectrum. KIST301135 was initially tested in a single dose duplicate mode at a concentration of 20 nM over a panel of 53 kinases against Staurosporine as a positive control. KIST301135 has showed inhibitions above 75% in only 2 kinases (FLT3 and JAK3 kinases) of the 53 tested kinases, while Staurosporine has showed inhibitions above 80% in about 62% of the tested kinases. KIST301135 was retested at a 5-dose testing mode over the 9 kinases inhibited by percentages over 20 at the single dose testing in order to determine its $IC_{50}$ values. KIST301135 has shown much improved kinase inhibitory selectivity relative to Staurosporine in its potency at JAK3 kinase and CAMK2b kinase.

Although the pyrimidine derivatives were obtained in low yields ranging from 8% to 20%, we reported the successful preparation of N,N-diaryl-pyrimidin-2-amine derivatives starting from the corresponding 2-aminopyrimidines (1a-1c), by direct palladium-catalyzed arylation using different arylbromides. The reasons of low yields are thought to be the electronic and steric effects by the neighboring aromatic systems. The absorption spectra and photoluminescent spectra of compounds (3a 3g and 4a-4c) were measured using dichloromethane on the concentration of 25 mM by UV vis spectroscopy and luminescent spectroscopy. Pyrimidine derivatives 4a, 4b, and 4c showed moderate emission maxima at 474 nm, 481 nm, and 367 nm, respectively, while other compounds showed very weak photoluminescence or no photoluminescence at all.

Aims: Hepatitis C virus (HCV) is a major risk of hepatocellular carcinoma (HCC), however, the molecular mechanism of HCV-related hepatocarcinogenesis is still unclear. The polycistronic microRNA (miR)-17~92 cluster, designated also as “OncomiR-1”, plays a role in tumorigenesis and is transactivated by c-MYC oncogene. The cluster negatively regulates phosphatase and tensin homolog (PTEN) and activates nuclear factor-kappa B (NF-κB). The present study was designed to evaluate plasma levels of miR-17 host gene (MIR17HG) protein, encoded by miR-17~92 cluster host gene, in patients with HCV-related HCC in relation to tumor c-MYC, PTEN and NF-κB expression. Methods: Forty five patients with chronic HCV infection [18 patients with chronic hepatitis C (CHC), 12 patients with cirrhosis and 18 patients with HCC] and 15 healthy subjects were enrolled in the study. The HCC stage was determined according to the Barcelona Clinic Liver Cancer (BCLC) staging system and Cancer of the Liver Italian Program (CLIP). Quantitative measurement of plasma MIR17HG protein levels was performed using an enzyme-linked immunosorbant assay kit. The HCC histologic grade was assessed according to Edmonson and Steiner scoring system. Immunohistochemical staining of liver specimens was done using anti-human antibodies against c-MYC, PTEN and NF-kB and was scored semiquatitatively. Results: Plasma MIR17HG protein levels showed significant increases in CHC and cirrhotic patients with and without HCC compared with healthy subjects and in patients with HCC compared with those without HCC (P < 0.001). By plotting ROC curve, the sensitivity and specificity of plasma MIR17HG protein levels in detecting HCC were 100% and 93.3% respectively at a cut-off level of 257.25 pg/ml [AUC = 0.996]. The HCC tissues showed a significant increase in c-MYC, and NF-kB expression and a significant decrease in PTEN expression compared with CHC, cirrhotic and surrounding non-neoplastic liver tissues (P < 0.05). The plasma MIR17HG protein levels were positively correlated with tumor size (P = 0.001), stage (P = 0.001) and HCC histological grade (P = 0.002) and c-MYC (P = 0.001) and NF-kB expression (P = 0.015) and were inversely correlated with PTEN expression (P = 0.001). Conclusions: Activation of miR-17~92 cluster induced by c-MYC may play an important role in the development and progression of HCV-related HCC, possibly, through inhibition of PTEN and activation of NF-kB and could provide a potential therapeutic target for HCC. Circulating MIR17HG protein could be a useful non-invasive biomarker for the detection of HCC in chronic HCV infection.
