Gentamicin sulfate (GS)-loaded poly(L-lactide-co-glycolide) (PLGA) wafers and powders were prepared by direct compression method and melting extrusion method for the sustained local antibiotic delivery, respectively. We have investigated the effects of initial drug loading, wafer size, amount of NaCl and PVP as water soluble additives and powder size. The in vitro release of GS were analyzed by high performance liquid chromatography (HPLC) and the morphology of GS/PLGA wafers and powders were examined with scanning electron microscope (SEM) before and after in vitro release test for the explanation of biodegradation mechanism of GS/PLGA wafers and powders. The increasing initial drug and additives loading amounts led to the increase of GS release. Also, the small size of powder affected GS release. From this results, the release profiles from the GS/PLGA wafers and powders could be controlled by varying preparation conditions such as initial drug loading, additives type and amounts, wafer size and powder size.
