The aqueous and methanolic extracts of 110 crude drugs were screened for hyaluronidase inhibitory activity using a microplate assay. Among them, MeOH extract of 15 crude drugs inhibited more than 80% of hyauluronidase activity at the concentration of 5mg/ml. The active principles of Anemarrhenae Rhizoma, Rhei Rhizoma, Ephedrae Herba, Pteropi Faeces and Ginseng Radix alba were transferred into organic solvents.

Hyaluronidase is one of the mucopolysaccharide-splitting enzyme and is related to the permeability of the vascular system and inflammation. An anti-hyaluronidase assay guided fractionation of the methanolic extract of Uncariae Ramulus et Uncus has furnished a pentacyclic triterpene, ursolic acid (compound I). Compound I exhibited hyaluronidase inhibitory activity with $IC_{50}$ value of 0.15 mM, and disodium cromoglycate showed the inhibitory activity with $IC_{50}$ value of 1.78 mM as a positive control.

From the 60% aqueous methanolic fraction of Moutan Cortex Radicis two hyaluronidase inhibitors were isolated and their structures were elucidated by spectroscopic methods. Their structures were identified as paeoniflorin (com-pound I) and oxypaeoniflorin (compound II). Compound I and II exhibited hyaluronidase inhibitory activities with $IC_{50}$ of 1.71 and 1.73mM, respectively.

From the ethyl acetate fraction of the methanol extract of the fruits of Opuntia ficus-indica var. saboten, two dihydroflavonois were isolated and identified as (+)-trans-dihydrokaempferol (1) and (+)-trans-dihydroquercetin (2) by spectroscopic methods.

One hundred and seventeen crude drugs were screened for DNA-binding activity in vitro. The DNA-methyl green assay is a useful biochemical screen for the detection or development of biologically active compounds which bind to DNA. This assay is based upon the fact that free methyl green undergoes rapid spontaneous molecular rearrangement to its colorless carbinol base so that the liberation of the dye from DNA by displacement can be followed spectrophotometrically as a decrease in absorbance at 65Onm. Seven methanolic extracts of crude drugs including Cinnamomi Cortex spissus, Coicis Semen, Coptidis Rhizoma, Perillae Semen, Plantaginis Semen, Polygalae Radix and Zanthoxyli Fructus showed less than 2,000mcg/ml as their IC50 values. The active principles of Plantaginis Semen and Zanthoxyli Fructus were transferred into organic solvents, which showed the IC50 values with 588 and 574mcg/ml, respectively. These fractions have been selected for isolation of biologically active constituents.

Effects of the aqueous extract of Aquilariae Lignum (Thymelaeaceae) on the allergic reactions were investigated. Oral administration of this extract (50, 250, and 500mg/kg) exhibited a dose-dependent inhibition on passive cutaneous anaphylactic reactions in rats. Administrations of this extract (500mg/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) decreased the mortality rates to 0, 0, and 14.2%, respectively. The aqueous extract of Aquilariae Lignum (0.05 ~ 1.6mg/ml) showed a dose-related inhibition on histamine release from rat peritoneal mast cells. The morphological examination also clearly showed that the aqueous extract of Aquilariae Lignum prevented the degranulation of mast cells in rats.
