Metamodels such as response surface model (RSM) and kriging model are used to meet weight minimization of a double-deck train car-body. Weight minimization problem of the double-deck train car-body is required to decide 36 design variables of thicknesses for large aluminum extruded panel while satisfying stress constraints. Therefore, we attempt to reduce the number of design variables by using RSM as a screening process based on orthogonal array design. Global sensitivity and Pareto plot of responses with respect to design variables are employed as screening criterion. After screening design variables, maximum entropy sampling is employed to select additional sample points. We construct kriging model for the mass and maximum stress of a double-deck train car-body with the reduced design variables. Optimization based on kriging model is performed and the merits of this design methodology are discussed.

3T3-L1 지방전구세포 분화 억제에 대한 Thaumatococcus daiellii 열매 유래 토마틴의 항비만 효과를 검토하였다. 3T3-L1 지방전구세포에 토마틴을 0-5 μM 농도로 처리한 결과 세포생존율은 처리 농도 의존적으로 감소하였는데, 토마틴 8일 처리 후 1 및 3 μM 농도에서 각각 97% 및 88.3%의 세포생존율을 보였다. 또한 토마틴 3 μM 처리농도에서 Oil-Red-O염색 지방구가 현저히 감소된 것으로 나타났다. 3T3-L1 세포 내 중성지방 농도는 양성 대조군에 비해 농도 의존적으로 감소하였다. 따라서 천연 식물성 토마틴은 3T3-L1 지방전구세포의 세포증식 억제 및 중성지방 농도 감소 효과를 보여 항비만 효과가 있는 것으로 밝혀졌다. The effects of thaumatin isolated from katemfe fruit of Thaumatococcus daiellii Benth on 3T3-L1 preadipocyte differentiation was investigated in vitro. 3T3-L1 adipocytes were treated with various concentrations of thaumatin ranging in 0-5 μM. Thaumatin reduced fat accumulation in differentiated 3T3-L1 adipocytes in a dose-dependent manner. 3T3-L1 cell proliferation was 97.0 and 88.3% at 1 and 3 μM after 8 days of thaumatin treatment, respectively. Thaumatin showed a potent inhibitory effect on stained lipid droplets at a concentration of 3 μM. Thaumatin reduced triglyceride accumulation in differentiated 3T3-L1 cells in a dose-dependent manner, compared with positive control cells. This study provides basic information on the anti-obesity activity of thaumatin.
Ge-MOSFETs을 위한 Ni-Co 합금을 이용한 Ni-germanide의 열안정성 개선

In this paper, Ni-Co alloy was used to improve thermal stability of Ni Germanide. It was found that uniform germanide is obtained on epitaxial Ge-on-Si substrate by employing Ni-Co alloy. Moreover, neither agglomeration nor penetration is observed during post-germanidation annealing process. The thermal stability of Ni germanide using Ni-Co alloy is improved due to the less agglomeration of Germanide. Therefore, the proposed Ni-Co alloy is promising for highly thermal immune Ni germanide for nano scale Ge-MOSFETs technology.
알코올 대사 효소 alcohol dehydrogenase (ADH) 및 acetaldehyde dehydrogenase (ALDH) 활성에 미치는 아미노산의 영향

본 연구에서는 숙취해소에 좋은 것으로 알려진 식품 소재의 주요 아미노산을 포함하여 효소 활성에 영향을 미치는 것으로 알려진 아미노산을 선택하였고, 효소 활성도가 상대적으로 높은 yeast와 rat liver 유래의 ADH 및 ALDH 효소를 대상으로 알코올 대사에 관련된 효소 활성의 촉진 효과에 대하여 검토하였다. Rat liver 유래의 ADH 활성은 처리한 아미노산 중에서 arginine에서 가장 높았다. Arginine의 첨가 농도를 달리하여 효소 활성을 측정한 결과 10~50 ㎎/㎖ 농도에서 118~120.6%로 양성대조구의 90.6% 보다 약간 높은 것으로 나타났다. 또한, yeast 유래의 ADH 활성은 methionine에서 가장 높은 활성을 보였고, methionine의 처리 농도를 달리한 경우에서는 첨가 농도 의존적으로 높은 활성을 보였다. Rat liver 유래의 ALDH 활성은 methionine이 가장 높은 활성을 보였다. Methionine의 첨가 농도별 측정에서는 10 ㎎/㎖에서 30 및 50 ㎎/㎖ 첨가 농도에서 보다 높은 활성을 보였으며, 이들 모든 처리 농도에서 양성대조구 보다 상당히 높은 활성을 보였다. 한편 yeast 유래의 ALDH 활성은 각 아미노산별 큰 차이는 없었으나, arginine에서 높은 활성을 보였다. Arginine의 첨가 농도별 측정에서는 처리 농도 의존적으로 활성이 약간씩 증가하는 경향을 보였으며, 양성대조구 보다 높은 활성을 나타내었다. 효모 유래 ALDH 및 rat liver 유래 ADH 효소 활성을 촉진시키는 작용을 가진 arginine을 효모 배양에 첨가시킬 경우 세포 내 ALDH 및 ADH 활성 염색 정도가 증가함으로써 arginine은 ALDH 및 ADH 활성을 촉진시키는 효능이 in vivo 실험계에서도 확인되었다. 이상의 실험 결과에서 아미노산 중에서는 arginine과 methionine이 ADH 및 ALDH 활성을 촉진시키는 작용에 의해 알코올 분해뿐만 아니라 acetaldehyde의 분해도 촉진시킬 가능성이 높아 숙취해소 효과는 물론 간 보호 효과도 동시에 있을 것으로 시사 되어 진다. 따라서 arginine과 methionine과 같은 아미노산을 주류 제품에 첨가하게 될 경우 숙취해소 경감과 간 보호 효능을 어느 정도 나타낼 수 있을 가능성이 제기되었다. The present study examined the comparative effects of various amino acids on the alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities of yeast Saccharomyces cereviciae and rat liver homogenate in vitro. Methionine showed the highest activity in yeast ADH among the amino acids used in this study, but this was not higher than that of the hangover product, Condition-Power (CP) used as positive control. Methionine was also found to be the best amino acid in terms of the ALDH activity in rat liver homogenate among the treatment amino acids, which was comparatively higher than that of positive control CP. It was chosen for further experiments and yeast ADH activity increased in parallel with increased methionine concentration, but not rat liver ALDH activity, and it was comparatively higher than those of the positive control. Arginine showed the highest values in yeast ALDH and rat liver ADH activities among amino acids, and it was chosen for further experiments. Yeast ALDH activity increased in parallel with increased arginine concentration, which was higher than that of positive control CP, and rat liver ADH activity was also comparatively higher in all treatment concentrations of arginine than that of positive control CP. The native electrophoresis of ADH and ALDH from cell-free extracts of yeast Saccharomyces cerevisiae cultured in the growth medium containing various arginine concentrations by 0~0.1% showed two active bands upon zymogram staining analysis, and the straining intensity of ADH and ALDH active bands in arginine treatment yeast was stronger than that of non-yeast or low treatment yeast. These results indicate that alcohol metabolizing enzyme activities can be enhanced by arginine and methionine, suggesting that arginine and methionine have potent ethanol-metabolizing activities.

In engineering design, metamodel has been widely used to improve the efficiency of optimization. However, global metamodels of constraints in constrained optimization is required good accuracy around near optimum point. To satisfy this requirement, more sampling points must be located around the boundary of feasible region or inside of it. Therefore, a new sampling strategy that is capable of defining feasible domain should be applied to select sampling points satisfying constraints. In this research, penalty function method is employed to locate sampling points within feasible domain. For this reason, the proposed sampling strategy is referred to as feasible domain sampling. To validate the excellence of feasible domain sampling, the results of the proposed method are compared with those of conventional global space-filling sampling for a variety of optimization problems. The advantages of the feasible domain sampling are discussed based on optimization results.

For optimal design of a deep-sea ocean mining collector system,based on self-propelled mining vehicle, it is imperative to develop and validate the dynamic model of a tracked vehicle traveling on soft deep seabed. The purpose of this paper is to evaluate the fidelity of the dynamic simulation model by means of response surface methodology. Various statistical techniques related to response surface methodology, such as outlier analysis, detection of interaction effect, analysis of variance, inference of the significance of design variables, and global sensitivity analysis, are examined. To obtain a plausible response surface model, maximum entropy sampling is adopted. From statistical analysis and prediction for dynamic responses of the tracked vehicle, conclusions will be drawn about the accuracy of the dynamic model and the performance of the response surface model.
크리깅 모델의 평균과 분산을 이용한 모델의 정확도 평가법

To validate rigorously the accuracy of metamodel is an important research area in metamodel techniques. A leave-k-out cross-validation technique not only requires considerable computational cost but also cannot measure quantitatively the fidelity of metamodel. Recently, the average validation technique has been proposed. However the average validation criterion may stop a sampling process prematurely even if kriging model is inaccurate yet. In this research, we propose a new validation technique using an average and a variance of response during a sequential sampling method, such as maximum entropy sampling. The proposed validation technique becomes more efficient and accurate than cross-validation technique, because it integrates explicitly kriging model to achieve an accurate average and variance, rather than numerical integration. The proposed validation technique shows similar trend to root mean squared error such that it can be used as a strop criterion for sequential sampling.

Double-deck train has been attracted growing attention as next generation transportation around metropolis because of high passenger carrying capacity. In order to develop high-speed double-deck train with low operational costs, the carbody must be designed as light as possible. In addition, the carbody must be strong enough to ensure the safety of passengers. For these purposes, weight minimization that determines thickness of aluminum extruded panels(AEP) of the carbody can be performed. First, we perform the screening process that select sensitive or/and important design variables through design exploration because the design variables of carbody are too many to treat all of them in the design optimization. Then, weight minimization is accomplished under multi-loading condition such as vertical, compressive and torsional load, while satisfying user specified strength constraints. Finally, the result of design optimization is discussed by comparison with it of initial design.
