동충하초(冬蟲夏草)가 Hydrocortisone을 투여한 흰쥐의 Nitric Oxide Synthase 활성 및 Testosterone 함량에 미치는 영향

The fallowing are the results of the experimental studies of Cordyceps militaris (CM) on the penile nitric oxide synthase (NOS) activity and the level of blood testosterone in hydrocortisone acetate-treated rats. CM was tested for the effects on activity of xanthine oxidase and lipid peroxidation in penis of hydrocortisone acetate-treated rats. In vitro, CM didn't effect the levels of lipid peroxide and the activity of NOS. In the penis of hydrocortisone acetate-treated rats, lipid peroxide, the activities and ratio of type conversion of xanthine oxidase were increased but activity of NOS and content of nitrite were decreased. In vivo, after administration of CM to hydrocortisone acetate-treated rats, levels of lipid peroxide in penis was decreased. Also, the activities and ratio of type conversion of xanthine oxidase were decreased, too. The body weight and concentration of testosterone in the blood were increased. The effects of Cordyceps militalis Broth did better than the effects of Cordyceps militalls Mycelia, These results suggest that CM decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide and increase the penile NOS activity and the level of blood testosterone in hydrocortisone acetate-treated rats. Conclusively, CM is capable of improving of sexual ability in hydrocortisone acetate-treated rats.

Objective : Peroxynitrite $(ONOO^-)$, formed from the reaction of $O_2^-$ and NO, is a cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA. It has been implicated in the aging process and age-related disease such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate scavenging activities of $ONOO^-$ and its precursors. NO and $O_2^-$ and its scavenging mechanism using fluorescent probes, DCFDA, DAF-2 and DHR 123.. Methods : Psoralea corylifolia was crushed. The crushed Psoralea corylifolia was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under reduced pressured using a rotary evaporator to yield 16g. This was done to investigate scavenging activities of $ONOO^-$, NO, $O_2^-$ and its scavenging mechanism using fluorescent probes, DCFDA, DAF-2 and DHR 123. Results : After Psoralea corylifolia was added authentic $ONOO^-,\;{\cdot}\;O_2^-$ and NO was markedly scavenged. Also, $ONOO^-$ induced by $O_2^-$ and NO (these derived from SIN-1) was inhibited. The data showed a decrease in $ONOO^-$ mediated nitration of tyrosine through electron donation after Psoralea corylifolia was added. Data showed a dose-dependent correlation with inhibition of nitration of bovine serum albumin induced by $ONOO^-$, Furtheremore, LPS-induced ROS and RNS generation was blocked. Conclusions: These results suggest potential for use of Psoralea corylifolia as an effective $ONOO^-$ scavenger to counter the aging process and age-related diseases.

Objectives : Cordyceps Sinensis (CS) was tested for the effects of antioxidant enzymes and lipid peroxidation in the liver. Methods : We measured the changes in body weight, enzyme activity, lipid peroxide and the death rate in the hydrocortisone acetate-treated rats. Results : In vitro, CS didn't effect levels of lipid peroxide. the activities, and the ratio of type conversion of xanthine oxidase. In the hydrocortisone acetate-treated rats, lipid peroxide, the activities, the ratio of type conversion of xanthine oxidase, and the death rate all increased. But, glutathione peroxidase and superoxide dismutase decreased. In vitro, after CS was administered to hydrocortisone acetatetreated rats, the levels of lipid peroxide in the liver, and the death rate decreased. However, the activities, and the ratio of type conversion of xanthine oxidase decreased. The body weight, glutathione peroxidase, and superoxide dismutase in+creased. The effects of Sinensis Cordyceps Broth did better than the effects of Sinensis Cordyceps Mycelia. Conclusions : These results suggest that CS decrease the activities of free radical generating enzymes which form lipid peroxide and increase the activities of oxygen free radical scavenging enzymes.

Objective : his study was carried out to investigate the effect of Dansameum (DS) on the gastrohemorrhagic lesion induced by ammonia in rats. Methods : Rats were pretreated with DS extract 25 mg/kg for 10 days and then were given amonia through gastric tube. The animals were killed 1 hr after amonia treatment. Results : DS extract siginificantly reduced the gastrohemorrhagic lesion score, the gastric lipid peroxide level, the gastric urease activity, the gastric myeloperoxidase activity, the gastric acid phosphatase activity, xanthine oxidase activity and type conversion, and increased the gastric glutathione level considerably. In photomicrographs of stomach tissue in rat, we could see the gastrohemorrhagic lesion induced clearly. conclusions : These results suggest that DS extract may be effective in peptic ulcer.

Objectives: This study was undertaken to investigate the action mechanism of Holotrichia (HT) at parameter related to dementia. Methods: HT was tested for the effects on acetylchonine esterase and monoamine oxidase activities, lipid peroxidation, antioxidation in brain of lead acetate-treated rats. Lead acetate were treated firstly into samples for 10 days, and then lead acetate and HT were set with them for 20 days. Results: The level of lipid peroxide, acetylcholine esterase and monoamine oxidase activities, enzyme activities and ratio of type conversion of xanthine oxidase increased in lead acetate-treated rats were decreased as highly as normal group by HT. Superoxide dismutase, catalase and glutathione peroxidase activities, the level of acetylcholine decreased in lead acetate-treated rats were increased as lowly as normal group by HT. Conclusions : These results suggest that HT might have an effect on treatment of dementia according to decreasing the activities of acetylcholine esterase, monoamine oxidase and level of lipid peroxide in brain.
양생활력단(養生活力丹) 추출물이 흰쥐의 음경발기에 미치는 영향

Objective : This study investigated the effects of Yangsaengwhallyukdan on nitrite oxide synthase (NOS) activity, nitrite level, antioxidation and erectile response in rat corpus cavernosum penis. Methods : Yangsaengwhallyukdan was formulated to contain various natural products that have been used to treat erectile dysfunction. Yangsaengwhallyukdan suspended in a 0.5% CMC solution was oral-administered, 50 mg or 100 mg per 1 kg of body weight for 30 days, while the normal group was administered only with a 0.5% CMC solution. The efficacy of the Yangsaengwhallyukdan was assessed with regard to erectile function. Results: After the Yangsaengwhallyukdan was administered to the rat for 30 days, the urethral NOS activity increased. The level of urethral nitrite, glutathione and testosterone increased too. The level of urethral lipid peroxide was decreased in Yangsaengwhallyukdan treated rats. The erectile response to a cavernous nerve stimulation in L-NAME$(10^4)$-treated rats in rats given Yangsaengwhallyukdan reached the level of the normal group. Conclusions : This study suggest that Yangsaengwhallyukdan is effective for the treatment of erectile dysfunction.

Objective : This study was to investigate diagnosis and treatment based on an overall analysis of signs and symptoms on a hyperthyroid patient. Methods : The observation of the clinical progress was carried out by conducting diagnosis and treatment based on an overall analysis of signs and symptoms with the patient diagnosed hyperthyroidism. Results : Treatments such as the invigoration of qi (益氣) nourishing m (養陰) was given because the patient showed the qi deficiency (氣虛) and m deficiency (陰虛). Conclusion : Diagnosis and treatment based on an overall analysis of signs and symptoms of a hyperthyroid patient had moderate effects on the improvement of the patients condition.

Objectives : The following are the results of the experimental studies of Samjasan (SJS) extract on the nitric oxide synthase (NOS) activity and the level of lipid peroxide in the penises of rats. Methods : Cnidii Fructus, Cuscutae Semen, Schizandrae Fructus constitute SJS. 150 g of crushed crude drug was extracted with methyl alcohol, under reflux, for 24 hours, three times; the total extractive was evaporated under reduced pressure to give 28.6 g. Results : In vitro, the SJS extract didn't effect the activity of NOS. However, the SJS extract decreased the activities, the ratio of type conversion of xanthine oxidase, the levels of lipid peroxide, In vitro, after administration of the SJS extract to rats, the activities and ratio of type conversion of xanthine oxidase decreased, but the activity of NOS and the content of nitrite increased. Also, the levels of the superoxide anion radical and lipid peroxide decreased in the penises of rats. But, after administration of the SJS extract to rats, the levels of glutathione did not increase. The effects of the SJS extract did better as the dosage and the length of treatment increased. Conclusions : These results suggest that the SJS extract decreases the activities of free radical generating enzymes which form lipid peroxide and increases the NOS activity in the penises of rats. Therefore, the SJS extract is capable of improving of sexual ability in rats.

潛陽法은 平肝治法의 일종으로 刊陽이 上亢해서 나타나는 頭目昏暈, 수족추휵, 구안괘사, 舌乾語澁, 頭痛不止, 猝然倒地 등의 症狀에 쓰는 治法이며 中風의 標實證을 다스리는 목적으로 多用되고 있다. 中風에서의 活用은 淸代에 이르러 본격화되었는데, 葉天士는 "자액식풍, 濡養營絡, 補陰潛陽"을 사용하였고, 張伯龍은 "潛鎭攝納" 하는 치료법을 사용하였다. 中風의 病理가 本虛標實하므로 항상 滋陰, 補水하는 治法과 相互補完的으로 사용되고 있다. 潛陽法이 多用된 병증은 刊胃陰虛, 風陽上擾로 인한 中經絡證과 中腑證 중에서 陽閉證이며, 脫證의 일부와 中風日久로 因한 癲癎에도 사용 되었다. 潛陽藥 중 珍珠母, 石決明, 玳瑁, 牡蠣, 見齒, 龜板, 鱉甲 등 介類의 효력이 뛰어나고, 治方으로는 鎭肝??風陽, 虎潛丸, 固本丹, 復脈湯, 天麻鉤藤飮, 羚羊角湯 등이 활용되고 있다. The methods of checking the exuberance of yang (潛陽法) are a kind of medical treatments Lvhich cure various symtoms caused by hyperactivity of liver-yang, at the same time used for treating apoplexy. Thus, I investigated the application of checking the exuberance of yang to treat stroke. The conclusions are as follows. We can find application of checking the exuberance of yang to apoplexy on Synopsis of the Gold Chamber. After Ye Gui who persisted the theory of endogenous wind, many physicians applied the method of checking the exuberance of yang and this method has usually applied with nourishing yin. The methods of checking the exuberance of yang are concretely applied as follows. 1) a portion of apoplexy involving the channels and collaterals, which is caused by deficiency of \-in and wind-fire pathogen. 2) excessive-syndrome of coma accompanied by heat syndrome. 3) epilepsy caused old apoplexy. The majority of shells for example abalone shell, hawksbill shell, oyster shell, tortoise-shell etc. are the most frequent use of herbal drug which check the exuberance of yang. The precriptions of checking the exuberance of yang are Ho-jam-hwan(虎潛丸), Ko-bon-dan(固本丹), Cheon-ma-gu-deung-eum(天麻鉤藤飮), Chin-gan-sik-pung-tang(鎭肝湜風湯) etc.
