Cardiovascular disease is the most common cause of death in diabetic patients. Hyperglycemia is the primary characteristic of diabetes and is associated with many complications. The role of hyperglycemia in the dysfunction of human cardiac progenitor cells that can regenerate damaged cardiac tissue has been investigated, but the exact mechanism underlying this association is not clear. Thus, we examined whether hyperglycemia could regulate mitochondrial dynamics and lead to cardiac progenitor cell dysfunction, and whether blocking glucose uptake could rescue this dysfunction. High glucose in cardiac progenitor cells results in reduced cell viability and decreased expression of cell cycle-related molecules, including CDK2 and cyclin E. A tube formation assay revealed that hyperglycemia led to a significant decrease in the tube-forming ability of cardiac progenitor cells. Fluorescent labeling of cardiac progenitor cell mitochondria revealed that hyperglycemia alters mitochondrial dynamics and increases expression of fission-related proteins, including Fis1 and Drp1. Moreover, we showed that specific blockage of GLUT1 improved cell viability, tube formation, and regulation of mitochondrial dynamics in cardiac progenitor cells. To our knowledge, this study is the first to demonstrate that high glucose leads to cardiac progenitor cell dysfunction through an increase in mitochondrial fission, and that a GLUT1 blocker can rescue cardiac progenitor cell dysfunction and downregulation of mitochondrial fission. Combined therapy with cardiac progenitor cells and a GLUT1 blocker may provide a novel strategy for cardiac progenitor cell therapy in cardiovascular disease patients with diabetes.

Surgical-orthodontic treatment is aimed to restore occlusal function, harmonize a maxillofacial complex, and improve facial esthetics. As patients with dentofacial deformities desire an immediate improvement of appearance, surgery-first or surgery-early approach with minimal presurgical orthodontics has been reported. In addition, three-dimensional analysis using CBCT and computer-aided surgical planning (CASP) have made it possible to perform more accurate treatment plans for orthognathic surgery. This case report describes a successful surgery-early approach in a Class III patient with facial asymmetry. A 23-year-old male patient presented to our clinic with facial asymmetry and mandibular prognathism. The three-dimensional analysis showed mandibular deviation to the right side due to overgrowth of the left mandibular body, and the dentition presented canting of the occlusal plane to the left side with dental compensation of the maxillary right molars. Minimal presurgical orthodontic treatment was performed to improve arch coordination and eliminate premature contacts to achieve stable postoperative maxillo-mandibular relation. Afterward, canting correction and superior impaction of the posterior maxilla, and differential setback of the mandible were planned using CASP. Early bimaxillary surgery was performed with a 3D printed wafer. After a total of 20 months, the patient achieved a favorable facial profile and functional occlusion as planned. Biological Control Activity of Two Isolates of Pseudomonas fluorescens against Rice Sheath Blight

Two isolates of mucous bacteria, mc75 and pc78, were isolated from fungal culture plate as culture contaminants with an interesting swarming motility. Both isolates were identified as Pseudomonas fluorescens based on microscopy, biochemical analysis, Biolog test and DNA sequence analysis of the 16S rRNA gene. Both strains have the exactly the same 16S rRNA gene sequences, and yet their biological control activity were not identical each other. In vitro analysis of antagonistic activity of two isolates against several plant pathogenic fungi indicated that both produced diffusible and volatile antifungal compounds of unknown identities. Treatment of the bacterial culture of P. fluorescens pc78 and its culture filtrate exhibited a strong biological control activity against rice sheath blight in vivo among six plant diseases tested. More effective disease control activity was obtained from treatment of bacterial culture than that of culture filtrate. Therefore, in addition to antifungal compound and siderophore production, other traits such as biofilm formation and swarming motility on plant surface may contribute to the biological control activity of P.fluorescens pc78 and mc75.
FCP 15 : A case of extranodal NK/T cell Lymphoma mimicking refractory Behcet`s disease

A 39-year-old female was diagnosed with complete type Behcet’s disease (BD) having symptoms of oral ulcer, genital ulcer, uveitis and erythema nodosum-like skin lesions. Colchicine, azathioprine or mycophenolate mofetil and systemic corticosteroid were prescribed to reduce the symptoms. However, active uveitis and cutaneous lesions persisted despite treatment. Under suspicious lung involvement of BD, infliximab was started to lessen systemic inflammation yet wihout much effect. Subsequent lung biopsy to investigate pulmonary infiltrate revealed opportunistic infection by Pneumocystis jiroveci. After stopping infliximab infusion, prednisolone in 1mg/kg/day dose was maintained with other immunosuppressants. Surprisingly, cytological specimen from palliative surgery to improve visual acuity discovered a concurrent involvement of vitreoretinal large cell lymphoma. Additional biopsy performed on hard palate ulceration which initially thought to be a symptom of BD revealed unexpected extranodal NK/T cell lymphoma involvement, and retrospective review of pathological slides of skin also revealed the presence of lymphoma. BD should be always suspected of its final diagnosis when systemic vasculitic inflammation persists without any plausible cause. If symptoms are not alleviated even with intense systemic treatments, other suspicious diagnosis including malignancy should be doubted in practice.

As one of the most important crop, rice is not only a staple food of half world’s population but a wonderful model plant, which has been leading the evolution and functional genomics study. The next-generation sequencing technology are expediting rice genomic study, by providing a simple but powerful way. In this study, we re-sequenced a core collection of 137 rice accessions from all over the world along with 158 Korean breeding varieties. Finally, 6.3G uniquely mapped reads were obtained, and about 10 million SNPs and ~1.2 million InDels were identified with average sequencing depth of 7.5X. These will help us to maximize our germplasm utilization and assists all the deep research in population dynamics and functional studies. Here, we’d like to show the approaches applied to resequencing data mining and on-going activities.

<B>ABSTRACT</B><P>A microbial community analysis of forest soil from Jindong Valley, Korea, revealed that the most abundant rRNA genes were related to <I>Acidobacteria</I>, a major taxon with few cultured representatives. To access the microbial genetic resources of this forest soil, metagenomic libraries were constructed in fosmids, with an average DNA insert size of more than 35 kb. We constructed 80,500 clones from Yuseong and 33,200 clones from Jindong Valley forest soils. The double-agar-layer method allowed us to select two antibacterial clones by screening the constructed libraries using <I>Bacillus subtilis</I> as a target organism. Several clones produced purple or brown colonies. One of the selected antibacterial clones, pJEC5, produced purple colonies. Structural analysis of the purified pigments demonstrated that the metagenomic clone produced both the pigment indirubin and its isomer, indigo blue, resulting in purple colonies. In vitro mutational and subclonal analyses revealed that two open reading frames (ORFs) are responsible for the pigment production and antibacterial activity. The ORFs encode an oxygenase-like protein and a putative transcriptional regulator. Mutations of the gene encoding the oxygenase canceled both pigment production and antibacterial activity, whereas a subclone carrying the two ORFs retained pigment production and antibacterial activity. This finding suggests that these forest soil microbial genes are responsible for producing the pigment with antibacterial activity.</P>
