Large conductance Ca^(2+)-activated K^(+) (maxi-K) channel was studied using inside-out patches of smooth muscle cell from rabbit portal vein. Single smooth muscle cells were obtained through proteolytic enzyme digestion (collagenase) and patch clamp technique was used. The maxi-K channel showed a single channel conductance of about 200 pS in physiological concentration of K^(+) on both sides of patch. The channel activity was dependent on intracellular Ca^(2+) concentration change. Application of estrogen with physiological dosage (10 ng/dl) greatly increased the maxi-K channel activity time-dependently. From these results, it is strongly suggested that maxi-K channels of rabbit portal vein were activated by estrogen through membrane-delimited pathway.

To demonstrate and study the functions of calcium channel of the contractile responses of rabbit clitoral smooth muscle. The clitoris is one of major female genital organ. New Zealand White rabbits, around 2.5 kg, were used for the experiment. They were anesthetized with urethane (1 g/kg) then the clitoral tissues have been taken and prepared, using force transducer to record the contractility. Phenylephrine (PE) and highly concentrated potassium (K) induced contractions of the clitoral smooth muscle strips in a dose dependent manner. This contractile response was divided into tonic and phasic components. The phasic component only occurred with high concentration K solution over 35 mM Verapamil, L-type Ca channel antagonist, revealed concentration-dependent relaxation when the tonic contractions were induced by PE and K. However, it rarely affected the phasic contractions. The relaxations caused by verapamil was pronounced in the contraction evoked by K. When the contraction by PE, prazocin, α-adrenergic antagonist, was shown to be more pronounced than verapamil. NiCl₂, T-type Ca channel antagonist, has no effect on tonic contraction. However, it inhibited phasic contraction by over 80%. In conclusion, it is likely there are receptor-operated and voltage- operated Ca channels in the rabbits' clitoral smooth muscle. L-type and T-type Ca channels seem to be coexist in the voltage-operated Ca channel. The contraction and relaxation by the Ca channels of the clitoral smooth muscle functions might be regulated in many complicated ways at the physiological environment.
