All muscle movements including breathing, walking, and fine motor skills rely on the function of the spinal motor neuron to transmit signals from the brain to individual muscle groups. Motor neuron diseases (MNDs) result from the progressive degeneration and deat of motor neurons. The two most studied MNDs are spinal muscular acrophy (SMA), a childhood genetic disease, and amyotrophic lateral sclerosis (ALS), a adult-onset neurodegenerative disease. Both diseases involve neuromuscular dysfunction progressively leading to fatal paralysis. The potential of human pluripotent stem cells to treat patients with neurodegenerative disease is enormous. Stem cells can be introduced into clinical applications in several different ways, such as disease modeling, drug screening, and cell replacement therapy. Even though it may take quite a long time to address some problematice, the establishment and optimization of human stem cell differentiation protocols for clinical applications may hold the key to cure diverse disorders.

Mycoplasmas are smaller than bacteria (0.3-0.8 μm in diameter) and can be observed as filamentous or coccal forms. They may remain undetected by microscopic observation but can cause a reduction of growth rate, morphological changes, chromosomal aberrations, induction or suppression of cytokine expression, a change in membrane composition and alterations in the amino acid and nucleic acid metabolism of infected cells. In this study, we tried to remove the mycoplasma from infected human embryonic stem cells (hESCs) by combinational antibiotics treatment. In case of only moxifloxacin treatment, mycoplasma-specific gene was still detected in cultured hESCs. However, in case of double treatment of moxifloxacin and genramicin, mycoplasma-specific gene was removed completely. We confirmed normal karyotype from the double-treated hESCs by karyotype analysis. Our results suggest a stable and simple method for mycoplasma treatment in hESC culture.

Adult mesenchymal stem cells (MSCs) are self-renewable adherent stromal cells and differentiate into variable cell types. MSCs can be obtained from distinct tissue sources and have turned out to be successfully manipulated in vitro. It is now accepted that stem cells reside within the post-natal dental pulp. These postnatal populations also have MSC-like characteristics including the self-renewal and multilineage differentiation potential. Characterization of these cells and determination of their potential in terms of specificity of regenerative response may help direct new clinical treatments. Such approaches may provide innovative and novel tools for generation of clinical materials and/or treatments. This study aimed to review the possibility of dental stem cells and related technology for their future clinical applications in dental disease.

Parkinson`s disease (PD) is the second most prevalent neurodegenerative disease, next to Alzheimer`s disease, affecting approximately 0.5% of 65-69 year old adults. PD is a characterized by a progressive degeneration and loss of dopaminergic neurons within the substantia nigra pars compacta, leading to the classical symptoms of the disease, such as tremor, rigidity, and hypokinesia. Stem cells have a high capacity for self-renewal and differentiation potential and some of them can be differentiated into dopaminergic neurons on specific conditions. However, there are some postulations for clinical application of the stem cell-derived dopaminergic neurons. The stem cells grafts should exhibit a regulated release of dopamine and molecular, electrophysiological, and morphological properties similar to those of substantia nigra neurons, reestablish the dopamine network within the striatum and restore the functional connectivity with host extra-striatal neural circuitries and produce no adverse-effects such as tumor formation, immune reactions and graft-induced dyskinesias. In this review, we described the advantages and disadvantages of each characeteristic of variable stem cells with regard to its potential use for PD treatment.

Human pluripotent stem cells including human embryonic stem cells and induced pluripotent stem cell can potentially be used for cell replacement therapies after the differentiation into various cell types although some remaining risk of cell therapeutics, clinical trials for pluripotent stem cell-based therapies is proceeding carefully. At present, applicable cell types and disease are limited. However, expanded clinical application is expected in the next few years. In this review, we will introduce a current state of ongoing clinical trials and refer to current and future perspectives for cell replacement therapy using human pluripotent stem cells.

Parkinson`s disease (PD) is a neurodegenerative disorder involved motor symptom of the central nervous system and characterized by progressive and selective loss of dopaminergic (DA) neurons in the midbrain substantia nigra. Some drugs such as levodopa and surgical procedure including deep brain stimulation are used for PD treatment. However, current treatments are not enough to restore the damaged brain and have some side effects. Recently, cell replacement therapy is considered an alternative approach for PD treatment. In this review, we will refer to current clinical trial for cell replacement therapeutics of the PD.

Stem cells are good cell sources for cell replacement therapy of degenerative or incurable disorders. However, there are some limitations such as immune response, differentiation efficiency, safety and ethical issues for clinical application. These barriers must be considered thoroughly from starting point of the investigational study. For clinical application and commercialization of stem cell-derived cell therapy products, strict preparation is required for formal approval of regulatory body at each stage of the clinical process. This preparation process can be another barrier and challenge to the researcher or developer related to stem cell study. However, These challenges will become increasingly important to stem cell research as clinical translation progresses and helpful for birth of authentic novel cell therapy products. In this brief review, we will suggest more effective strategy of the stem cell research and development and introduce the clinical process for cell therapy products briefly.
