      • Association of Extracellular Cleavage of E-Cadherin Mediated by MMP-7 with HGF-Induced in vitro Invasion in Human Stomach Cancer Cells

        <P><I>Background:</I> Proteolytic shedding of the ectodomain of a variety of transmembrane proteins, including cell-to-cell adhesion molecules, has been observed in solid cancers. We have investigated whether extracellular cleavage of E-cadherin mediated by matrix metalloproteinase-7 (MMP-7) is involved in hepatocyte growth factor (HGF) induced in vitro invasion in stomach cancer cells. <I>Methods:</I> The effects of HGF on the expression of E-cadherin/β-catenin and MMP-7 at both the protein and mRNA levels were assessed in stomach cancer cells, NUGC-3 and MKN-28, and in cells in which the expression of MMP-7 was downregulated by transfection with a MMP-7 short hairpin RNA plasmid. <I>Results:</I> Treatment with HGF increased the extracellular cleavage of E-cadherin and the release of MMP-7 and reduced the level of E-cadherin in a dose- and time-dependent manner. HGF treatment repressed the phosphorylation of β-catenin in a Triton-soluble fraction, but enhanced this phosphorylation in a Triton-insoluble fraction. The association of E-cadherin with β-catenin was decreased by HGF treatment in the Triton-soluble fraction. In addition, treatment of MMP-7 short hairpin RNA transfected NUGC-3 cells with HGF resulted in no extracellular cleavage of E-cadherin and also decreased the in vitro cell invasion. <I>Conclusions:</I> These results suggest that incubation with HGF mediated the release of MMP-7, resulting in extracellular cleavage of E-cadherin from stomach cancer cells.         Purposes :The purposes of this study was to investigate the self-managing status according to the life styles and the diabetic knowledge of early adulthood. The subjects in this study were 240people, the early adult aged twenties to thirties living in the metropolitan area. Data were collected by using self-administered questionnaires that measure about the knowledge of diabetes; the dietetic behavior; the knowledge of diet; the exercise behavior; the knowledge of exercise. Descriptive statistics, t-test, ANOVA, and correlation were used for data analysis. Result :1. At the knowledge of diabetes degree based on the general character, the subjects who had experiences of diabetic education have higher degree of the diabetic knowledge. 2. At the knowledge of diabetes degree according to the life style, the subjects who have regular diet are appeared highly. Also the subjects who don't take much food of animal origin have higher degree. 3. At the knowledge of diet based on the life style, the subjects who don't keep three times meals a day are appeared highly. Also the subjects who have regular diet have higher degree. 4. At the knowledge of exercise according to the life style, the subjects who take extra nutritions 3-4 times a week are appeared highly. Although the subjects tend to know about dietary, exercise and health management, it showed that they actually don't perform what they know in their everyday of life. The reason for this could be the lack of perceived seriousness about diabetes mellitus. Thus as nursing professionals, we should provide them proper health education and support. Moreover, advanced research on the empowerment is warranted.

      • 700-nm Zwitterionic Near-Infrared Fluorophores for Dual-Channel Image-Guided Surgery

        <P>Purpose: The purpose of this study was to develop a family of 700-nm zwitterionic pentamethine indocyanine near-infrared fluorophores that would permit dual-channel image-guided surgery. Procedures: Three complementary synthetic schemes were used to produce novel zwitterionic chemical structures. Physicochemical, optical, biodistribution, and clearance properties were compared to Cy5.5, a conventional pentamethine indocyanine now used for biomedical imaging. Results: ZW700-1a, ZW700-1b, and ZW700-1c were synthesized, purified, and analyzed extensively in vitro and in vivo. All molecules had extinction coefficients >= 199,000 M-1 cm(-1), emission >= 660 nm, and stability >= 99 % after 24 h in warm serum. In mice, rats, and pigs, >= 80 % of the injected dose was completely eliminated from the body via renal clearance within 4 h. Either alone or conjugated to a tumor targeting ligand, ZW700-1a permitted dual-channel, high SBR, and simultaneous imaging with 800-nm NIR fluorophores using the FLARE (R) imaging system. Conclusions: Novel 700-nm zwitterionic NIR fluorophores enable dual-NIR image-guided surgery.</P>

      • Gold-coated silicon nanowire-graphene core-shell composite film as a polymer binder-free anode for rechargeable lithium-ion batteries

        <P>We designed and fabricated a gold (Au)-coated nanowires/graphene (Au-SiNWs/G) hybrid composite as a polymer binder free anode for rechargeable lithium ion batteries (LIBs). A large amount of SiNWs for LIB anode materials can be prepared by metal assisted chemical etching (MaCE) process. The Au-SiNVVs/G composite film on current collector was obtained by vacuum filtration using an anodic aluminum oxide (AAO) membrane and hot pressing method. Our experimental results show that the Au-SiNVVs/G composite has a stable reversible capacity of about 1520 mA h/g which was maintained for 20 cycles. The Au-SiNWs/G composite anode showed much better cycling performance than SiNWs/ polyvinylidene fluoride (PVDE)/Super-P, SiNWs/G composite, and pure SiNWs anodes. The improved electrochemical properties of the Au-SiNWs/G composite anode material is mainly ascribed to the composite's porous network structure.,(C) 2014 Elsevier B.V. All rights reserved.</P>

      • Fabrication and characterization of Cu-Ni-YSZ SOFC anodes for direct use of methane via Cu-electroplating

        The Cu-Ni-YSZ cermet anodes for direct use of methane in solid oxide fuel cells have been fabricated by electroplating Cu into a porous Ni-YSZ cermet anode. The uniform distribution of Cu in the Ni-YSZ anode was obtained by electroplating in an aqueous solution mixture of CuSO<SUB>4</SUB>.5H<SUB>2</SUB>O and H<SUB>2</SUB>SO<SUB>4</SUB> for 30min with 0.1A of applied current. When the Cu-Ni-YSZ anode was exposed to methane at 700<SUP>o</SUP>C, the amount of carbon deposited on the anode decreased as the amount of Cu in the Cu-Ni solid solution increased. The power density (0.24W/cm<SUP>2</SUP>) of a single cell with a Cu-Ni-YSZ anode was slightly lower in methane at 700<SUP>o</SUP>C than the power density (0.28W/cm<SUP>2</SUP>) of a single cell with a Ni-YSZ anode. However, the performance of the Ni-YSZ anode-supported single cell degraded steeply over 21h because of carbon deposition, whereas the Cu-Ni-YSZ anode-supported single cell showed enhanced durability up to 200h.

        경사지에서 고추 정식시기에 따른 토양유실과 유출수에 대한 식생피복 효과

        As sloped farmland is subject to runoff and soil erosion and consequently require appropriate vegetative coverage to conserve soil and water, a field study was carried out to evaluate the impact of crop canopy coverage on soil loss and runoff from the experimental plot with three different textural types (clay loam, loam, and sandy loam). The runoff and soil loss were examined at lysimeters with 15% slope, 5 m in length, and 2 m in width for five months from May to September 2009 in Suwon (37° 16" 42.67" N, 126° 59" 0.11" E). Red pepper (Capsicum annum L. cv. Daechon) seedlings were transplanted on three different dates, May 4 (RP1), 15 (RP2), and 25 (RP3) to check vegetation coverage. During the experimental period, the vegetation coverage and plant height were measured at 7 day-intervals and then the "canopy cover subfactor" (an inverse of vegetation cover) was subsequently calculated. After each rainfall ceased, the amounts of soil loss and runoff were measured from each plot. Under rainfall events >100 mm, both soil loss and runoff ratio increased with increasing canopy cover subfactor (R²=0.35, p<0.01, R²=0.09, p<0.1), indicating that as vegetation cover increases, the amount of soil loss and runoff reduces. However, the soil loss and runoff were depending on the soil texture and rainfall intensity (i. e., EI30). The red pepper canopy cover subfactor was more highly correlated with soil loss in clay loam (R²=0.83, p<0.001) than in sandy loam (R²=0.48, p<0.05) and loam (R²=0.43, p<0.1) plots. However, the runoff ratio was effectively mitigated by the canopy coverage under the rainfall only with EI30<1000 MJ mm ha<SUP>-1</SUP> hr<SUP>-1</SUP> (R²=0.34, p<0.05). Therefore, this result suggested that soil loss from the red pepper field could be reduced by adjusting seedling transplanting dates, but it was also affected by the various soil textures and EI30.

        Design of the RAON Accelerator Systems

        The RAON is the name of the heavy ion accelerator facility under construction in Korea thatincludes the In-flight Fragment (IF) and Isotope Separation On-Line (ISOL) facilities to supportcutting-edge research in various science fields. The superconducting linac is the driver for the IFfacility that can accelerate beams from proton to uranium with 200 MeV/u, 400 kW (for uraniumbeam). A 70-MeV, 1-mA H− cyclotron is the driver for the ISOL facility and is followed by apost-accelerator consisting of s superconducting linac that can accelerate rare-isotope (RI) beamsand deliver them to experimental halls. These facilities provide high-intensity stable ion and rareisotope (RI) beams for domestic and international users. In this paper, design and prototypingefforts for the RAON accelerator systems are presented.

        Obesity phenotype and cardiovascular changes

        OBJECTIVE: Healthy obese phenotype with favorable metabolic profiles is proposed. However, whether healthy obesity leads to target organ changes is controversial. We investigated the impact of a healthy obesity on cardiovascular structure and function. METHODS: A total of 2540 participants without known cardiovascular disease were enrolled. According to BMI and the metabolic syndrome (MetS) component, the participants were divided into six groups: healthy (none of five MetS components) normal weight (BMI <23 kg/m), unhealthy (one or more of five MetS components) normal weight, healthy overweight (BMI 23–24.9 kg/m), unhealthy overweight, healthy obesity (BMI ≥25 kg/m), and unhealthy obesity. The cardiovascular changes were assessed by echocardiography, tissue Doppler imaging (TDI), carotid ultrasonography, and pulse wave velocity (PWV). RESULTS: In a multivariate analysis after adjusting for age, sex, heart rate, high-sensitivity C-reactive protein, and medication for hypertension and diabetes mellitus, the unhealthy overweight and obese groups showed statistically significant changes in the left ventricular mass index, mitral E/A ratio, E/Ea ratio, TDI Ea velocity, common carotid artery intima–media thickness (CCA-IMT), and brachial-ankle PWV (P < 0.001), compared with the healthy normal weight individuals. In the healthy overweight and obese groups, CCA-IMT and brachial-ankle PWV values were similar, but left-ventricular mass index and TDI Ea velocity were significantly different (P < 0.001). CONCLUSION: Healthy obesity was associated with subtle changes in left ventricular structure and function. These data provide evidence that metabolically healthy phenotypes with excess weight may not be a benign condition.

      • Computational Methods for Patient-Specific Perfusion Simulations of Coronary Arteries

        Patient-specific computational simulations of blood flow are utilized to diagnose and predict treatment outcomes of coronary artery disease. The computational simulations, however, are limited when estimating perfusion in the myocardium as multiscale vessels from arteries to capillaries need to be developed. We propose a multiscale patient-specific computational model framework to simulate blood flow from large coronary arteries to myocardial tissues. Patient vasculatures were segmented from coronary computed tomography angiography data and then extended from the image-based model down to the arteriole level using a space-filling synthetic forest of arterial trees. Blood flow is modelled by coupling a 1-D model of the coronary arteries to a single-compartment Darcy myocardium model. Simulated results for 5 patients with non-obstructive coronary artery disease are compared to [<sup>15</sup>O]H<sub>2</sub>O PET exam data for both resting and hyperemic conditions. Results on a patient with a severe disease demonstrate coronary artery disease can predict myocardial regions with perfusion deficit. This multiscale computational model of simulating blood flow from the epicardial coronary arteries to the left ventricle myocardium will be further validated and applied to human data.

        For the agricultural uses of Manbok alpine area of 900m and 1000m high above sea level located in Gure-gun, Jeonnam province, investigations on the growth and yield of potato, radish and forage crops obtained by different seeding dates and survey on some meteorological conditions of the area were carried out. This experiment was conducted as the second yearly trial in 1971. The results may be summarized as follows: 1. The yearly variations of average air temperature, minimum air temperature and difference between maximum temperature and minimum temperature of the Manbok region were larger than those of plane region. But the yearly variation of maximum air temperature of the region comparatively small. On the other hand the yearly variation of relative humidity of the region was not very large. Relative humidity, amount of cloud and precipitation of the region were higher and more than those of plane region. 2. The growth and yield of potatoes showed better and more in the May 5 seeding plot than those in the May 15 seeding plot at both regions of 900m and 1000m high above sea level. The potato yield of that plot was 578∼757㎏/10a and this was 50∼66% as compared with average yield of Gure-gun, 557∼1% as compared with average yield of the country and 20∼26% as compared with the yield of Daegwanryung Highland Experimental Station. 3. The yield of radish showed more in the June 16 seeding plot than that in the May 28 seeding plot at both regions of 900m and 1000m high above sea level. The radish yield of that plot was 1696∼2234㎏/10a and this was 147∼194% as compared with average yield of the country. 4. The yield in mixed seeding of orchardgrass and ladinoclover showed more in the May 5 seeding plot than that in the May 15 seeding plot at both regions of 900m and 1000m high above sea level. The dry matter yield of that plot was 215∼387㎏/10a and this was 26∼49% as compared with the yield of Daegwanryung Highland Experimental Station.
