Objective : This experimental studies were performed in order to prove the effect of Hirudin Herbal-acupuncture by using rats that had neuronal damage due to the Middle Cerebral Artery Occulsion(MCAO). Method : We observed the change of extracellular concentrations(${\mu}M$) of dopamine, DOPAC, HVA, HIAA, glutamate, aspartate, GABA, glysine, taurine, alanine, and tyrosine as extracted by vivo microdialysis, in the Hirudin Herbal-acupuncture administrated rats($240{\sim}260g$, Sprague-Dawley) subjected to the MCAO. The dialysates were extracted three times before the MCAO and six times after the MCAO every 20 minutes, and analysed by highperformance liquid chromatography(HPLC). Results : Hirudin Herbal-acupuncture significantly inhibited glutamate, aspartate, and tyrosine which are stimulant neurotransmitters at brain ischemia, and it significantly decreased glycine, GABA, taurine, and alanine which are inhibitory neurotransmitters at brain ischemia. Conclusion : Hirudin Herbal-acupuncture may prevent delayed neuronal death(DND) in selectively vulnerable focal areas of the brain effectively.
Bee Venom 및 Melittin 약침액(藥鍼液)이 신경아세포종(神經芽細胞腫)의 세포사(細胞死)에 미치는 영향(影響)

Objective : This study was designed to analyze the effects of bee venom and melittin on cell death in neuroblastoma cell line. Methods : MTT assay, morphologic method, DNA fragmentation, flow cytometry, immunocytochemistry analysis, RT-PCR and Western blot were performed. Results : The obtained results are summarized as follows: 1. The MTT assay demonstrated that neuroblastoma cell viability was significantly inhibitted dose-dependently by treatment with bee venom and melittin in comparison with control. 2. Cell culture demonstrated that control group proliferated highestly at he 5th day in comparison with the 4th day in bee venom and melittin group. And in bee venom and melitti group cell proliferation decreased 2.5 times than control group. 3. The morphologic study demonstrated that neuroblastoma cell showed apoptosis after treatment with bee venom and melittin for 6 hours using microscope. 4. The Flow cytometry demonstrated that apoptosis of neuroblastoma cell treated with bee venom and melittin was related with stop of cell cycle in stage of $G_0/G_1$. 5 .DNA fragmenation demonstrated that neuroblastoma cell treated with bee venom and melittin showed DNA ladder below 1 Kbp. 6. Immunocytochemistry assay demonstrated that Fos and MAPK which are related with cancer were down-regulated by treatment with bee venom and melittin. 7. RT-PCR analysis demonstrated that Fos and MAPK mRNA were transcripted. Fos was down-regulated form treatment with $5{\mu}g/ml$ bee venom and MAPK was down-regulated form $1{\mu}g/ml$ bee venom. 8. Western blot demonstrated that Fos was down-regulated from $1{\mu}g/ml$ bee venom whereas MAPK was expressed by $1{\mu}g/ml$ bee venom but down-regulated by $10{\mu}g/ml$ bee venom. Conclusions : We found that some cancer related genes ware down-regulated by treatment with bee venom and melittin. Further study is needed for investigating the anti-cancer effect of bee venom and melittin.
배합비율(配合比率)에 따른 당귀보혈탕(當歸補血湯) 약침(藥鍼)이 빈혈백서(貧血白鼠)에 미치는 영향(影響)

Objective; This study was investigated to test the anti-anemic effects of Danggwibohyeol-tang for herbal acupuncture on rat with anemia induced by phenylhydrazine according to composition ratio. Methods; Danggwibohyeol-tang herbal-acupuncture was administered into Kyoksu(膈兪穴, BL17) and RBC, Hemoglobin, Hematocrit, MCV, MCH, MCHC, RDW, platelet, MPV, PDW, indirect bilirubin and lactate dehydrogenase were tested. Results; The obtained results are summarized as follows: (1) In control group RBC was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group. The effect of Danggwibohyeol-tang group C was similiar to that of Danggwibohyeol-tang group B and was higher than that of Danggwibohyeol-tang group A. (2) In control group hemoglobin was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group but there was no significant difference between Danggwibohyeol-tang group A, B, C. (3) In control group hematocrit was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group. The effect of Danggwibohyeol-tang group C was higher than that of Danggwibohyeol-tang group A or B. (4) In case of MCV, MCH, MCHC and RDW there was no significant anti-anemic effect. (5) Danggwibohyeol-tang group significantly inhibited the growth of platelet. In case of Danggwibohyeol-tang A and B group there was more inhibitory effect than Danggwibohyeol-tang C group but there was no significant difference between Danggwibohyeol-tang A and B group. (6) In case of MPV, PDW and indirect bilirubin there was no significant difference between Danggwibohyeol-tang group and control group. (7) Danggwibohyeol-tang group significantly inhibited the growth of lactate dehydrogenase. In case of Danggwibohyeol-tang C group there was more inhibitory effect than Danggwibohyeol-tang B group and Danggwibohyeol-tang B group showed more inhibitory effect than Danggwibohyeol-tang A group. Conclusion; From above results Danggwibohyeol-tang for herbal acupuncture showed significant anti-anemic effect in some parameters. Further study is needed for the effect of Danggwibohyeol-tang according to different composition ratio.

Objective : This study was designed to find out whether NS(normal saline) is able to be constituted as an appropriate control group for HHT(Hwangryunhaedoktang herbal acupuncture) and HP(Homnis Placenta herbal acupuncture). Methods : NS and two herbal acupuncture were inserted into Quchi(LI 11) of the subjects. After 5 minutes the subjects completed a questionnaire rating the intensity of 21 kinds of acupuncture sensation; hurting, penetrating, sharp, aching, intense, spreading, radiating, tingling, pricking, stinging, pulling, heavy, dull, numb, electric, shocking, hot, burning, cool, pulsing, and throbbing. We compared the subjective acupuncture sensation between the two groups. Results : As for HHT seven items of the acupuncture sensation were significantly different from NS. In case of HP just two items were significantly different from NS. Conclusion : We found that NS may be able to be an placebo herbal acupuncture for HP. Further study is needed for the appropriate placebo herbal acupuncture.

Objectives: In order to prove the effect of Phospholipase A2(PLA2) Herbal-acupuncture, this experimental studies were performed by using rats that had neuronal damage due to the Middle Cerebral Artery Occulsion(MCAO). Methods: Microdialysis probes were implanted into the coordinate of striatum of anesthetized rats which consist of sham-operated 8 rats, MCAO-operated 8 rats and PLA2 Herbal-acupuncture administrated 8 rats before MCAO operating. The PLA2 Herbal-acupuncture(0.5mg/kg) was administrated to rats 30 minutes before having an operation causing the MCAO. The surgical excision lead the cross resected brain to the acute ischemic state. The brain was sliced in 2㎜ thickness and stained with cresyl violet buffer for the measurement of cerebral infarcted area and volume. Results: Based on the result of the tissue inspection for the cerebral ischemic cell, PLA2, Herbal-acupuncture significantly protect neurocytes. Conclusions: We suggest PLA2, Herbal-acupuncture produces protective effects against the neuronal damage induced by MCAO. Therefore, PLA2, Herbal-acupuncture may prevent delayed neuronal death(DND) in selectively vulnerable focal areas of the brain effectively.
급성 좌섬요통 환자에 대한 습식 부항요법과 건식 부항요법의 비교

Objectives : This study was designed to compare depletion and cupping therapy with cupping therapy on acute lumbosacral strain patients. Methods : This study was carried out on the 36 acute lumbosacral strain patients. We divided these patients into 2 groups. The GroupⅠ was treated with the depletion and cupping therapy and the GroupⅡ was treated with the cupping therapy. We evaluated by visual analogue scale(VAS) score after each treatment and Oswestry disability index(ODI) score before and after treatment. Result : The VAS score of GroupⅠ was statistically lower than GroupⅡ after 1st treatment. But there was no significant difference between two groups after 2st treatment. In each group, the VAS score and ODI score was statistically decreased after treatment. Conclusions : In this study, depletion and cupping therapy was more effective than cupping therapy after 1st treatment, but there was no difference between two kinds of cupping therapy after 2nd treatment on acute lumbosacral strain patients.
