본 연구에서는 Landrace, Large White와 Duroc 3품종의 3원 교잡 시스템으로 비육돈을 생산하는 9개 농가를 대상으로 Landrace와 Large White 교배를 통해 생산된 F1 모돈과 Duroc 웅돈 그리고 비육돈을 이용하여 기 보고한 바 있는 돼지 이력추적 및 브랜드육 식별을 위한 13종의 MS marker의 개체판별능과 혈연관계 추정 효율을 실증하였다. 우선 F1 모돈과 웅돈 즉 부모 집단을 대상으로 API-CALC version 1.0 프로그램을 이용하여 무작위 교배집단, 반형매 교배집단 그리고 전형매 교배집단으로 가정 시 개체 판별능이 각 각 4.94×10(-34), 8.16×10(-23) 그리고 2.01×10(-08)으로 추정되었으며, 비육돈을 포함한 추정치는 3.74×10(-35), 5.48×10(-25) 그리고 2.96×10(-11)으로 추정되었다. 또한 Cervus version 2.0을 이용하여 친자감별률을 추정한 결과 100% 인 것으로 추정되었다. 이론적으로 산출된 상기의 수치들을 검증하기 위해 PAPA version 2.0 프로그램을 이용하여 비육돈 전체 452두에 대한 친자감별을 실시한 결과 100% 친부모를 확인 할 수 있었으며, Cervus 프로그램을 이용하여 전체 축군에서 동일한 대립유전자형을 가진 개체의 출현을 조사한 결과 동일개체는 존재하지 않는 것을 확인하였다. 비록 개체 판별능에 대한 실증은 제시한 이론적 판별능에 상응하는 두수를 검증하지는 못하였으나, 친자확인의 경우는 제시한 이론적 효율성 100%는 실증적으로 검증되었다. 따라서 현재 국내에서 행해지는 3 품종 교잡에 의한 비육돈 생산 시스템의 경우 본 연구의 결과로 비추어 볼 때 13 MS marker를 이용하여 체계화된 전산정보와 병행하여 계열화된 생산체계하의 브랜드 단위 또는 지역별 권역으로 구분하는 이력추적이 충분히 가능하다고 사료된다. Using the materials collected from nine farms in a three-way cross system to produce commercial pigs produced from F1 sows (Landrace×Large White)×Duroc, the power of individual discrimination and parentage of the 13 microsatellite(MS) marker set that has been suggested for individual/brand identification(traceability) was empirically tested. Initially, genotypes of the parental population(F1 sows and Duroc), and commercial pigs were determined and the genotype frequency and polymorphic index were estimated using the Cervus 2.0 program. The probability of identity among genotypes of random individuals, that random half sibs and that of full sib individuals, based on the genotypes from 91 F1 sows and Duroc were expected to be 4.94×10(-34), 8.16×10(-23) and 2.01×10(-08), respectively, using the API-CALC version 1.0 program. When commercial pigs were included, the estimates increased to 3.74×10(-35), 5.48×10(-25) and 2.96×10(-11), respectively. For the empirical verification of the estimated powers of individual discrimination and parentage, the parentage test was performed for 452 commercial pigs using PAPA version 2.0, and individuals with the same genotype were investigated using the Cervus version 2.0 program. Parents for all commercial pigs were successfully estimated and no identical individual was identified in the pedigree. Although the individual discriminating power was not fully verified because of the lack of individuals corresponding with the theoretical power, the 100% efficiency of parentage test was clearly confirmed. Therefore, we believe that the 13 MS marker set in conjunction with management record/information for the pig production kept in a farm/brand should be useful in the pork traceability in a brand unit.

The aims of the present study were to estimate the possibility for toxicity test and compare acute toxicity of potassium dichromate in the larva stage of Neocaridina denticulata, Daphnia magna and the juvenile stage of Oryzias latipes. N. denticulate, a freshwater shrimp lives in Korea, is an indigenous species and considered to be useful for toxicity test. D. magna and O. latipes were recommended as a test species for the OECD test guideline. The 96 h-LC50 potassium dichromate value was 0.62 mg L^-1 for the larva stage of N. denticulata and 168.44 mg L^-1 for the juvenile stage of O. latipes. The 48 h-EC50 value was 1.27 mg L^-1 for the D. magna. The study was confirmed higher sensitivity of the larva stage of N. denticulata to potassium dichromate compared to the D. magna and the juvenile stage of O. latipes.

쥐의 mesangial 세포를 초회 배양하여 Transforming Growth Factor-$\beta$(TGF-$\beta$)가 세포의 복제 및 collagen 합성에 미치는 영향을 조사하였다. TGF-$\beta$는 사람 혈소판에서 산성 에탄올 침전법 및 FPLC를 이용한 젤 크로마토그래피의 방법으로 정제되었다. TGF-$\beta$가 mesangial 세포를 복제 및 collagen 합성에 미치는 영향은 TGF-$\beta$ 농도와 세포의 밀도에 따라 달라졌다. Confluent 세포의 경우 0.1~15 ng/ml의 농도에서 TGF-$\beta$는 mesangial 세포의 복제를 35%까지 감소시켰다. Collagen 합성은 0.5 ng/ml의 TGF-$\beta$ 농도에서는 2배로 증가되었으나 그 이상의 농도에서는 감소하기 시작하는 두 가지의 조절 양상을 보였다. 그러나 subconfluent 세포의 경우에는 세포의 복제 빛 collagen 합성 모두에 TGF-$\beta$는 아무런 효과를 나타내지 못하였다. 본 연구결과는 TGF-$\beta$가 glomerulosclerosis에 있어서 중요한 역할을 하리라는 가능성을 제시해주고 있다. The effect of TGF-$\beta$ on cell replication and collagen synthesis in primarily cultured mesangial cells from rats was studied in vitro. The TGF-$\beta$ was purified from human platelets by acidic ethanol extraction and gel filtration with FPLC. The effect of TGF-$\beta$ on the DNA synthesis and the collagen synthesis in mesangial cells depended on both TGF-$\beta$ concentration and cell density in monolayer culture. Concentrations of TGF-$\beta$ ranging from 0.1~15 ng/ml were evaluated for their effects upon DNA synthesis. In confluent cells, when compared to the growth rate in standard medium, TGF-$\beta$ concentrations in the range 0.1~15 ng/ml reduced a cell proliferation up to 35%, suggesting inhibition of cell division. The effect of TGF-$\beta$ on collagen synthesis in mesangial cells was biphasic. In confluent cells, the addition of TGF-$\beta$ induced a 2-fold increase in the production of collagen when compared with cells grown in standard medium. The maximal effect of TGF-$\beta$ occurred at 0.5 ng/ml. At higher concentrations there was a decrease in the action of TGF-$\beta$ with the effects on collagen production. In subconfluent cells, however, TGF-$\beta$ showed no significant effects on the DNA synthesis and the collagen synthesis at all concentrations. These results support that TGF-$\beta$ may play an important role as a multifunctional regulator in the pathogenesis of glomerulosclerosis.

Purpose: The purpose of this study is to evaluate the clinical results of 23-gauge sutureless vitrectomy. Methods: We retrospectively analyzed 111 eyes that had undergone 23-gauge sutureless vitrectomy. The eyes underwent surgery from February 2006 to July 2006 and were examined for surgical outcome as well as intraoperative and postoperative complications. Results: The preoperative best corrected visual acuity (BCVA) was 1.43±0.12 and increased to 0.88±0.11 (P>0.01). Thirteen (11.7%) of 111 eyes developed intraoperative complications, and 21 (18.9%) of 111 eyes developed postoperative complications. Intraoperative complications included wound‐leaking (seven eyes), conjunctival suture (three eyes), and difficult insertion (two eyes). Postoperative complications included postoperative hypotony (seven eyes), vitreous hemorrhage (seven eyes), hyphema (five eyes), and infectious endophthalmitis (two eyes). Conclusions: Unlike 20-gauge vitrectomy, sutureless 23-gauge vitrectomy offers all the advantages of minimally invasive sutureless vitrectomy. J Korean Ophthalmol Soc 49(6):911-916, 2008

Reconstruction of subtotal maxillectomy defects with columella deficit is challenging. We report a unique case of facial reconstruction using a free radial forearm flap and a free preauricular flap for the maxillectomy and columella deficit. A 73-year-old woman was diagnosed with recurrent sebaceous carcinoma of the nose. We performed wide excision, including areas of the right cheek, nose, upper lip, maxilla, and columella. The resultant subtotal maxillectomy defect was reconstructed using a three-dimensional flap. First, a free radial forearm flap was transfered to resurface the nasal, oral, and external facial side. Second, a preauricular flap was positioned into the columella defect and anastomosed with the distal portion of the radial forearm flap pedicle. The two flaps survived, and the patient recovered uneventfully. We believe the radial forearm and preauricular double free flaps with the pedicle connection method were effective in reconstructing the present case of subtotal maxillectomy defect.
