        소유적 관계가 자아형성에 미치는 부정적 영향: 『빌러비드』를 중심으로

        The purpose of this paper is to analyze the negative influence of possessive relationship on forming personal identity in Tony Morrison’s Beloved. Dividing into two parts, one examines slavery’s impact on black people’s identity, and the other that of possessive love on other’s self, not true love. First, black peoples’ self was broken completely under the slavery, for it regarded black slave as possessions, not as human being. In the mean while, Sethe, the main character of this novel, misunderstands possessive love as “thick,” which is due to the slavery. Accordingly, she considers it better to die than to live as a slave, so she commits infanticide of her own baby, Beloved. With this prejudice, Sethe leads a very lonely life alienated from her community. In conclusion, Possessiveness, whether slavery or love, is a will to control the others, preventing their character from growing. Also those who try to possess others may be ruined by their prejudice and arrogance. (Chungnam National University)

        효율적인 유전자 전달을 위한 Transferrin과 PEI가 도입된 O-carboxymethyl Chitosan의 합성

        본 연구에서는 키토산의 유전자 전달 효율과 암세포 표적효율을 향상시키기 위하여 OCMCh의 amine group과 carboxyl group에 각각 표적물질인 TF와 양이온고분자인 PEI를 도입하여, 유전자 전달체인 POPT를 제조하였다. 제조된 POPT는 1 H-NMR 을 통해 성공적으로 합성되었음을 확인하였고, DLS를 통해 입자크기를 측정한 결과 POPT의 비율이 증가함에 따라 입자크기가 작아지며, POPT가 나노 크기의 입자를 형성함을 확인하였다. 또한 전기영동 실험을 통해 POPT가 pEGFP-N1와 복합체를 잘 형성하며, heparin 실험결과 혈액 내에서 pEGFP-N1를 잘 방출할 수 있으며, DNase로부터 pE GFP-N1를 보호할 수 있음을 확인하였다. POPT의 유전자 전달효율은 정상세포와 암세포를 사용하여 확인한 결과 POPT 가 증가할수록 유전자 전달 효율이 향상되며, TF의 표적능력으로 인해 암세포에서만 유전자가 전달됨을 확인할 수 있었다. 이 같은 실험 결과들을 통해 POPT는 암세포만을 표적해 유전자를 전달할 수 있는 전달체로써 사용 가능성이 있는 우수한 물질로 사료된다. 추후에 수행할 실험에서는, 이러한 시스템의 가능성을 보다 더 정확히 확인하기 위하여 동물실험을 진행할 예정이며, 질병을 치료할 수 있는 유전자를 전달체를 통하여 전달하는 실험을 통하여 유전자 치료를 수행할 수 있는지 여부를 확인하는 연구를 진행할 것이다. The polyethyleneimine / O-carboxymethyl chitosan / polyethyleneglycol / transferrin (POPT) copolymer was evaluated as nonviral gene delivery carrier. The structural analysis of POPT copolymer was confirmed by 1-proton nuclear magnetic resonance ( 1 H-NMR). The particle size of - pEGFP-N1/POPT polyplexes was confirmed by dynamic light scattering (DLS). The results of DLS showed that the particle sizes of pEGFP-N1/POPT polyplexes has small size of 130~260 nm. The binding ability with pEGFP-N1, pEGFP-N1 protecting assay against DNase, and pEGFP-N1 release assay through heparin of POPT were evaluated by gel retardation assay. In addition, gene transfection effect of pEGFP-N1/POPT polyplexes was evaluated in fibroblast cell line (L929, without transferrin receptor) and cervical cancer cell line (HeLa-S3, with transferrin receptor) cells. The results of transfection were shown that green fluorescence was cancer cell more expression than normal cell due to receptor mediated endocytosis between transferrin of POPT and transferrin receptor on the cancer surface. Therefore, pEGFP-N1/POPT polyplexes could be a promising gene delivery system.

        토니 모리슨의 『재즈』: 제목 적합성 연구

        토니 모리슨의 『재즈』에는 제목 이외에 재즈라는 단어가 전혀 나타나지 않고 다만 이미지와 메타포로써 재즈형식이 적용되었다. 이것은 흑인들이 재즈시대를 통과하면서도 과거 남부에서의 고통과 함께 현재 도시에서의 좌절과 소외감으로 인해 재즈음악에 대해서 알지 못하는 고난의 삶을 역설적으로 강조하려는 전략이다. 이러한 『재즈』의 집필 목적은 미국역사에서 제외된 흑인들의 할렘 이주역사를 복원하려는 것이다. 그런데 기록으로 정확하게 남아있지 않은 역사이기 때문에 이야기를 전달하는 장치로 믿을 수 없는 화자가 등장한다. 그래서 독자는 화자의 서술을 의심하면서 작품에 참여하게 되고 이러한 참여는 재즈의 즉흥적인 변주에 해당된다. 그리고『재즈』는 다른 작품에서는 찾아볼 수 없는 독특한 형식의 장으로 구성되어 있다. 숫자나 제목 없는 장과 장은 빈 페이지로 나뉘고 단어와 내용이 반복되면서 확장, 연결되어 선창-응창하는 재즈형식과 동일하다. 재즈는 메타포로도 적용되었는데 흑인들이 과거의 비참한 삶에서 벗어나 조화로운 공동체를 이루어 자아를 찾아가는 삶의 발달로 나타난다. 이처럼 『재즈』는 재즈형식의 화자, 장 구성, 메타포로 구성되어 재즈시대의 역사임을 미학적으로 표현하고, 내용에 재즈를 사용하지 않아 할렘 르네상스 시기 흑인들의 고통스런 당시 상황을 강조한다. The purpose of this paper is to examine the rightness of the title Jazz by Toni Morrison. In the novel, the word ``jazz`` doesn`t appear literally except the title, but jazz itself is mare in the setting of the book as image and metaphor. The text, divided into unnumbered sections, separated by blank pages, is structured as jazz itself through repetition, amplification, and variation. The narrator is neither omniscient nor reliable, and asks the readers to participate in the book, reflecting the improvision and unpredivtability of jazz. Likewise, the characters are evolving, being entirely different from the conclude, jazz is in the background of the book, implying the jazz age where the black people were experiencing aspiration, disillusion, and loss. Jazz is concerned with African history of the Jazz Age, the presences of which was denied in the American history. Escaping poverty and violence from the rural South even after slavery, black people migrated into the Northern city longing for a better life. However, they lost themselves in the traumatic past and present frustration. Despite that, they recover their identities with communal love and solidarity.

        Histochemical Analysis of the Cutaneous Wound Healing in the Amphibian

        상처회복과정은 염증, 재상피화, 그리고 기질의 재형성등을 포함한 포괄적인 생물학적 반응이다. 생물학적 분류체계에서, 회복능력은 매우 다양한데, 일반적으로 하등동물이 고등동물에 비교해 단순하며 뛰어난 것으로 알려져 있다. 따라서, 하등동물들을 이용한 실험모델은 상처회복과 치유의 기작을 연구하는데 자주 이용되고 있다. 양서류를 이용한 피부 상처회복과정 동안의 조직화학적 특성들을 규명하기 위하여 한국산 무당개구리(Bombina orientalis)를 이용하였다. 1일, 10일 그리고 16일의 조직에서 점액물질이 활발하게 분비되는 것을 확인하였으며, PAS (periodic acid shiff)와 Alcian blue (pH 2.5) 반응에 강하게 염색되었다. PTAH(phosphotungstic acid hematoxylin) 염색을 통하여 상처 후 10일 조직에서 상처주변의 진피층에서 콜라겐의 합성이 증가하는 것을 확인하였다. 또한, 세포의 유사분열의 시점을 확인하기 위한 MG-P (methyl green pyronin)염색에서 초기에는 3시간과 6시간에 강한 염색반응을 관찰하였고, 이는 23일과 27일 조직에서도 동일한 패턴으로 이어졌다. The wound healing is very complex biological processing including inflammatory, reepithelialization and matrix construction. According to the biological systematic category, the ability of the healing is very different. Generally healing ability of the lower animal group has been known more excellent compared to its higher group. Therefore, lower animals have been used as the experimental model to explore the mechanism of the wound healing or repair. To verify histochemical characteristics of the wound healing, we have used skin of the frog (Bombina orientalis) as known common amphibian. At day 1, 10, and 16, the mucous substance was very actively synthesized and strong positive by PAS and Alcian blue (pH 2.5). Day 10 after wounding, margin of the wound was gradually strong positive by PTAH staining for detection of collagen synthesis. At 3 to 6 hour and day 23 to 27, we have found the cell division was active through the MG-P staining, in which the concentration and division of DNA in nucleus was green to deep blue color.

        표적성 pH-sensitive 약물전달을 위한 folic acid와 doxorubicin이 결합된 O-carboxymethyl chitosan의 평가

        In this study, target ligand and anticancer drug were conjugated with O-carboxymethyl chitosan (FPOCM-DOX). Targeting ligand was used folic acid (FA) for specific binding of FA receptor overexpressed cancer cell. Anticancer drug model was used doxorubicin (DOX). DOX was conjugated with 2, 3-dimethylmaleic anhydride (DMMA) (DMMA-DOX) for pH-sensitive drug release. The FPOCM-DOX of chemical structure was analyzed by 1 H-NMR and FT-IR. DLS study that the FPOCM-DOX was self-aggregated to an average diameter of 432 nm. In addition, the antitumor activity of FPOCM-DOX was demonstrated against HeLa cell by MTT assay. The IC50 of OCM-DOX and FPOCM-DOX were confirmed 11.25 μg, 5.6 μg in HeLa cell. The antitumor activity considered that FPOCM-DOX might be increased antitumor activity by receptor mediate endocytosis. The result suggested that FPOCM-DOX might be a promised anticancer drug carrier for cancer therapy.

        인간배아줄기세포 유래 배상체내의 신경세포군 확인과 분화과정 동안 이뤄지는 신경발생과정의 이해

        Recently, human embryonic stem cells(hESCs) have been highlighted to understand early embryogenesis as well as to apply to clinical cell therapy. During human embryogenesis, dynamic processes of early neuronal development are very important to elucidate their developing mechanism. However, the mechanism has not been entirely clear. The formation of embryoid bodies(EBs) derived from hESCs is a tissue-like spheroids in suspension culture and has a potential to differentiate into the three germ layers. Even though many kinds of specific cell makers have been studied, the specific proteins expressed from differentiated cells have been elusive. The objective of this study is to investigate the expression of neuronal protein while EBs is differentiating from hESCs(day 5, 10, 15 and 20). The expressions of neuronal specific makers such as Neurofilament 68kDa(NF 68), tubulin III(TuJ-1) and microtubule-associated protein 2(MAP2) in each EBs stage were analyzed by immunofluorescence(IF). The expressions and localization of neuronal specific markers were shown to be different depending on EBs stages. NF 68 positive cells were expressed on the surface of EBs at day 10 and the expression of NF68 almost disappeared at day 20. MAP2 was not detected in EBs at day 10, whereas it began to be slightly detected on the surface of EBs at day 15. On the other hand, TuJ-1 positive cells were found at all stages and their expression levels were maintained until day 20. These results demonstrate neuronal specific markers can be expressed differently depending on EBs stages, suggesting this allows us to probably understand the early development of neurogenesis in hESCs. Furthermore, this idea expands that we are able to understand the development of all specific cells differentiated from early human embryogenesis.

        Human embryonic stem cell(hESC) can be very valuable source for the cell therapy due to their pluripotency when they differentiated into specialized cell types. Conventionally, differentiated cell population were obtained from spontaneously formed embryoid body(EB) which could contain various cell types. In this study, we described the optimization of culture condition for endothelial precursor cells derived from human embryonic stem cells(hESC-EPCs). First, we compared the expression of endothelial precursor cell markers such as CD133, CD34 and KDR in day 7 hEBs and isolated CD133 and KDR double positive population as hESC-EPCs. Second, we compared the culture medium and ECM coating to optimize culture condition for hESC-EPCs. As the result, we could confirm EGM-2 and collagen coating were optimized condition to culture of hESC-EPCs. After conducting optimized culture condition for hESC-EPCs, we examined the maintenance of characteristics of hESC-EPCs cultured in optimized culture condition as vascular cells. Finally, we could confirm the characteristics of hESC-EPCs cultured in optimized condition as vascular cells were maintained in developed optimized culture condition. Furthermore, the optimized culture condition developed in this study will be useful to culture of other vascular lineage cells derived from hESCs.
