여러 가지 카이랄 금속 촉매를 이용한 one oxygen atom transfer(oxo-transfer) 반응은 생체 내 산화 반응에 있어서 중요한 역할을 해왔을 뿐만 아니라, 활성 천연물 및 의약품 합성시 중요한 중간체의 비대칭 합성에 많이 응용되어왔다.
이 중 chiral manganese(III) salen complexes를 이용한 unfunctionalized olefins의 asymmetric epoxidation은 이 분야에서 널리 사용되어질 수 있는 유용한 반응이라 할 수 있다. 따라서 본 연구에서는 chiral binaphthol 유도체를 이용하여 비대칭 산화반응에 이용될 수 있는 manganese(III) salen complexes를 개발하여 olefin의 촉매적 비대칭 산화 반응을 수행하고자 하였다. 그 결과 적당한 steric effect를 줄 수 있는 MnBI2B, MnBI2C 촉매를 개발하였으며, 이 촉매를 이용하여 산화제 sodium hypochlorite와 함께 conjugated cis-olefins에 적용시킴으로써 경제적 및 산업적으로 유용한 입체 선택적인 epoxide를 얻을 수 있었다. Chiral binaphthalene 모핵과 diamine 모핵의 상대적인 chirality는 비대칭 에폭시화 반응시 입체 선택성에 영향을 주었으며, 생성물의 configuration은 diamine의 chirality에 의해 결정됨을 알 수 있었다.
또한, sulfides의 asymmetric oxidation에 대해서도 유사한 방법으로 적용하고자 하였는데, 다음의 2가지 부류의 촉매 및 산화제를 이용하여 chiral sulfoxides를 합성하였다. 첫번째는 mono- 또는 di-aldehyde binaphthol 모핵과 aminoalcohol 모핵으로 구성된 광학활성의 oxovanadium(IV) complexes를 합성하여 산화제 hydrogen peroxide와 함께 sulfide의 비대칭 산화반응을 하였고, 두번째는 비대칭 에폭시화 반응에서 사용한 chiral manganese(III) salen complexes를 이용하여 산화제 iodosobenzene와 함께 비대칭 설폭시화 반응을 하였다. 그 결과 vanadium(IV) complexs를 촉매로 사용하였을 경우, H4BI(S)Lu2(S)와 H2BI(R)Lu(S) ligand가 효과적이었으며, manganese(III) complexes를 사용하였을 경우, MnBI2(R)C(R)가 thioanisole의 산화반응에 있어서 현재까지 보고된 결과보다 높은 입체 선택성을 보여주었다.
한편, 비대칭 산화반응을 제외하고라도 일반적인 sulfide의 산화반응에서 sulfoxide 또는 sulfone이 생성되는데, 이중 한가지만 생성되도록 하는 선택적 산화는 상당히 어려운 것으로 알려져 있다. 이에 우리는 sulfide의 선택적 산화 방법을 찾고자 노력하였으며, 그 결과 저가의 산화제인 과산화수소와 trirutile-type solid oxide (AMMOx)를 촉매로 개발하여, 메탄올 용매 하에서 다양한 sulfide(aryl alkyl sulfide, allyl sulfide)에 대하여 높은 수율의 선택적 산화 반응을 성공시켰다. 또한 trirutile type solid oxide 촉매는 heterogeneous 촉매여서, 생성물과 분리가 쉽고 재사용 할 수 있었다.
이상에서 저자는 비대칭 산화반응(epoxidation, sulfoxide)에 유용한 촉매와 trirutile type solid oxide 촉매 개발을 통하여 새로운 chiral drug 이나 racemic switch로 재개발되고 있는 의약품의 순수 이성질체 합성 및 천연물의 중요한 중간체로 사용될 수 있는 sulfide의 선택적 산화반응에 유용하게 이용되어질 것으로 기대한다.

The one oxygen atom transfer(oxo-transfer) reactions using various chiral metal catalysts have been provided important roles in many types of reactions, such as not only the biological oxidative reaction but also the asymmetric synthesis of important intermediates for the synthesis of drug and natural compound. Among these types of reactions, the asymmetric epoxidation of the unfunctionalized olefins using by chiral manganese(III) salen complexes is the useful reaction for this purpose. Therefore, the purpose of our research was the synthesis of new types of manganese(III) salen complexes that was composed with chiral binaphthol moieties, and the applications to asymmetric oxidation of various olefins using our new catalysts. As a result, we could synthesize new catalysts, MnBI2B and MnBI2C, which have proper steric effect, and successfully applied these catalysts to the chiral epoxidation of conjugated cis-olefins with NaOCl. The stereoselectivity of the epoxidation reaction was dependent on the relative chirality of binaphthyl and diamine moiety of the catalyst, and the configuration of the products was affected from the chirality of diamine moiety.
We also studied asymmetric oxidations of various sulfides in the similar methods. We have been employed following methods for the preparation of chiral sulfoxides: The first method used the optically active oxovanadium(IV) complexes, which were consisted of mono or di-formyl binaphthol moiety and amino alcohol moiety. In this case, hydrogen peroxide was used as an oxidant for the asymmetric sulfoxidation. The second method used the chiral manganese(III) salen complexes similar to our epoxidation studies. The oxidant used iodosobenzene. As for the as asymmetric sulfoxidation reaction using the vanadium(IV) complexes as catalysts, H4BI(S)Lu2(S) and H2BI(R)Lu(S) were shown to be effective ligands. In the reaction with manganese(III) complexes, the enantioselectivity obtained with MnBI2(R)C(R) in sulfoxidation of thioanisole was better than any previously published result.
The selective oxidations from sulfides to sulfoxides have been known to be very difficult because the sulfones were generally produced in the normal oxidation of sulfides. Therefore, we tried to solve this problem. Fortunately, we could achieve the highest selective oxidation of various sulfides at room temperature in methanolic solution using hydrogen peroxide as an oxidant, and trirutile-type solid oxide (AMMOx) catalysts. These catalysts were heterogeneous, easily separated and reused.
From these oxidation results, it is expected that our new types of catalysts are served to solve the key steps of the syntheses of the enantiomerically pure drugs and natural compounds.
