ERIC(enterobacterial repetitive intergenic consensus) 염기서열과 REP(repetitive extragenic palindromic) 염기서열의 생물학적 기능과 다양한 세균 균주들의 분자생물학적 동정에의 응용에 관하여 고찰하였다. ERIC 혹은 REP 염기서열은 염기서열 그 자체는 진화론적으로 상당히 보존되었으나 그들의 염색체상의 위치는 세균 종마다 다르다. 그러므로, ERIC 혹은 REP 염기서열과 상보적인 PCR용 primer를 고안하여 연쇄중합 반응을 시키면, ERIC 혹은 REP 염기서열 사이로부터 증폭된 크기가 다른 PCR 생산 DNA단편들이 만들어진다. 이때 증폭된 크기가 다른 DNA 단편들을 이용하여 각 세균 균주마다 고유한 DNA fingerprinting pattern이 결정될 수 있다. 그렇게 결정된 pattern들을 서로 비교하여 검체 균주간의 상보성과 유전학적 연관성 등이 결정될 수 있다. 그러므로 ERIC 혹은 REP 염기서열을 이용한 PCR반응은 식품으로부터 분리될 수 있는 다양한 세균 균주의 동정에 이용될 수 있는 최근에 개발된 새로운 분자생물학적 기법이다. A family of interspersed repetitive DNA elements in prokaryotic genomes, emphasizing on enterobacterial repetitive intergenic consensus (ERIC) sequences and repetitive extragenic palindromic(REP) sequences, have been discussed in terms of their biological functions and their applications on genotyping diverse bacterial strains. ERIC or REP sequences are highly conserved at the nucleotide sequence level, but their chromosomal locations differ between species. Several features of ERIC sequences resemble those of REP sequences although the nucleotide sequence is entirely different. ERIC or REP sequences can be used as oligonucleotide primer binding sites for polymerase chain reaction (PCR)-mediated genomic fingerprinting. By using outwardly facing oligonucleotide primers complementary to ERIC or REP sequences. PCR reactions can amplify differently sized DNA fragments consisting of sequences lying between elements. Consequently, the multiple amplicons of different sizes can be used to establish DNA fingerprinting patterns specific for individual bacterial isolates. The established patterns can be compared to estimate relative degrees of similarity between isolates and to determine whether isolates are clonally related. Therefore, the ERIC or REP sequence-based PCR fingerprinting can be a powerful tool to discriminate diverse foodborne bacterial isolates from each other.
